Latar BelakangBiji kedelai Detam 1 dan daun jati Belanda berefek menghambat kenaikan berat badan, akan tetapi dikhawatirkan mempengaruhi organ hepar.
Tujuan penelitianTujuan penelitian untuk mengetahui efek ekstrak etanol biji kedelai Detam 1 (EEKD), ekstrak etanol daun jati Belanda (EEJB) dan kombinasinya terhadap penghambatan kenaikan berat badan dan gambaran histopatologis hepar pada tikus Wistar yang diberi pakan tinggi lemak (PTL).
Metode penelitianPenelitian merupakan eksperimental laboratorium dengan rancangan acak lengkap bersifat komparatif. Sebanyak 40 ekor tikus Wistar jantan dibagi secara acak menjadi 8 kelompok perlakuan, masing masing terdiri dari 5 ekor. Selanjutnya diberi perlakuan selama 28 hari, semua kelompok kecuali kelompok kontrol negatif (KN), tetap diberi PTL. Pada hari ke-29, seluruh tikus dikorbankan dan semua hepar tikus, kecuali kelompok Orlistat (K6), dibuat sediaan histopatologis dengan pewarnaan Haematoxylin Eosin (HE).
HasilPenghambatan kenaikan berat badan terjadi pada semua kelompok perlakuan, kelompok K3 (EEKD 10 mg : EEJB 20 mg) menunjukkan penghambatan kenaikan berat badan yang paling baik dan potensinya setara dengan kontrol positip (KP) atau Orlistat. Pada semua kelompok perlakuan (K1, K2, K3, K4 dan K5) terjadi perubahan struktur arsitektur dan inflamasi di daerah portal namun tidak menyebabkan bengkak keruh dan degenerasi lemak.Kesimpulan: Pemberian kombinasi EEKD 10 mg : EEJB 20 mg menunjukkan penghambatan kenaikan berat badan yang paling baik. EEJB sediaan tunggal menyebabkan perubahan gambaran histopatologis hepar paling buruk pada tikus Wistar jantan yang diinduksi pakan tinggi lemak.
Background: Anti-Nutritional-Factors (ANF) are natural or synthetic compounds that interfere with the absorption of nutrients. ANF may cause changes in the histopathology of the small intestinal mucosa, and may affect the digestive, secretory and absorptive functions of the small intestine. Soybean and Jati Belanda leaves which can promote weight loss, may contain ANF. In this study, soybean seeds variety Detam 1 and Jati Belanda leaves were used to assess their potential effects on weight gains and jejunum functions. Research Objective: To study the effects of ethanol extracts of Detam 1 raw soybean (EEDS) and Jati Belanda (EEJB) Bumi Herbal Dago variety and their combinations on weight gain and jejunum histopathological changes in male Wistar rats. Research Methods: Phytochemical analyses of EEDS and EEJB ethanol extracts were performed, then assayed for the above mentioned activities after administered to high-fat induced male Wistar rats for 28 days. Weight gain was determined and histopathological examination of jejunum was achieved after fixation and staining with Hematoxylin Eosin. Results: The combinations of EEDS:EEJB (2:1 and 1:2 w/w) reduced the weight gain with the effect being more pronounced (5.95%) for the combination 1:2 (w/w). Such effect may be attributed to ANFs contained in EEDS and EEJB such as tannin, saponin, flavonoids and phenolics. The histopathologic examination of the positive control group was not different from all treatment groups (p>0.05). Conclusion: The combination EEDS:EEJB (1:2 w/w) is effective in inhibiting weight gain without remarkable histopathological changes in male Wistar rat jejunum.
High-intensity physical exercises can increase oxidative stress, causing damage to reproductive system. The purpose of this study was to determine the effect of mangosteen peel extract on sperm motility and sperm count of Swiss Webster mice induced by high-intensity physical exercise. This was a real experimental with complete randomized design. Twenty five male Swiss Webster mice were divided into five groups (n = 5). Negative control (KN) was without any treatment, positive control (KP) was given treadmill exercise only, the treatment groups (KM1, KM2, KM3) were given mangosteen peel extract orally 2, 4, 8 mg respectively as well as treadmill exercises. The treadmill exercises was conducted for 35 days. The sperm motility (mm/second) and the sperm count (per mm3 ) were assessed manually using Improved Neaubauer chamber. The result were analyzed using one-way ANOVA followed by Tukey HSD. The result showed high significant differences of sperm motility and sperm count between KM2 and KP, also between KM2 with KM1 and KM3 (p=0,000). The conclusion was mangosteen peel extract improved the sperm motility and sperm count of Swiss Webster mice induced by high-intensity physical exercise with 4 mg as the optimal dose.
