Bekasam adalah produk fermentasi ikan. Bekasam diduga mempunyai aktivitas Angiotensin I Converting Enzyme (ACE) inhibitor, yang disebabkan oleh terbentuknya peptida ACE inhibitor hasil aktivitas proteolitik enzim endogenous ikan dan bakteri asam laktat. Penelitian ini bertujuan mengetahui perubahan populasi bakteri asam laktat, pH, total asam, perubahan protein terlarut dan besarnya aktivitas ACE inhibitor selama proses fermentasi bekasam. Jumlah populasi bakteri laktat meningkat dari 5,16 menjadi 8,15 log CFU/g, nilai pH menurun dari 6,5 menjadi 4,41 disertai dengan peningkatan total asam. Bekasam bandeng menunjukkan aktivitas antihipertensi sebesar 51,77% yang terbentuk pada hari ke 6, dan tidak menunjukkan aktivitas penghambatan pada proses fermentasi selanjutnya. Besarnya aktivitas ACE inhibitor berkorelasi dengan terjadinya kenaikan protein terlarut selama proses fermentasi bekasam.

The study was aimed to investigate chemical composition, dietary fiber content and the antioxidant activity of the hydrophilic extract of varieties of rice bran IR-64, Sintanur, Rajalele and Menthikwangi. Rice bran from each sample was analyzed proximate and fiber content. Hydrophilic extract was obtained by extracting each sample of bran with methanol solvent. Total phenolic content was tested by Folin Ciocalteu reagent and antioxidant activity of the extract was tested by DPPH (2,2-diphenyl-1-picrylhydrazyl) method. The results showed that all the four samples had differencesamount of their moisture, ash, protein and fat contents. Fat content varies from 16.80±0.05 % - 23.75±0.04 %db, which the highest level of fat content was possessed by Sintanur bran (23.75±0.04 %db) and IR-64 bran (16.80±0.05 %db) was the lowest level. The levels of soluble fiber from four varieties of rice bran were not significantly different, between4.07 – 4.14 %db but there was a difference in the content of insoluble fiber, which had effect on the total of fiber content. The insoluble fiber content of the rice bran varieties of Menthikwangi, Rajalele, IR-64 and Sintanur were 27.64±0.46, 28.04±0,25, 29.15±0.26 dan 30.44±0.60 %db respectively. The content of total phenols and the highest antioxidantactivity of the hydrophilic extract was owned by Menthikwangi ie. 2794.28±181.83 μg galic acid equivalen (GAE)/g bran for total phenol and 41,28±0,60 % for antioxidant activity.Keywords: Rice bran, hydrophilic extract, rice variety, phenol, DPPH ABSTRAKPenelitian bertujuan untuk mengetahui komposisi kimia, kadar serat pangan serta aktivitas antioksidan ekstrak hidrofilik bekatul dari empat varietas padi yaitu IR-64, Sintanur, Rajalele dan Menthikwangi. Masing-masing sampel bekatul dianalisis proksimat dan kadar serat pangannya. Ekstrak hidrofilik diperoleh dengan cara mengekstrak masingmasingsampel bekatul dengan pelarut metanol. Ekstrak diuji kandungan total fenol dengan reagen Folin Ciocalteu dan aktivitas antioksidannyadengan metode DPPH (2,2-diphenyl-1-picrylhydrazyl). Hasil penelitian menunjukkan bahwa keempat sampel memiliki kadar air, kadar abu,kadar protein dan kadar lemak yang bervariasi. Kadar lemak bervariasidari 16,80±0,05 % - 23,75±0,04%db, dengan kadar tertinggi dimiliki bekatul varietas Sintanur (23,75±0,04 %db) dan terendah bekatul varietas IR-64 (16,80±0,05 %db). Kadar serat larut (soluble fiber, SF) bekatul dari keempat varietas padi tidak berbeda secara signifikan yaitu antara 4,07 - 4,14 %db namun terdapat perbedaan pada kandungan serat tak larut (insoluble fiber, IF) yang berpengaruh pada kadar serat total. Kandungan serat tak larut bekatul dari varietas Menthikwangi,Rajalele, IR-64 dan Sintanur berturut-turut adalah 27,64±0,46, 28,04±0,25, 29,15±0,26 dan 30,44±0,60 %db. Kandungan total fenol dan aktivitas antioksidan tertinggi dimiliki oleh ekstrak hidrofilik dari bekatul verietas Menthikwangi, masing-masing 2794,28±181,83 μg ekivalenasam galat (EAG)/g bekatul untuk total fenol dan aktivitas antioksidanmenangkal (scavenging) radikal bebas (DPPH) sebesar 41,28±0,60 %.Kata kunci: Bekatul, ekstrak hidrofilik, varietas padi, fenol, DPPH

Fish has a high nutritional value and is a major food source in many countries. Fish lipid has a high content of polyunsaturated fatty acids (PUFAs), especially eicosapentaenoic acid (EPA; 20: 5n-3) and docosahexsanoic acid (DHA; 22: 6n-3). The objective of this research was to determine fatty acids composition of snapper (Lutjanus sp) fillet and its damage during the storage process. The results showed that total of the saturated fatty acids (SFA) increased from 4.35% to 25.55%, 28.06%, 32.73%, and 61.75% during storage at 0 °C, 10 °C, 20 °C, 30 °C, and 40 °C, respectively. Total mono-unsaturated fatty acids (MUFA) were 23.72%, 23.69%, 14.4%, 22.66%, and 29.4% at storage temperature of 0 °C, 10 °C, 20 °C, 30 °C, and 40 °C. Total PUFA decreased from 25.06% to 15.98%, 14.99%, 10.32%, and 8.84% at 0 °C, 10 °C, 20 °C, 30 °C, and 40 °C. Peroxide value, as primary peroxide of snapper fillet, increased about 10.60 times with an increased in storage temperature from 0 °C to 40 °C. Value of TBA increased 6.60 times with an increased in temperature from 0 °C to 40 °C during 45 days.ABSTRAKIkan memiliki nilai gizi tinggi dan merupakan sumber makanan utama di banyak negara. Lipid ikan memiliki kandungan tinggi asam tak jenuh ganda (Poly Unsaturated Fatty Acid, PUFA), terutama asam eikosapentanoat (EPA; 20:5n-3) dan asam docosahexsanoat (DHA; 22:6n-3). Penelitian ini bertujuan untuk mengetahui komposisi asam lemak fillet ikan kakap (Lutjanus sp) dan kerusakan akibat proses penyimpanan. Hasil analisis asam lemak jenuh (Saturated Fatty Acid, SFA) menunjukkan bahwa asam lemak jenuh meningkat dari 4,35% menjadi 25,55%, 28,06%, 32,73%, dan 61,75% selama penyimpanan pada 0 °C, 10 °C, 20 °C, 30 °C, dan 40 °C. Total asam lemak tak jenuh (Mono Unsaturated Fatty Acid, MUFA) adalah 23,72%, 23,69, 14,4%, 22,66%, dan 29,4% pada penyimpanan 0 °C, 10 °C, 20 °C, 30 °C, dan 40 °C. Sedangkan total PUFA turun dari 25,05% menjadi 15,98%, 14,99%, 10,32%, dan 8,84%  pada penyimpanan 0 °C, 10 °C, 20 °C, 30 °C, dan 40 °C. Angka  peroksida sebagai produk primer dari oksidasi  fillet ikan kakap  meningkat 10,6 kali dengan kenaikan suhu dari 0 °C sampai 40 °C. Synthesis of fatty acid methyl ester from coconut oil has conducted by chemical methanolysis. The objectives of the research were to evaluate the effect of temperature reaction on yield and profile of fatty acid methyl ester and the fractionation process on medium chain fatty acids methyl ester content. Coconut oil was extracted from 11-12 months of Mapanget Tall coconut variety fruit. The oil was then analyzed to evaluate the physycochemical properties and fatty acids profile. Methanolysis reaction was conducted using potassium metoxide as a catalyst at 50, 55 and 60oC for 2 hours. The methanolysis products were then measured for the yield and were determined for fatty acids profile. Furthermore, the fatty acid methyl ester was franctionated by melting from solid to liquid form. The results showed that the coconut oil had a good quality and medium chain fatty acid content was 61.93%, whereas lauric acid was the major component about 48.24%. The yield of fatty acid methyl ester was 85.41-86.69% and medium chain methyl ester content were 60.81%, 61.84% and 59.85% at reaction temperature of 50oC, 55oC and 60oC, respectively. Ester methyl lauric was a major component of 48.84 – 49.04%. Dry fractionation process only increased 6.68% of fatty acids methyl laurate content.ABSTRAKPada penelitian ini dilakukan sintesis ester metil asam lemak minyak kelapa dengan cara metanolisis kimiawi. Tujuan dari penelitian ini adalah mengetahui pengaruh suhu metanolisis terhadap yield dan profil ester metil asam lemaknya serta perlakuan fraksinasi terhadap kandungan ester metil asam lemak rantai medium. Minyak kelapa diekstraksi dari daging buah kelapa varietas Dalam Mapanget umur buah 11-12 bulan. Minyak kelapa yang diperoleh dianalisis sifat kimianya dan profil asam lemaknya. Reaksi metanolisis dilakukan secara kimiawi menggunakan katalisator kalium metoksida pada suhu 50, 55 dan 60oC selama 2 jam. Hasil metanolisis diukur yield dan proporsi asam lemaknya. Selanjutnya ester metil yang dihasilkan difraksinasi dengan metode beku ke cair. Hasil analisis diperoleh bahwa minyak kelapa mempunyai kualitas yang baik dan asam lemak rantai medium sebesar 61,93% yang didominasi oleh asam laurat sekitar 48,24%. Yield ester metil yang dihasilkan berkisar 85,41-86,69% dengan total kandungan ester metil rantai medium yang dihasilkan berturut-turut 60,81%, 61,84% dan 59,85% masing-masing pada suhu 50oC, 55oC dan 60oC. Proporsi tertinggi adalah ester metil laurat yaitu sebesar 48,84 - 49,04%. Proses fraksinasi kering dengan metode beku ke cair hanya dapat meningkatkan proporsi ester metil laurat sebesar 6,68%.

ABSTRAKProtein ikan mudah rusak akibat oksidasi selama penyimpanan. Kecepatan reaksi oksidasi dapat didekati melalui orde ke nol maupun orde pertama. Penelitian ini bertujuan untuk mempelajari oksidasi selama penyimpanan dengan menentukan besaran energi aktivasi (Ea) dan konstanta perubahan (k). Hasil menunjukkan bahwa nilai k meningkat dari 0,0617 menjadi 0,311 dengan peningkatan suhu dari 0 °C ke 40 °C. Energi aktivasi reaksi oksidasi yang menyebabkan terjadinya oksidasi protein adalah 42,015 Kj/mol.K untuk orde nol. Kinetika peningkatan protein karbonil: semakin tinggi suhu penyimpanan protein ikan kakap (Lutjanus sp) semakin besar nilai konstanta (k) yang diperoleh. Studi kinetika juga memperlihatkan bahwa peningkatan laju reaksi kerusakan oksidasi protein ikan kakap selama penyimpanan mengikuti reaksi orde ke nol atau reaksi berjalan lambat.Kata kunci: Energi aktivasi; protein ikan; kinetika reaksi ordo nol; reaksi orde pertama ABSTRACT Fish protein is oxidased easily during storage. The oxidation reaction rate can be approached through the order to zero or first order. The objective of this research was to study the oxidation rate during storage by determining the amount of activation energy (Ea) and constant change (k). The results showed that the increased of temperature storage from 0 °C to 40 o C can increased the k value from 0.0617 to 0.311. The carbonyl content of red snapper protein isolate can be increased to higher level as storage temperature increase to 40 °C with higher level increase at higher temperature. The activation energy of oxidation reactions that cause oxidation of the protein is 42.015 Kj/mol.K to zero order. Kinetics increase in protein carbonyls: the higher the temperature storage protein isolate red snapper (Lutjanus sp), the greater the value of a constant (k) is obtained. Protein ikan dalam bentuk konsentrat telah digunakan sebagai suplementasi bahan pangan berprotein rendah untuk golongan rawan gizi (Wang dkk., 2010). Menurut Mercier dkk. (2011) penelitian oksidasi protein relatif baru dan pembentukan karbonil merupakan salah satu perubahan yang paling menonjol dalam protein teroksidasi.

Incorporation of pectin during biosynthesis of bacterial cellulose by Gluconacetobacter xylinus InaCC B404: Possibility for producing green food packaging. Biodiversitas 22: 2548-2553. Bacterial cellulose-pectin biocomposite is developed as a potential environmentally friendly food packaging material. The objective of this research was to investigate the effects of pectin supplementation to the growth medium of Gluconacetobacter xylinus on the properties of the biocomposite produced. Pectin was added into the growth medium at different concentrations, i.e.: 0.1, 0.2 and 0.3% (w/v). The bacterial cellulose-pectin biocomposites produced were analyzed for structure, mechanical strength, crystallinity, morphology, color and opacity. FTIR analysis suggested that interaction between bacterial cellulose and pectin was hydrogen bonding. Generally, pectin addition decreased the crystallinity, tensile strength, tear strength and lightness of biocomposite produced. The tensile and tear strength of bacterial cellulose-pectin biocomposites were in the range of 23.05-25.72 MPa and 13.30-15.72 g/?m respectively, comparable to those conventional non-biodegradable plastics such as high-density polyethylene and low-density polyethylene with opacity much lower than HDPE. These results suggesting the possibility of cellulose-pectin biocomposite as semi-translucent green food packaging.

The quality of tempe is influenced by raw materials, processing and type of inoculum used. Black soybeans can beused as raw material for making tempe that has quality like tempe made from yellow soybeans. This study aims to determine the effect of  inoculum type and duration of incubation on the mold growth, organoleptic properties and antioxidant activity of black soybean tempe. This study uses mallika black soybean varieties as raw material for making tempe. Black soybeans that have been discarded skin, soaked and steamed,  then mixed with inoculum derived from pure cultures of Rhizopus stolonifer, R. oligosporus and R. oryzae. After it was incubated for 24, 30, 36 and 42 hours at a temperature 25-27 oC. The parameters were used mold growth, organoleptic properties and antioxidant activity of black soybean tempe. Results showed the treatment inoculum type and duration of incubation, effect on mold growth, organoleptic properties and antioxidant activity. Mold growth increased up to 36-hour long incubation, then decreased. Panelists gave the highest value in tempe that were incubated for 36 hours. There are a tendency long incubation tempe increased antioxidant activity.The conclusion of this research has the characteristics of Rhizopus stolonifer  relatively highest mold growth, organoleptic properties and antioxidant activity  that compared to other types of mold at the old30-hour incubation.ABSTRAKKualitas tempe dipengaruhi oleh bahan baku, proses pengolahan dan jenis inokulum yang digunakan. Kedelai hitamdapat digunakan sebagai bahan baku pembuatan tempe yang mempunyai kualitas seperti halnya tempe yang terbuat dari kedelai kuning. Penelitian ini bertujuan untuk mengetahui pengaruh jenis inokulum dan lama inkubasi terhadap pertumbuhan jamu, sifat organoleptik dan aktivitas antioksidan tempe kedelai hitam. Penelitian ini menggunakan kedelai hitam varietas mallika sebagai bahan baku pembuatan tempe. Kedelai hitam yang telah dibuang kulitnya, direndam dan dikukus kemudian dicampur dengan inokulum yang berasal dari biakan murni Rhizopus stolonifer, R. oligosporus dan R. oryzae. Setelah itu diinkubasi selama 24, 30, 36 dan 42 jam pada suhu 25-27 oC. Parameter yang diamati adalah pertumbuhan jamur, sifat organoleptik dan aktivitas antioksidan tempe kedelai hitam. Hasil penelitian menunjukkan perlakuan jenis inokulum dan lama inkubasi berpengaruh terhadap pertumbuhan jamur, sifat organoleptik dan aktivtas antioksidan. Pertumbuhan jamur meningkat sampai lama inkubasi 36 jam, kemudian turun. Panelis memberikan nilai tertinggi pada tempe yang diinkubasi selama 36 jam. Ada kecenderungan lama inkubasi tempe meningkat aktivitas antioksidannya. Kesimpulan dari penelitian ini Rhizopus stolonifer mempunyai karakteristik relatif lebih tinggi pertumbuhan jamur, sifat organoleptik dan aktivitas antioksidan dibandingkan jenis jamur yang lain pada lama inkubasi 30 jam.

Penelitian ini dilakukan untuk mendapatkan data laju penurunan mutu filet ikan nila (Oreochromis niloticus) yang dikemas vakum dengan HDPE. Hasil analisis digunakan untuk menentukan indikator yang paling tepat untuk persamaan prediksi umur simpan menggunakan persamaan regresi. Dalam percobaan yang dilakukan filet ikan nila yang dikemas vakum dengan HDPE disimpan pada suhu 0, 10, 20, dan 30 o C. Parameter yang dianalisis adalah TVB-N, pH, TBA, Organoleptik (hedonik), dan TPC (aerob dan anaerob). Data yang dihasilkan menunjukkan bahwa suhu dan lama penyimpanan berpengaruh nyata terhadap laju penurunan mutu filet ikan nila (P<0,05). Berdasarkan tingkat kecepatan parameter mutu untuk mencapai limit toleransi, nilai TVB-N merupakan parameter mutu yang paling tepat untuk dijadikan parameter penentu kinetika pembusukan filet ikan nila. Kandungan TVB filet ikan nila yang disimpan pada suhu 30, 20, 10, dan 0 °C telah melampaui batas mutu (30 mg-N/100 g) secara berturut-turut pada penyimpanan 9, 24, 72, dan 168 jam. Berdasarkan hasil pengolahan data nilai TVB filet ikan nila pada beberapa suhu penyimpanan menggunakan persamaan Arrhenius, nilai Ea yang didapatkan sebesar 72,17 KJ/mol dengan menggunakan nilai TVB 30 mg-N/100 g sebagai nilai batas penolakan mutu filet ikan nila. Persamaan prediksi umur simpan (t=jam) filet ikan nila dalam HDPE vakum yang didapatkan adalah ln A = ln A 0 + (t.exp [26,44-8681(1/T)]) dengan tingkat akurasi nilai prediksi terhadap nilai mutu filet ikan nila percobaan adalah 73-78%.
