ABSTRAKTujuan dari penelitian ini adalah mengetahui pengaruh penambahan kuning telur dalam pengencer CEP-2 terhadap motilitas dan viabilitas spermatozoa sapi Limousin. Penelitian menggunakan rancangan acak kelompok dengan 10 kali ulangan dari 10 ejakulat dari sapi yang berbeda. Spermatozoa disimpan selama delapan hari. Motilitas spermatozoa diamati dengan mikroskop cahaya (200x) pada suhu 37 C. Viabiltas spermatozoa diamati dengan pewarnaan eosin-negrosin menggunakan mikroskop cahaya (400x). Hasil penelitian menunjukkan suplementasi kuning telur pada pengencer CEP-2 berpengaruh terhadap motilitas dan viabilitas spermatozoa sapi Limousin selama penyimpanan pada suhu 4-5° C. Konsentrasi kuning telur terbaik adalah 20% dalam mempertahankan motilitas (44,25±3,92%) dan viabilitas (87,46±5,40%) spermatozoa sapi Limousin setelah penyimpanan delapan hari pada suhu 4-5° C. ____________________________________________________________________________________________________________________ Kata kunci: CEP-2, kuning telur, motilitas, viabilitas, spermatozoa sapi Limousin ABSTRACT The aim of this research was to study the effect of egg yolk supplementation in CEP-2 extender on motility and viability of Limousin bull sperm. Analisis cluster merupakan seperangkat metode yang digunakan untuk mengelompokkan objek ke dalam sebuah cluster berdasarkan informasi yang ditemukan pada data. Analisis cluster dapat diterapkan pada data runtun waktu, di mana terdapat prosedur dan algoritma pengelompokkan yang berbeda dibandingkan dengan pengelompokkan data cross-section. Banyak teknik pengelompokkan data runtun waktu yang dikembangkan di antaranya adalah penggunaan jarak pengukuran kemiripan yang sesuai dengan karakteristik data runtun waktu, pemilihan algoritma pengelompokkan yang optimal sampai dengan penentuan banyaknya cluster yang representatif. Tujuan dari penelitian adalah untuk memperoleh jarak pengukuran kemiripan terbaik, kemudian memperoleh algoritma pengelompokkan metode agglomerative yang optimal serta memperoleh jumlah cluster yang representatif. Pemilihan jarak pengukuran kemiripan terbaik dan algoritma yang optimal menggunakan koefisien korelasi cophenetic, sedangkan untuk penentuan jumlah cluster menggunakan koefisien silhouette. Data pada penelitian adalah data jumlah penduduk Kabupaten/Kota di Provinsi Kalimantan Timur dari Tahun 2005-2017. Berdasarkan hasil analisis, diperoleh jarak pengukuran kemiripan terbaik dalam mengelompokkan Kabupaten/Kota di Provinsi Kalimantan Timur adalah jarak autocorrelation based distance (ACF) dengan nilai koefisien korelasi cophenetic sebesar 0,99. Algoritma pengelompokkan yang optimal adalah algoritma average linkage, dikarenakan memiliki nilai koefisien korelasi cophenetic yang terbesar diantara algoritma pengelompokkan lainnya, dengan jumlah cluster yang representatif berdasarkan koefisien silhouette adalah 2 cluster.
As an underground crop, tuberous root of sweet potato (Ipomoea batatas) is the main economical yield expected from various level of cultivation. Aside rich in carbohydrate, sweet potato root contains vitamin and mineral. The degree of utilization reflects the level of subsistence to commercialization, where the desire or needs can be easily met was defined as prosperity. Supplying continuously the demand of sweet potato for subsistence and commercialization oriented to bio-ethanol and food related industries depend on the sustainable production system. To sustain sweet potato production system, the severe biotic and a-biotic stresses have to be alleviated.
Sperm can change physiology and structure during storage in refrigerator temperature or frozen temperature that caused by cold shock or free radical. The aim of this study to evaluate ultrastructure and fertilizing ability of Limousin bull sperm after storage in cauda epididymal plasma-based (CEP-2) extender with or without 20% egg yolk concentration at refrigerator temperature. Semen sample collected from three Limousin bull were diluted with CEP-2 with 20% egg yolk and CEP-2 without egg yolk, cooled and stored at 4-5 degrees C during eight days. Sperm ultrastructure were observed with scanning electron microscopy (SEM). Fertilizing ability of Limousin bull sperm were assessed on cleavage rate of embryo using in vitro fertilization method. The percentage data were transformed into arcsine before being analysis with ANOVA and Duncan's multiple comparison test. The result of study showed morphologically normal sperm after storage in CEP-2 with 20% egg yolk, whereas in CEP-2 without egg yolk morphologically abnormal sperm especially neck was fractured and head was destroyed. Fertilizing ability of Limousin bull sperm were significantly higher in CEP-2 extender with egg yolk 20% (74.29 +/- 4.95%; p < 0.05) than without egg yolk (30.00 +/- 12.02%; p < 0.05). Egg yolk 20% in CEP-2 extender protected ultrastructure and fertilizing ability after storage during eight days.
COVID-19 is a syndrome affecting pulmonary function but rather in serious conditions leads to death. Kencur (Kaempferia galanga L.) is a type of rhizome plant in Indonesia that is used as an herbal medicine called Jamu because it is believed to be able to cure various types of diseases. One of which is for anti-virus. The goal of this study was to see how effective the compounds in kencur are against COVID-19 with a molecular docking strategy. Kencur biological activities were obtained from the library and the design of the Acute Respiratory Syndrome Main protease (Mpro) has been gained from the protein data bank website. In addition, the biological activities in kencur were examined utilizing Lipinski's five-point concept was used to evaluate their substance molecular characteristics. Molecular docking analysis was performed with the PyRx Virtual Screening Tool software. The PyRx program was used for molecular docking simulation. While, the Discovery Studio Visualizer program was used to visualize the interaction between SARS-CoV-2 (Mpro) and the pharmacologically active metabolites in kencur. The docking evaluation on three antiviral substances revealed that Quercetin had the lowest binding energy when bound with Mpro and thus had the greatest potential as a viral inhibitor.
Analisis data time series menggunakan metode Winter's exponential smoothing dan Pegel's exponential smoothing merupakan analisis data yang dipengaruhi oleh pola data musiman. Winter's exponential smoothing merupakan metode peramalan yang mengasumsikan pola data bersifat trend aditif sedangkan Pegel's exponential smoothing menyajikan sembilan model klasifikasi yang memisahkan faktor trend dan musiman. Penelitian ini bertujuan untuk memperoleh model yang tepat dan hasil peramalan dari data produksi kelapa sawit Provinsi Kalimantan Timur periode Januari 2014 sampai Desember 2017. Hasil peramalan diverifikasi menggunakan metode tracking signal. Hasil penelitian menunjukkan bahwa model musiman multiplikatif tanpa trend pada metode Pegel's exponential smoothing dengan nilai MAPE sebesar 7,04% memiliki akurasi peramalan yang lebih baik daripada metode yang lainnya. Berdasarkan pemantauan menggunakan tracking signal diperoleh satu hasil peramalan yang bersifat bias. Model musiman multiplikatif tanpa trend dapat digunakan untuk meramalkan 3 bulan ke depan yaitu Januari, Februari dan Maret Tahun 2018. Hasil peramalan 3 bulan ke depan mengalami penurunan secara berturut-turut.
: This research was conducted at the Center of Artificial Insemination (BBIB) Singosari , Malang, East Java. The purpose of this research was to determine the parameters of fresh semen quality including volume, pH, individual motility and concentration of frozen semen production and recovery rate of Bali bulls in different seasons. The material used were fresh semen of 14 heads of Bali's bull in 3, 5, 6, 10, and 11 years and secondary data from November 2015 to September 2016 at BBIB Singosari. The method used in this research was case study and the data were analyzed descriptively. The results showed that the highest volume semen Bali bull in summer was 6.05±1.50 ml. The pH in rainy season and summer were 6.5±0.07. Semen of Bali's bull dominated by the white-milk color. The highest of color (white-milk), individual motility, concentration and total of frozen semen production were at summer in September. The average of color, individual motility, concentration and total of fozen semen production were 89%±0.10, 66.98 ± 0.05%, 1159.25 ± 211.28 million/ml and 22682 ± 707.95 straw. Post thawing Motility (PTM) and RR value were 45.28 ± 0.02%; 66.79± 0.05 semen of Bali's bull highest at summer in August. The conclusion showed the differences of season will affect to volume, color, individual motility, concentration, PTM, the value of RR and production of frozen semen Bali bull, but it is not effect for pH semen.
