Обоснование. Известно, что механизм цитотоксического действия противоопухолевого средства-каталитической системы «терафтал + аскорбиновая кислота» (ТФ+АК)-связан с образованием в клетке активных форм кислорода (ROS) и индукцией в ней окислительного стресса. Система «гемоксигеназа-1/ ферритин» (HО-1/Ft) защищает клетки от окислительного стресса. Цель. Исследовать значение системы HО-1/Ft в защите лейкозных клеток от токсического воздействия противоопухолевого средства ТФ+АК. Материалы и методы. В работе использовались лейкозные клетки человека линий K562 и U937. Базальную и препарат-индуцированную экспрессию HО-1/ Ft на уровне мРНК и белка изучали методами ОТ-ПЦР в реальном времени и вестерн-блоттинга, концентрацию ROS в клетках-методом проточной цитометрии, цитотоксичность препаратов-методом МТТ. Результаты. По нашим данным, в миеломонобластах линии U937 присутствует конститутивно-активная НО-1, а в эритробластах линии K562 экспрессия этого белка заблокирована на уровне мРНК. Гемин, агонист НО-1, индуцирует в клетках U937 коэкспрессию HО-1 и Ft на уровне мРНК и белка. Активация системы HО-1/Ft гемином в клетках U937 не влияет на чувствительность их к ТФ+АК и увеличивает чувствительность, например, к цитарабину в 2 раза. Установлено, что система ТФ+АК вызывает up-регуляцию генов HО-1/Ft, экспрессия которых увеличивается в 4 и 1,5 раза соответственно по сравнению с базальным уровнем. Предынкубация миеломонобластов U937 с хелатором железа дефероксамином приводит к увеличению их устойчивости к ТФ+АК в 2 раза. В то же время в присутствии железосодержащего аналога ТФ цитотоксичность этой системы увеличивается в 2 раза. Заключение. Background. The purpose of this investigation is the search of new compounds with high selectivity of antitumor action on the tumor of the gastrointestinal tract (GIT) in the series of analogues of the peptide hormone gastrin. Objective: synthesis of 2 analogues of gastrin, 1 of which contains the cytotoxic group, the study of their cytotoxic and antitumor activity. Materials and methods. Synthesis of peptides was carried out by classical methods of peptide chemistry. Cytotoxic activity was studied on the cell culture НСТ116. Antitumor activity of analogues of gastrin were studied on transplanted tumors of the GIT of mice AKATOL and AKATON. Results. Two analogues of gastrin (octapeptide) were synthesized, 1 of which contains a cytotoxic group. Analogue containing cytotoxic group revealed the cytotoxic activity. Antitumor activity of two analogues of gastrin were studied on transplanted tumors of the GIT of mice AKATOL and AKATON. The cytotoxic and non-cytotoxic analogues of gastrin showed antitumor activity only on AKATON. Inhibition of tumor growth is 74 and 84 %, respectively. Conclusions. The therapeutic effect of two analogues of gastrin on adenocarcinome AKATON, probably, is associated with the expression of gastrin receptors ССК2 in this tumor.
Introduction. The molecular basis of radio- and photodynamic therapy (PDT), the mechanism of action of a number of antitumor chemotherapy drugs is oxidative stress (OS). The enzyme hemoxygenase-I (НO-1), a molecular marker of OS, is a key participant in the system of protection and adaptation of tumor cells under stress.Objective. To find out whether the sensitivity of human melanoma tumor cells to OS depends on the basal and modulator-induced levels of НO-1 expressionMaterial and methods. Human melanoma cell lines were used in the study. The expression of mRNA НO-1 in cells was studied by real-time RT-PCR, the reactive oxygen species content in cells – by flow cytometry and the cytotoxicity of drugs – by MTT assay.Results. According to our data, human melanoma cells have different basal levels of HO-1 transcription: high (3.0–3.5 o. u.) in lines MelIL, MelP, medium (1.5 o. u.) in lines MeWo, MelZ, MelIbr and low (0.5 o. e.) – MelMe, A375).There is no direct correlation between the level of basal cell expression of HO-1 and their sensitivity to the OS inducer – Н2О2. The hemin-induced increase in HO-1 expression in cells is accompanied by doubled resistance to Н2О2. It was found that HO-1 repression in the presence of apigenin was registered in melanoma cells with different basal levels, but sensitization to Н2О2 (2–4 times) was observed only for cells with medium (MeWo) and low (A375) levels of basal HO-1 expression. It was found that the decrease in basal expression of HO-1 induced by apigenin is accompanied by an increase in the reactive oxygen species content in cells.Conclusions. The results of our research allow us to recommend natural flavon apigenin, a modulator of HO-1 expression, for inclusion in the chemotherapy and PDT regimens to increase the effectiveness of human melanoma treatment.
