Benzene is a ubiquitous environmental pollutant and an important industrial chemical present in both gasoline and motor vehicle emissions. Occupational human exposure to benzene occurs in the petrochemical and petroleum refining industries as well as in gas-station workers, where it can lead to benzene poisoning (BP), but the mechanisms of BP are not completely understood. In Brazil, a significant number of gas-station service workers are employed. The aim of the present study was to evaluate alterations related to BP and metabolic polymorphisms in gas-station service workers exposed to benzene in the city of Rio de Janeiro, Brazil. Occupational exposure was based on clinical findings related to BP, and metabolic polymorphisms in 114 Brazilian gas-station attendants. These workers were divided into No Clinical Findings (NCF) and Clinical Findings (CF) groups. Neutrophil and Mean Corpuscular Volume (MCV) showed a significant difference between the two study groups, and neutrophil has the greatest impact on the alterations suggestive of BP. The clinical findings revealed higher frequencies of symptoms in the CF group, although not all members presented statistical significance. The frequencies of alleles related to risk were higher in the CF group for GSTM1, GSTT1, CYP2E1 7632T > A, but lower for NQO1 and CYP2E1 1053C > T genotypes. Moreover, an association was found between GSTM1 null and alterations related to BP, but we did not observe any effects of other polymorphisms. Variations in benzene metabolizing genes may modify benzene toxicity and should be taken into consideration during risk assessment evaluations.

Resumo Introdução: a avaliação de uma exposição mensura sua intensidade, frequência e duração, podendo detectar danos precoces que, se ignorados, podem evoluir para um quadro nocivo. Nos campos da saúde ambiental e ocupacional, os biomarcadores de genotoxicidade tem sido largamente utilizados para essa avaliação. Objetivo: identificar, descrever e discutir os principais bioindicadores de genotoxicidade e seu uso conjunto com técnicas de avaliação de expressão gênica em estudos de exposição ocupacional ao benzeno em postos de revenda de combustíveis (PRC). Métodos: revisão bibliográfica de trabalhos publicados entre 1995 e 2015. Resultados: as técnicas identificadas foram: ensaio cometa, estresse oxidativo, micronúcleos, aberrações cromossômicas, polimorfismos, adutos de DNA e proteínas, fatores epigenéticos e expressão gênica. Foi observado que testes de danos genéticos e epigenéticos são utilizados em frentistas de PRC que participam de programas de saúde do trabalhador ou de pesquisas, embora um baixo número de publicações sobre o tema tenha sido identificado. Esse fato talvez possa ser explicado pelos poucos países onde a profissão persiste e pelas limitações para o desenvolvimento de pesquisas nesses países. Conclusão: os bioindicadores de genotoxicidade e as técnicas de expressão gênica são úteis na detecção de dano precoce desta exposição ocupacional e devem ser avaliados em conjunto.

The importance of atmospheric lead in environmental global contamination is receiving increasing scientific attention. The main exposure monitoring approach is biological, but the environmental one has a key role, since the environment is the major source of exposure. The study aimed to identify the contamination sources and potential risk factors of the exposure in a 64 subject group formed by 0-16 years-old children from an economically deprived community in Rio de Janeiro. Lead concentrations in soil, water, dust and air were determined and neurological and carcinogenic risk factors for ingestion and inhalation were calculated. Blood samples were collected and used in the analysis of Pb-B, ALA-D% and ALA-D genotyping. The observed neurological risk factor was 549 times higher than reference dose for dust and 554 times higher in the case of ingestion. The carcinogenic risk factor for ingestion was about 4 times. Mean Pb-B was 5.6 microg/dL and 40% of the children presented Pb-B levels above the 6 microg/dL cutoff value. Mean ALA-D% was 40.3% and a correlation between Pb-B and ALA-D% was observed. ALAD1-2 genotype was identified in 10% of the children. The results will allow the understanding of our reality, supporting public health and environment organizations to carry out control actions and all-encompassing environmental surveillance.

O uso indiscriminado de um número cada vez maior de substâncias químicas vem aumentando e a contaminação ambiental associada tem trazido sérias consequências para o sistema público de saúde devido à elevação de danos para a saúde humana. Uma das substâncias que desperta grande interesse devido à contaminação contemporânea é o benzeno, composto aromático classificado pela International Agency for Research on Cancer como reconhecidamente carcinogênico para humanos. O objetivo do presente estudo foi elaborar e discutir um panorama sobre a contaminação por benzeno, seu metabolismo, consequências para a saúde e sua determinação ambiental e biológica a partir de informações existentes na literatura científica. O levantamento de dados possibilitou o acesso a mais de 200 artigos científicos tanto de âmbito nacional quanto internacional, demonstrando a atualidade do tema e a necessidade de minimização da exposição humana a essa substância. A maioria preocupa-se em explorar o metabolismo e investigar indicadores de exposição, muitos já amplamente estudados e com sérias limitações. Contudo, um crescente número de pesquisadores estão empenhados em elucidar fatores relacionados à suscetibilidade e à interferência da exposição no material genético e proteico. Indicadores de exposição inovadores têm sido propostos com o objetivo de complementar as lacunas de informações anteriormente obtidas, contribuindo para o delineamento da estrutura da biologia de sistemas orgânicos frente à exposição ao benzeno.

Resumo Introdução: trabalhadores de postos de combustíveis estão expostos às diversas substâncias químicas presentes no ambiente de trabalho, destacando-se entre elas o benzeno, devido às suas propriedades carcinogênicas. Objetivo: avaliar os danos genotóxicos relacionados à exposição ocupacional ao BTEX (benzeno, tolueno, etilbenzeno, xilenos) em trabalhadores de cinco postos de combustíveis do município do Rio de Janeiro, RJ. Metodologia: foram analisadas concentrações de BTEX no ar; atividades das enzimas catalase e glutationa S-transferase; e ensaio cometa em amostras de sangue total de 97 trabalhadores. Resultados: as concentrações de BTEX estavam dentro dos valores preconizados pela NR 15, incluindo Anexo 13-A. Entretanto, uma oscilação nos resultados de ensaio cometa foi observada entre os trabalhadores dos diferentes postos de combustíveis, principalmente em trabalhadores de postos com menores concentrações de benzeno. Discussão: esse resultado está de acordo com a literatura científica atual, que indica uma curva dose-resposta supralinear para o benzeno, observando-se em baixas concentrações um aumento não linear do risco de leucemia, provavelmente relacionado à maior metabolização do benzeno e à maior produção de seus metabólitos tóxicos nessas concentrações. Conclusão: os resultados deste estudo sugerem que a exposição ao BTEX, mesmo em baixas concentrações, contribui para o risco genotóxico à saúde humana.

Benzene is one of the most important substances for assessment, due to its significant use, the environmental contamination resulting from its emission and the effects on human health. It is classified by the International Agency for Research on Cancer (IARC) as a known carcinogen to humans (group 1) and associated with the development of leukemia. In general, the population is exposed to this substance by inhaling contaminated air, which varies according to the location and intensity of its potential sources. The petrochemical industry is one of the most important sources of this compound. The municipality of Duque de Caxias, specifically the Campos Elíseos district, in Rio de Janeiro State, Brazil, houses the Industrial Complex of Campos Elíseos (PICE), a grouping of over 25 industries, which includes the second largest oil refinery in Brazil. Environmental contamination from the PICE has been recognized, but there is a lack of studies concerning its impact on the health of the surrounding population. S-phenylmercapturic acid (S-PMA) concentrations ranging from 0.80 to 8.01μg.g-1 creatinine were observed in the local population, apparently related to hematological changes also observed in exposed population. The quantifiable presence of urinary S-PMA from the benzene metabolism is associated with the fact that 60% of the participants present specific hematological changes, which may be due to the environmental benzene exposure. The allele and genotype frequencies of the CYP2E1 and NQO1 enzymes observed in the study population were similar to those reported in other studies. The presence of the variant allele in the NQO1 genotype may be a risk factor for the observed hematological changes.

Introduction: Tobacco smoking represents the leading preventable cause of morbidity and mortality in adults. Nicotine is responsible for tobacco addiction and is known to elevate brain dopamine, leading to feelings of pleasure and reward. The MAOA enzyme is responsible for degrading dopamine. It is known that a polymorphism in the promoter region of the MAOA gene has been implicated in smoking behavior. In this study, we investigated the association between the MAOA VNTR polymorphism and smoking behavior in Brazilian males. Methods:A cross-sectional study was conducted with 121 Brazilian males over 18 years of age. The polymorphism was genotyped using PCR. Multiple logistic regression were used to verify the association of MAOA gene polymorphisms with smoking status. Results:We not found significant association between the MAOA polymorphism and smoking status. Likewise, no association of the VNTR polymorphism with elements of smoking behavior, such as smoking duration, smoking initiation, relapses, smoking cessation, and degree of nicotine dependence, was observed. Conclusions:In conclusion, no association was observed between the MAOA polymorphism, smoking status and smoking behavior in population of Brazilian males studied. Further studies with larger sample sizes will be needed to elucidate these issues.
