Введение. Функции низкомолекулярных РНК <нмРНК> исследованы до сих пор недостаточно. В обзорах по нмРНК упоминается ряд внутриклеточных процессов, которые протекают с участием нмРНК [1, 2], однако надежные доказательства известны в единичных случаях. В частности установлено, что нмРНК участвуют в терминации трансляции секретируемых белков [3]. Появление нескольких гипотез, описывающих участие нмРНК в сплайсинге высокомолекулярных РНК [4][5][6][7][8], сыграли инициирующую роль в повышении интереса к исследованию нмРНК. Недавно были получены первые доказательства в пользу этих гипотез [9,10].Значительный интерес привлекает возможность участия низкомолекулярных РНК в регуляции транскрипции. При использовании системы изолированных ядер наблюдают в основном ингибирующий эффект нмРНК или же его отсутствие [И-13], а в живых клетках нмРНК стимулируют транскрипцию [14,15]. При этом активные фракции нмРНК, выявленные в опытах, способны стимулировать транскрипцию и в системе изолированных ядер [16].Ранее нами было показано, что после инъекции в оплодотворенные яйцеклетки вьюна 0,35 М NaCl экстракта ядер, полученного из зародышей вьюна на стадии гаструлы, включение меченых предшественников в РНК после активации транскрипции возрастает [17]. Далее мы показали, что фракция нмРНК обладает стимулирующим транскрипцию действием. Предварительное сообщение о результатах этой работы было опубликовано ранее [18].В данной работе мы описали эффект усиления синтеза основных макромолекул клетки на ранних стадиях развития зародышей костистых рыб-вьюна (Misguruus fossilis L.) и форели (Salmo gairdnerii) вследствие введения нмРНК, исследовали распределение инъецированных нмРНК между клеточными компартментами, связь распределения нмРНК с внутриклеточным перемещением негистоновых белков (НГБ). Появление в клетках зародыша гомо-и гетерологичных нмРНК приводит к значительному усилению включения радиоактивных предшественников в макромолекулы клетки (ДНК, РНК и белок). Инъецированные нмРНК накапливаются в ядрах клеток зародыша и это координирует с переходом НГБ из цитоплазмы в ядра клеток.Материалы и методы. Synthesis of nuclear and cytoplasmic proteins was studied biochemically and autoradiographically in early loach (Misgurnus fossilis) and sea-urchin (Strongylocentrotus nudus) embryos. After incubation with [14C]amino acids for 5–120 min the ratio of the specific activities of nuclear, mitochondrial and 12000 g supernatant proteins was shown to be equal approximately to 6:1:2 in loach embryos and to 8:4:3 in sea-urchin embryos independently of the duration of labelling. After incubation with [3H]amino acids the number of silver grains per unit section was on the average 2·4 times higher for nuclei than it was for cytoplasm at mid-blastula and mid-gastrula stages. At the mid-gastrula the vegeto-animal gradient of protein synthesis was found. A higher level of the synthesis of nuclear proteins as compared with that of cytoplasmic proteins appears to be related to an increase in the nuclear volume and the nucleo-cytoplasmic ratio during the early development of the loach and sea-urchin embryos.
