Acute exposure to mercury chloride (HgCl 2 ) causes acute kidney injury (AKI). Some metals interfere with protein folding, leading to endoplasmic reticulum stress (ERS), and the activation of cell death mechanisms, but in the case of mercury, there is no knowledge about whether the ERS mediates tubular damage. This study aimed to determinate if HgCl 2 causes an AKI course with temporary activation of ERS and if this mechanism is involved in kidney cell death. Male mice were intoxicated with 5 mg/kg HgCl 2 and sacrificed after 24, 48, 72, and 96 h of mercury administration. The kidneys of euthanized mice were used to assess the renal function, oxidative stress, redox environment, antioxidant enzymatic system, cell death, and reticulum stress markers (PERK, ATF-6, and IRE1a pathways). The results indicate temporary-dependent renal dysfunction, oxidative stress, and an increase of glutathione-dependent enzymes involved in the bioaccumulation process of mercury, as well as the enhancement of caspase 3 activity along with IRE1a, GADD-153, and caspase 12 expressions. Mercury activates the PERK/eIF2a branch during the first 48 h. Meanwhile, the activation of PERK/ATF-4 branch allowed for ATF-4, ATF-6, and IRE1a pathways to enhance GADD-153. It led to the activation of caspases 12 and 3, which mediated the deaths of the tubular and glomerular cells. This study revealed temporary-dependent ERS present during AKI caused by HgCl 2 , as well as how it plays a pivotal role in kidney cell damage.
değişiklikleri ile ilgili yönetmelikler kapsamında, insan, hayvan ve çevre sağlığını iyileştirmeye yönelik olarak kurumların bilimsel ve teknolojik imkanlarından faydalanarak sahipsiz köpeklerin rehabilitasyonu ve viral enfeksiyon proflaksisinin sağlanmasıdır. 12 aylık çalışma kapsamında, Osmaniye ilinde yaşayan 296 sahipsiz köpekten 259'una rehabilitasyon uygulandı. Hastalık görülen 106 köpekten 77'de (%72,6) viral antijen saptandı ve hematolojik ve serolojik muayeneler yapılarak tedavi uygulandı. Köpeklerden 47'sinde (%44,3) Canine Parvovirüs (CPV), 9'da (%8,5) Canine Distemper Virüs (CDV),21'de (%19,8) Canine Coronavirüs (CCoV) ve 11'de (10,4) CPV ve CCoV koenfeksiyonu saptandı. Tüm köpeklere profilaktik ekto-ve endoparaziter tedavi uygulandı. Sağlıklı ve tedavi sonrası iyileşen 255 köpeğe Kuduz, CPV, CDV, Canine Adenovirus Tip-1 ve Tip-2 (CAV-1 ve CAV-2), Canine Parainfluenza Virüs (CPIV-2) ve Leptospira spp. enfeksiyonlarına karşı koruyucu aşılama uygulandı. 36 köpek sahipledirildi, 206'sı kayıtsızdı ve kulak küpesi ile mikroçip takılarak kayıt altına alındı, aktif olan 189'u kısırlaştırıldı ve hepsi bulundukları yere geri bırakıldı. Bu sayede Osmaniye ilinde yaşayan köpeklerde viral enfeksiyon hastalıklarının araştırılması, hasta ve taşıyıcı hayvanların tespiti, tedavisi ve bölgesel enfeksiyon kontrolü için koruyucu aşı çalışması yapıldı. Kuduz hastalığı ölümcül bir zoonotik enfeksiyon olup sahipsiz hayvanların aşılanmaları rutin olarak ülke genelinde yaygın bir uygulamadır ve kanun ve yönetmelikler ile sağlanmaktadır. CPV-2, CDV, CAV-1 ve CAV-2, CPIV-2 ve Leptospira spp. aşıları köpekler için yapılması gerekli aşılardır (Core Vaccines) ancak sahipsiz köpeklerin aşılanması ve mikroçip uygulaması ülkemizde yaygın bir uygulama değildir. Bu çalışma sahipsiz köpeklerin refahının sağlanması açısından bir 'İyi Veteriner Hekimlik ve Hayvan Refahı' uygulamasıdır. Halk, hayvan ve çevre sağlığının iyileştirilmesi açısından bir 'Tek Sağlık' uygulamasıdır. Sonuç: Bu çalışma Osmaniye Belediyesinin "Mutlu Şehir" projesini desteklemiştir. Yerel Yönetim, Üniversite ve Sivil Toplum Kuruluşu işbirliği ile 12 ayda 255 sahipsiz köpeği rehabilite edilerek ve aşılanarak viral enfeksiyon profilaksisi sağlanmıştır.
The equine locomotor system's health plays a key role on athletic performance. Bone and joint diseases are the major causes of lameness. Poor performance and diseases lead to great economic loss to equestrian sports and horse breeders. Therefore, prevention, early diagnosis, and therapy of joint diseases are important. A disintegrin-like and metalloproteinase with thrombospondin motifs (ADAMTS) proteinase family plays an important role in many physiological processes such as tissue reorganization, coagulation, and angiogenesis. Aggrecan proteinases ADAMTS-4 and ADAMTS-5 are physiologically responsible for the restructuring with enzymatic cleavage of the cartilage, specific biomarkers in the synovium or body fluids for early diagnosis, and potential specific therapeutic targets in order to their role on degenerative joint diseases physiopathology in humans and various animals.
The present study’s objective is to clarify the molecular mechanisms of tannic acid effects on the viability of human colorectal carcinoma (CRC). Tannic acid is stable for up to 48 h and is localized in both cytoplasm and nucleus. It dose-dependently inhibited the viability of CRC cell lines; SW-620 and HT-29 with IC 50 values of 7.2 ± 0.8 and 37.6 ± 1.4 µmol L–1. Besides, metastatic, invasive, and colony formation properties of CRC cells were significantly inhibited following the tannic acid treatment (p < 0.001). Tannic acid has been found to modulate enzyme, protein, and gene expressions of NQO1 in different levels and the upregulation of protein/gene expressions of p53 (p < 0.001), which leads the cells to trigger apoptosis. In conclusion, the present in vitro study may supply a significant background for in vivo studies in which the molecular mechanisms of antioxidant and chemopreventive activities of tannic acid will completely clarify.
This study was conducted to determine the serum levels of interleukin (IL)-1β, IL-6, IL-10 and tumor necrosis factor alpha (TNF-α) in thyroidectomized rats. Hypothyroidism was performed by surgical thyroidectomy in rats. Four weeks after from surgical procedure, selected cytokines levels were evaluated. Hypothyroidism was confirmed by elevated thyroid stimulating hormone (TSH) and decreased free tri-iodothyronine (fT3) levels (P<0.05) in thyroidectomized group. Serum IL-1β, IL-6, IL-10 levels have shown slight increase, whereas only TNF-α level (P<0.05) were significant statistically in thyroidectomized rats compared with control group. In conclusion, the obtained data suggest that elevated levels of cytokines could be as a consequence of thyroidectomy operation. Hipotiroidizm cerrahi yolla tiroidektomi yapılarak gerçekleştirildi. Cerrahi işlemden sonraki 4. haftada, seçili sitokinlerin seviyeleri ölçüldü. Hipotiroidizm, tiroidektomize grubun tiroid uyarıcı hormon (TSH) düzeyindeki artış (P<0.05) ve serbest tri-iyodotironin (fT3) düzeyindeki azalış (P<0.05) ile teyit edildi. Tiroidektomize ratların serum IL-1β, IL-6, IL-10 seviyelerinin kontrol grubuna kıyasla hafif düzeyde yükseldiği ancak, sadece TNF-α düzeyindeki artışın istatistiki olarak anlamlı (P<0.05) olduğu görüldü. Sonuç olarak elde edilen veriler, sitokin seviyelerindeki yükselmenin, tiroidektomi operasyonunun bir sonucu olabileceğini düşündürmektedir.
