‘Woody breast’ (WB) and ‘white striping’ in broiler meat is a global problem. With unknown etiology, WB negatively impacts bird health, welfare and is a significant economic burden to the poultry industry. New evidence has shown that WB is associated with dysregulation in systemic and breast muscle-oxygen homeostasis, resulting in hypoxia and anaemia. However, it has been observed that phytase (Quantum Blue (QB) a modified, E. coli-derived 6-phytase) super dosing can reverse dysregulation of muscle-oxygen homeostasis and reduces WB severity by ~5%. The objective of this study was to assess whether levels of Ins(1,3,4,5,6)P5, the main allosteric regulator of haemoglobin, are influenced by changes in plasma myo-inositol arising from super dosing with phytase. To enable this, methods suitable for measurement of myo-inositol in tissues and inositol phosphates in blood were developed. Data were collected from independent trials, including male Ross 308 broilers fed low and adequate calcium/available phosphate (Ca/AvP) diets supplemented with QB at 1,500 phytase units (FTU)/kg, which simultaneously decreased gizzard InsP6 (P<0.001) and increased gizzard myo-inositol (P<0.001). Similarly, male Cobb 500 broiler chicks fed a negative control (NC) diet deficient in AvP, Ca and sodium or diet supplemented with the QB phytase at 500, 1000 or 2,000 FTU/kg increased plasma (P<0.001) and liver (P=0.007) myo-inositol of 18d-old birds at 2,000 FTU/kg. Finally, QB supplementation of Cobb 500 breeder flock diet at 1,250 FTU/kg increased blood myo-inositol (P<0.001) and erythrocyte Ins(1,3,4,5,6)P5 (P=0.011) of their 1d-old hatchlings. These data confirmed the ability of phytase to modulate inositol phosphate pathways by provision of metabolic precursors of important signalling molecules. The ameliorations of WB afforded by super doses of phytase may include modulation of hypoxia pathways that also involve inositol signalling molecules. Elevations of erythrocyte Ins(1,3,4,5,6)P5 by phytase supplementation may enhance systemic oxygen carrying capacity, an important factor in the amelioration of WB and WS myopathy.

The objective of the trial was to determine the impact of corn source and xylanase on broiler performance, digestibility, and volatile fatty acid (VFA) profiles. Six corn samples were obtained from different regions of the US. Twelve treatments were derived using corn source, with each corn diet being fed with or without xylanase. Three dietary phases were used throughout the trial, starter (d 1–18), grower (d 19–31), and finisher (d 32–41). On d 18 and 41, ileal and excreta contents were collected for the determination of ileal digestible energy (IDE), ileal energy and nitrogen digestibility coefficients (IEDC and INDC), apparent metabolisable energy (AME), and caecal VFA profiles. Day 18 body weight (BW) was affected by corn source and varied between 724 and 764g (P = 0.001). For d 31 BW, there was an interaction of corn source with xylanase (P = 0.001), with the effect of xylanase being inconsistent. The effect of xylanase on feed conversion ratio (FCR) during the grower phase depended on corn source (interactive term, P = 0.021). Xylanase reduced (P = 0.026) FCR during the finisher phase (1.943 vs. 1.992). Variation of corn source influenced digestibility on all evaluated parameters. A range of 152 and 213 kcal/kg for IDE was observed on d 18 and 41, respectively (P = 0.005 and 0.001). The range of AME was 176 kcal/kg on d 18 of age which increased to 194 kcal/kg on d 41. Nitrogen digestibility was influenced by corn source, with an observed range of 4.4 and 6.1% for d 18 and 41, respectively, amongst all corn sources (P = 0.001). Xylanase increased (P = 0.031) the concentration of butyrate in the caecum on d 18. On d 41, an interaction between corn source and xylanase was observed with isovalerate in the caecal contents (P = 0.038). These data demonstrate the impact of varying corn nutrient profiles on nutrient utilisation and growth performance.

Supplementing phytase to diets deficient or submarginal in nonphytate P results in improved amino acid digestibility, energy digestibility, bone breaking strength, tibia ash, and BW gain [1][2][3][4][5]. The improvement in these parameters is dependent upon the supplemental phytase con- SUMMARYPhytase supplementation beyond the standard doses used for phosphorus release has been reported to result in extraphosphoric effects by enhancing nutrient digestibility resulting in improved performance of broilers. A study was conducted to examine the effects of the progressive addition of an enhanced Escherichia Coli phytase (400-1,600 phytase units; FTU) on growth performance and carcass characteristics from 1 to 42 d of age in male broilers. One thousand four hundred Hubbard × Cobb 500 1-d-old chicks were randomly distributed into 56 floor pens (0.08 m 2 /bird). Seven dietary treatments were provided in a 3-phase feeding program consisting of (1) a positive control (adequate Ca and nonphytate P; PC); (2) 1 negative control (Ca and nonphytate P reduced by 0.14% and 0.13%; NC); (3 to 6) the NC diet with 4 increasing supplemental phytase concentrations (NC + 400 FTU, NC + 800 FTU, NC + 1,200 FTU, and NC + 1,600 FTU, respectively); and (7) a low-energy NC diet without phytase and xylanase (reduced 66 kcal of AME n /kg). Body weight gain, feed conversion, mortality, weight and yield of whole carcass, abdominal fat, and pectoralis major and minor muscles were evaluated. Progressive supplementation of phytase decreased cumulative FCR linearly. Broilers fed diets containing 1,600 FTU had heavier total breast meat by 49 g compared with birds receiving the PC diets. Broilers consuming the NC + 400 FTU or the low-energy NC diet had similar growth performance and meat yield compared with birds provided PC diet. These data indicated that phytase supplementation beyond the need for phosphorus enhances growth performance and carcass characteristics.

In broilers, 2-hydroxy-4(methylthio) butanoic acid (HMTBA) can elicit a different dose response relative to dl-Met (DLM) such that birds could have lower gain responses at deficient TSAA concentrations but greater gain responses at maximum response concentrations. Two experiments tested if the 2 Met sources have a different dose response in 1-d-old turkeys using a 2 x 4 factorial plus a control design with 8 replicates of 12 toms per treatment. 2-Hydroxy-4(methylthio) butanoic acid and DLM were supplemented at equimolar concentrations of 0.05, 0.10, 0.15, and 0.20% or 0.04, 0.08, 0.16, and 0.32% for experiments 1 and 2, respectively, in commercial-type TSAA-deficient (0.99 to 1.02%) diets for 21 d. No differences in any performance parameter tested were found between HMTBA and DLM in either trial by ANOVA. Linear (LIN), quadratic (QUAD), and exponential regressions were fitted to the gain response of birds fed HMTBA or DLM. Equations with better goodness of fit as determined by Schwarz's Bayesian information criteria index were used for further calculations of predicted differences between HMTBA and DLM. In both trials, the shape of the dose response differed according to the Met source used, and best-fit equations were obtained when using Met intake over control rather than dietary Met concentration as the dependent variable. In experiment 1, the best-fit equations were an inverse QUAD for HMTBA and a LIN for DLM, and in experiment 2 with higher Met concentrations, the best-fit equations were a QUAD for DLM and a LIN for HMTBA. Feeding HMTBA at deficient TSAA resulted in lower (P <0.05) gains in experiment 1 but greater gains at maximum response concentrations (P <0.05) in both experiments. Plasma-free Met increased at 3 times the rate for DLM than HMTBA (P <0.01) with increasing Met concentration, which may play a role in the evolution of different dose responses at the extremes of the Met dose response. These results demonstrate that Met sources elicit a differential dose response in turkeys such that feeding HMTBA at deficient TSAA concentrations can be lower than DLM and can reach a higher maximum performance than with DLM.
