Abstract. In this study, while Osmanli strawberry, which has some superior characteristics like taste, aroma and male sterility that makes breeding studies easier, has been used as maternal parent, three local (Karacilek, Tuylu and Deli) cultivars in Turkey and three standard (Kabarla, Sweet Ann and Sweet Charlie) strawberry cultivars were used as paternal parents. Molecular analyses were conducted for determination of polymorphism levels by using 14 UBC-ISSR primers for the examination of 52 F1 genotypes that included selections as a result of hybridization studies, parents that were used in hybridization studies, and Fragaria chiloensis. 76 DNA bands were obtained in total and 60 of them were determined as polymorphic. The resulting data of the evaluation of ISSR-PCR bands, were analysed in Popgene32 and MEGA5.0 packaged software and the dendrogram was obtained according to the UPGMA method. The dendrogram was separated into two major groups at the 20% difference level, one is larger and the other is smaller. While Fragaria chiloensis, Tuylu-2 local cultivar, CC48 and CA97 F1 genotypes participated in the small group, the other F1 genotypes and parents participated in the larger group.
The production of strawberry cultivation in wide ecological regions, its aroma, rich mineral content and taste affect the preference of consumers positively. The high amount of aroma of local cultivars enables them to be used as parents in breeding studies. In this study, it was aimed to determine the morphological and pomological characteristics and phytochemical contents of the superior F1 strawberry genotypes selected after hybridization with the local Osmanli strawberry under the ecological conditions of Bilecik province. Morphological and pomological analyzes were made and the best ten genotypes were selected according to the weighted grading method. According to this, the titratable acidity amount of the selected genotypes was determined between 0.28–0.51, the total soluble solid amount was determined between 5–10.5%, and the pH amount was determined between 3.3–3.78. By looking at the average fruit weight, fruit width and fruit length values, genotypes that can compete with commercial cultivars were selected and these were determined as table genotypes. While the total phenolic content of the ten genotypes selected by phytochemical analyzes was found between 2675.56–3983.89 μg GAE/g TA, the total amount of anthocyanin was determined between 156.19–39.12 μg Plg-3-glu/gta. The antioxidant amount was 18.95 μmol TE/ g ta-7.54 μmol TE/ g in the FRAP method.
Bu çalışma, Bilecik koşullarında 2020-2021 yıllarında Heritage, Tulameen, Canby ahududu çeşitlerinin bazı fiziksel ve kimyasal özelliklerinin belirlenmesi amacıyla Bilecik Şeyh Edebali Üniversitesi Tarımsal Uygulama ve Araştırma Merkezine ait uygulama alanında yürütülmüştür. Ortalama meyve ağırlıkları bakımından denemenin iki yılında da çeşitler arasında istatistiki açıdan bir fark bulunmamıştır. İki yılda da ortalama meyve ağırlığı ve meyve eni incelendiğinde en iyi sonuç Heritage çeşidinden elde edilmiştir. 2020 yılında en yüksek meyve boyu Tulameen çeşidinde (19.30 mm), 2021 yılında ise Heritage çeşidinde (19,62 mm) tespit edilmiştir. 2021 yılında Suda çözünebilir kuru madde miktarı en yüksek Heritage (% 11.35) çeşidinde, pH Tulameen (3.77) çeşidinde, titre edilebilir asit miktarı ise Canby (% 4.03) çeşidinde belirlenmiştir. Denemenin iki yılında alınan sonuçlar değerlendirildiğinde Heritage çeşidinin ön planda olduğu belirlenmiştir.
This study was carried out to determine the genetic diversity of eleven service tree (Sorbus domestica L.) genotypes selected from Tokat province and taken under protection in the genetic resource plot. In the present study, ISSR-PCR studies were carried out on genotype DNAs obtained by Mini-CTAB method and the level of polymorphism between genotypes was determined us- ing seven UBC-ISSR primers. A total of 65 bands were obtained in service tree genotypes with seven primers and 56 of these bands were determined to be polymorphic. The number of bands obtained per primer varied between 5 and 12, the mean band number was 5.90, and the mean polymorphic band number was 5.09. The data obtained as a result of the evaluation of ISSR-PCR bands were analyzed in Popgene32 and MEGA5.0 computer package program, and a dendrogram was obtained according to the UPGMA method. In the dendro- gram obtained, it was determined that two groups were formed at the level of approximately 22% difference, and there was only one genotype in the small group while the other genotypes were in the large group.
