The objective of this study was to evaluate the antimicrobial potential of Lippia alba essential oil (EOLa) and to investigate the effect of food ingredients on its efficacy. The antimicrobial potential of the oil was determined by the presence or absence of inhibition zones, minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) against Escherichia coli, Listeria innocua, Listeria monocytogenes, Pseudomonas aeruginosa, Salmonella choleraesuis and Staphylococcus aureus. The effect of food ingredients and the pH on the antimicrobial efficacy of oil was assessed by monitoring the maximum growth rate of Listeria monocytogenes in model media. The model media included potato starch (0, 1, 5 or 10%), beef extract (1, 5, 3, 6 or 12%), sunflower oil (0, 5 or 10%) and TSB broth at pH levels of 4, 5, 6 or 7. The EOLa showed efficacy at all concentrations (50%, 25%, 6.25%, 3%, 1.5%, 0.8%, 0.4% and 0.2%) evaluated, against all bacterial species, Gram-positive and Gram-negative. The antimicrobial efficacy of EO was found to be a function of ingredient manipulation. Proteins and lipids had a negative impact on the oil effectiveness, indicating the protective action of both on the microbial specie tested. On the contrary, at the highest concentration of starch (10%), the lower rate growth of L. monocytogenes was detected, therefore indicating a positive effect of carbohydrates on the oil effectivenes. Regarding the pH, the studies showed that the rate of microbial growth increased with increasing pH. It was concluded that the use of EOLa is more effective control pathogenic and spoilage bacteria when applied to starchy foods under an acidic pH.
RESUMOA microbiota é um fator de grande importância na qualidade e conservação de vegetais minimamente processados. Muitos microrganismos podem afetar adversamente a qualidade e a segurança desses produtos, considerando que microrganismos patogênicos, que normalmente não estariam presentes, passam a compor a microbiota contaminante decorrente do manuseio a que são submetidos. Objetivou-se, neste trabalho, determinar a qualidade microbiológica de hortaliças, anterior e posteriormente ao processo de sanitização, como também o período final da vidade-prateleira dos produtos sanitizados, avaliando-se a presença de Listeria sp., Listeria monocytogenes e Salmonella sp. Foram analisadas amostras de agrião, alface, cenoura ralada, espinafre, repolho verde ralado e rúcula minimamente processados. As amostras não sanitizadas foram analisadas no dia da coleta, e as sanitizadas analisadas a 1, 5 e 8 dias após o processamento, a fim de determinar a condição microbiológica dos produtos. As hortaliças foram armazenadas sob temperatura de 5ºC. Salmonella sp. foi detectada em 12,7% das amostras, enquanto L. monocytogenes não foi encontrada em nenhuma delas, embora outras espécies de Listeria tenham sido isoladas de alface e cenoura. ABSTRACTThe microbiota is a factor of great importance to the quality and conservation of minimally processed vegetables. Many microorganisms may adversely affect the quality and safety of these products, considering that normally present pathogenic microorganisms may eventually compose the polluting microbiota whenever being submitted to handling. This work aimed to determine both: vegetable microbiologic quality, prior and post-sanitization process as well as products post-sanitization shelf-life expiring date, while the presence of Listeria sp., Listeria monocytogenes and Salmonella sp. was evaluated. The minimally processed vegetables samples analyzed were watercress, lettuce, grated carrot, spinach, grated green cabbage and rucula. The non-sanitized samples were analyzed on the first harvesting day (day 1), and the sanitized ones were analyzed at 1, 5 and 8 days after being processed in order to determine the product microbiological state. The vegetables were stored under a temperature of 5ºC. Salmonella sp. was detected in 12.7% of the samples, while L. monocytogenes was not found in any of them, though other species of Listeria were isolated from lettuce and carrot. According to the RDC nº 12, the presence of Salmonella sp. rendered 16 samples as being inappropriate for human consumption. Determinou-se o potencial antimicrobiano do óleo essencial (OE) de Cymbopogon citratus (DC) Stapf e o efeito dos componentes dos alimentos sobre sua efi cácia. O potencial antimicrobiano do óleo foi determinado pela presença de zona de inibição de crescimento e concentração inibitória mínima contra Listeria monocytogenes, L. innocua, Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa e Salmonella choleraesuis. O efeito dos componentes dos alimentos e do pH sobre a efi cácia antimicrobiana do óleo foi monitorado pelo crescimento de L. monocytogenes em meios de cultura modelos, constituídos de amido de batata (0, 1, 5 e 10 %), extrato de carne (1,5, 3, 6 e 12 %), óleo de girassol (0, 5 e 10 %) e caldo tripticase de soja (TSB) em pH 4, 5, 6 e 7. O OE mostrou efi cácia contra todas as espécies bacterianas testadas. O efeito antimicrobiano do óleo variou em função da concentração do componente manipulado. Proteínas e lipídios apresentaram impacto negativo na efi cácia do OE; quanto maior a concentração destes componentes nos meios maiores foram as médias do crescimento bacteriano. O amido apresentou impacto negativo na efi cácia do óleo nas concentrações de 1 % e 5 %. Contudo, na mais alta concentração avaliada (10 %) observou-se um declínio no crescimento bacteriano. A atividade antimicrobiana do OE aumentou com a diminuição do pH. Os resultados deste estudo comprovaram a efi cácia antimicrobiana do OE do C. citratus para agentes patogênicos e bactérias deterioradoras de alimentos, verifi cando-se maior efi ciência em alimentos proteicos e amiláceos em pH ácido.
We assessed at multiple length scales (nanometers to millimeters) the nanocoatings of silver nanoparticles (AgNPs) on model SiO 2 /Si substrates. The coatings from biogenic AgNPs (from yeasts Rhodotorula glutinis and Rhodotorula mucilaginosa) were compared to those formed from "synthetic" AgNPs capped with citrate and sodium dodecyl sulfate (SDS). With computational analysis of large-field (LF) X-ray images of the whole substrates (5 × 5 mm), we were able to assess the coatings homogeneity, relative amount of AgNPs, and their distribution as agglomerates. Surprisingly, by analyzing more than 100,000 elements (nanoparticles and agglomerates) in each sample, it was observed that the mentioned features have little dependence on the AgNPs morphology and capping agents. All silver nanocoatings resisted when immersed in phosphatebuffered saline medium by forming agglomerates of up to 10 μm 2 . However, coatings formed with synthetic AgNPs (capped with citrate and SDS) led to a higher antimicrobial efficiency against Staphylococcus aureus.Keywords: nanobiotechnology, image processing, confocal laser scanning microscopy, scanning electron microscopy, large-field X-ray imaging IntroductionSilver nanoparticles (AgNPs) have many technological applications as films and coatings over solid surfaces such as metals, ceramic, glasses and polymers, to prevent growth of bacteria and fungi. [1][2][3][4][5] For the AgNPs to be successfully attached, a suitable match between the solid surface and the nanoparticle physicochemical characteristics must occur. By synthesizing AgNPs with the mediation of bacteria and fungi extracts (biogenic AgNPs or bio-AgNPs), the resulting protein-stabilizing capping agent over the nanoparticle contains a variety of chemical groups bonded to the amino acids fragments. This latter aspect may lead to a myriad of possible chemical interactions (e.g. electrostatic, hydrophobic, hydrogen bonds and van der Waals) between the nanoparticles and solid surfaces. [6][7][8] Essentially, bio-AgNPs are formed from interaction mechanisms ruled by proteins present in the bacterial/ fungal extract or filtrate, which drive the bio-AgNPs' nucleation → growth → colloidal stabilization steps. 2,9,10 At the end of the kinetic process, a protein corona phase on the bio-AgNPs provides a long-term colloidal stability. PREPARATION AND CHARACTERIZATION OF PHOSPHORYLATED STARCH BLENDS WITH CHITOSAN AND POLYVINYL ALCOHOL. The aims of this study were to produce films from phosphorylated starch/chitosan/PVA blends and evaluate their physicochemical properties. The film properties were dependent on the starch percentage in the blend. The infrared spectra of the blends showed the appearance of bands at 1716 and 1733 cm -1 (stretching C = O of PVA acetate residues) and 1556 cm -1 (amide II deformation of the chitosan) indicating the incorporation of these components into the blends. The films with high phosphorylated starch content showed low thermal and mechanical strength and high water absorption capacity while blends with higher content of chitosan and PVA showed good mechanical properties and low water vapor permeability. Films containing 40% and 20% starch showed excellent properties such as high water absorption capacity and biodegradability for use as bandages and agricultural films and also as a source of organic matter in soil.Keywords: blends; starch; permeability; dressing. INTRODUÇÃOA elaboração de blendas com outros biopolímeros é uma das maneiras mais utilizadas para melhorar as propriedades mecânicas dos filmes e membranas de amido.1 No entanto, a adição de altas quantidades de amido a uma matriz polimérica diminui suas propriedades físicas e mecânicas, principalmente porque a imiscibilidade do amido hidrofílico e de polímeros hidrofóbicos, em nível molecular, leva a uma separação de fases.2 Dessa forma, torna-se importante modificar a estrutura do amido a fim de aumentar sua miscibilidade e suprir uma ou mais ausências e/ou deficiências de suas propriedades.A introdução de substituintes químicos na molécula de amido tem sido usada para melhorar propriedades funcionais tais como viscosidade, gelatinização, retrogradação, solubilidade em água, adesão e formação de filmes. Vários estudos sobre hidrólise ácida, oxidação, eterificação e esterificação em amido têm sido realizados. [3][4][5] Para melhorar a hidrofilicidade, o amido pode ser modificado através da fosforilação dos grupos hidroxilas reativos das unidades de anidroglicose do amido. As ligações cruzadas resultantes da modificação reforçam as ligações de hidrogênio que atuam como ponte entre as moléculas de amido. -This study evaluated the use of cashew fiber in combination with textured soybean protein for the preparation of vegetable burger. Two treatments were studied to reduce fiber size (mechanical pressing with or without enzymatic maceration) and its impact on physical chemical characteristics, sensory acceptance and stability of burgers stored at -18 °C for 180 days. Both burger formulations had high moisture and carbohydrate contents and low lipid contents. Enzymatic fiber maceration resulted in harder products with lower moisture content. During storage, reduction in pH and ascorbic acid and increase in acidity values were observed, with no tendencies for color values. Throughout the storage period, burgers were within microbiological standards of the Brazilian legislation for human consumption and average sensory acceptance was within acceptance zone. It was concluded that the inclusion of enzymatic maceration in the fiber treatment process has no advantages and that burgers elaborated with cashew fiber and textured soybean protein can be consumed up to six months of frozen storage. HAMBÚRGUERES VEGETAIS DE FIBRA DE CAJUE PROTEÍNA TEXTURIZADA DE SOJA RESUMO -Neste trabalho, avaliou-se a utilização da fibra de caju em associação à proteína texturizada de soja para elaboração de hambúrguer vegetal. Foram estudados dois tratamentos para a redução do tamanho da fibra (prensagem mecânica associada ou não à maceração enzimática) e seu impacto nas caraterísticas físico-químicas, aceitação sensorial e estabilidade dos hambúrgueres armazenados a -18ºC, por 180 dias. As duas formulações dos hambúrgueres apresentaram alto teor de umidade e carboidratos, e baixo teor de lipídeos. A maceração enzimática da fibra resultou em produtos com menor umidade e mais duros. Durante o armazenamento dos hambúrgueres, observaram-se redução de pH e de ácido ascórbico, e aumento de acidez, não tendo sido observadas tendências para os parâmetros de cor. Os hambúrgueres estiveram dentro dos padrões microbiológicos da legislação brasileira para o consumo humano, e as médias de aceitação sensorial, dentro da zona de aceitação durante todo o período de armazenamento. Conclui-se que, a inclusão de maceração enzimática ao processo de tratamento da fibra não apresenta vantagens e que os hambúrgueres obtidos de fibra de caju e proteína texturizada de soja podem ser consumidos até seis meses de armazenamento congelado.
