Зміни процесів вільнорадикального окиснення ліпідів і білків, антиоксидантного захисту у щурів із гіпофункцією щитоподібної залози на тлі дефіциту йоду та міді В експериментах на щурах із гіпофункцією щитоподібної залози на тлі монодефіциту йоду (ГЩЗ І ) та комбінованого дефіциту йоду й міді (ГЩЗ І+Cu ) вивчали тиреоїдний статус, баланс міді, співвідношення процесів перекисного окиснення ліпідів (ПОЛ), білків (ПОБ), антиоксидантого захисту (АОЗ). Установлено, що комбінований дефіцит мікроелементів супроводжується перерозподілом вмісту міді між різними тканинами (збільшення в еритроцитарній масі та головному мозку, зменшення у міокарді), суттєвими змінами показників гіпоталамо-гіпофізарно-тиреоїдної осі, киснезалежного метаболізму, протирадикального захисту, потенціюючи негативний вплив на організм кожного із них. З'ясовано, що ГЩЗ І+Cu зумовлює пригнічення киснезалежних процесів, про що свідчить зменшення вмісту дієнових кон'югатів (ДК) на 69,70 % у щитоподібній залозі, продуктів, що реагують на тіобарбітурову кислоту (ТБК-РП) на 47,72 % у проміжному мозку, продуктів окиснювальної модифікації білків на 37,10-98,98 % у проміжному мозку, щитоподібній залозі та печінці на тлі вибіркової активації пероксидації у сироватці крові (зростання ТБК-РП у 7,11 раза) та у міокарді (збільшення ДК -на 48,15 %, продуктів окиснювальної модифікації білків -у 2,64 раза) щодо аналогічних показників у інтактних тварин. Одержані результати (маніфестація ВСТУПОднією із пріоритетних медико-соціальних проблем є організація належної профілакти-ки, пошук нових можливостей ранньої діагностики та забезпечення ефективного лікування гіпотиреозу. Незважаючи на значні успіхи у вивченні патогенезу цієї патології, тенденція до прогресування захворювань щитоподібної залози є наслідком складної взаємодії ендо-та екзогенних чинників [1,2]. Етіологічна роль йоду у виникненні ендемічного зобу доведена давно [3,4]. Проте формування гіпофункції щитоподібної залози (ГЩЗ) відбувається також на тлі впливу неспецифічних чинників довкілля, що сприяють розвитку зобу. До них, зокрема, належать сполуки міді, надлишок або дис-баланс якої може зумовити розвиток тиреоїдної дисфункції [5]. З іншoго боку, цей мікроеле-мент необхідний для ефективного функціо-нуван ня щитоподібної залози, адже є ком- понентом ферментів (йодидпероксидази, тирозинази, тиреопероксидази та інших протеїназ), що забезпечують утворення тироніну, оксидацію йодиду в процесі синте зу тироксину [6,7]. Мідь також залу-чена в процес синтезу тиреотропного гор-мону аденогіпофіза (ТТГ) як компонент протеїнкінази [8]. Тому можна припустити, що комбінований дефіцит цих мікроеле-ментів може потенціювати негативний вплив на організм кожного із них, зокрема, супроводжуватися вираженими змінами тиреоїдного гомеостазу. Topicality. Pancreatic hormones play an important role in the process of remodeling of hard periodontal tissues. Violations of carbohydrate metabolism that occur under conditions of isolated insulin resistance (IR) and in combination with iodine deficiency (ID) can be the cause of dysmetabolic disorders of mineralization/demineralization physiological system of tooth-maxillary complex. The aim. To study the changes of remodeling processes of hard periodontal tissues in rats with isolated IR and impaired glucose tolerance against the background of ID. Materials and methods. The examination was carried on 90 male rats, which were divided into three groups: control (intact animals), group of rats with IR under conditions of adequate iodine supply, and with IR against the background of ID. The system of carbohydrate metabolism was studied by the level of insulin in blood serum, glucose and glycosylated hemoglobin (HbA1c) of blood with the following calculation of HOMA-IR index. The processes of mineralization/demineralization were detected by the content of calcium, magnesium, zinc, manganese, copper in cementum of tooth root and alveolar process, and by the activity of acid and alkaline phosphatases in blood serum. Results. Keeping of animals on a high-fructose diet led to the development of carbohydrate metabolism disorders (increase blood glucose and HbA1c levels, blood serum insulin, HOMA-IR index) and changes of remodeling processes in hard periodontal tissues (decrease the level of calcium, magnesium and manganese against the background of increase the zinc content in cementum of tooth root; decrease the content of macroelements in alveolar process; activation of acid phosphatase against the background of inhibition of alkaline phosphatase activity). The development of combined endocrinopathy was accompanied by more pronounced changes of studied parameters. Conclusions. The violation of glucose tolerance against the background of ID slows down the mineralization processes of hard periodontal tissues mainly due to the intensification of osteoresorptive processes. Carbohydrate metabolism markers, thyroid profile indexes, transaminase activity and structural features of liver were studied, taking into account sexual dimorphism, in rats which were keeping on a high-fructose diet under conditions of adequate iodine supply and pre- and postnatal iodine deficiency. It was found that under fructose loading of animals with iodine deficiency the risk of insulin resistance (IR) development increases (HOMA-IR index in insulin-resistant male rats under conditions of pre- and postnatal iodine deficiency increases respectively by 22 and 13% compared to the data in animals that received a high-carbohydrate diet and adequate iodine supply). It is important, that in males and females with IR, the secretion of free triiodothyronine decreased by 36 and 40%, thyroid-stimulating hormone – by 28 and 34%, respectively, compared to the data of intact animals. The development of IR was accompanied by the activation of aminotransferases in blood serum (more in males). In particular, in male rats with IR under conditions of pre- and postnatal iodine deficiency, the activity of aspartate aminotransferase increased by 2.7 and 2.4 times, alanine aminotransferase – by 3.9 and 2.7 times, respectively, which led to a decrease of the de-Ritis index (more significantly against the background of prenatal iodine deficiency – by 25%) in relation to the data in animals that were on a standard diet. The activity of aminotransferases in rats with IR under conditions of iodine deprivation was higher than in animals with isolated iodine deficiency – by 11-56% and IR – by 13-47 %. In animals with IR, the presence of binucleated hepatocytes with a diffuse decrease in the number of glycogen granules in them and accumulation of glycoproteins in the wall of liver vessels were found. The foci of fatty dystrophy and pronounced zonal character of dystrophic processes in hepatocytes were identified in females, diffuse dystrophy prevailed in males. In the cytoplasm of hepatocytes of animals with IR under conditions of prenatal iodine deficiency, the lightening and the granularity, single granules of glycogen were observed. Under these conditions, the average perimeter (by 74%) and area (by three times) of hepatocytes, the average area of nucleus (by 3.1 times), the average perimeter (by 50%) and area (by 66%) of the nucleolus were increased compared to the control values. The structural changes were consistent with an increase of the transaminase activity. Thus, fructose loading and the development of IR acts as a trigger for cytolysis of hepatocytes, which increases under conditions of iodine deficiency, especially prenatal.
