Twenty Holstein calves were used to investigate the effects of mannanoligosaccharides (MOS) supplementation in the whole milk on growth performance, faecal score, faecal pH, selected faecal bacterial populations and health during the preweaning period. Healthy calves selected by clinical examination were allocated to one of the two groups (control [CG] and experimental [EG]) at 5 days old. Each group consisted of 5 male and 5 female calves. Each calf in EG was supplemented with 7 g/d of a MOS product (Celmanax) from 5 days to 56 days of age. MOS supplement was mixed with the whole milk once in the morning and administered to the calves in EG via nipple bottle, whereas the calves in CG were fed the whole milk without MOS. Calves were weaned at 56 days of age. The final body weight, average daily weight gain (ADG) and average daily feed intake (ADFI) were statistically similar (p>0.05) but were higher by 3.70%, 6.66%, and 10.97%, respectively, in MOS than in control calves. Feed efficiency (ADG/ADFI) was also similar in two calves group. While faecal scores did not differ on day 5, 7, 14, 21, 28, 42, 49, and 56 between groups, EG had a higher faecal score (p = 0.05) than CG on day 35. Faecal concentration of Lactobacillus was lower (p<0.05) in EG compared with CG. No differences (p>0.05) in faecal concentrations of Bifidobacterium, Clostridium perfringens, and Escherichia coli were found between groups. Although there were no significant differences (p>0.05) in the incidence of diarrhoea, treatment days for diarrhoea and the costs associated with diarrhoea treatments between groups, collectively, the observed reductions in treatment days and the cost of diarrhoea treatments accompanying increases in final body weight, ADG and ADFI for EG may indicate potential benefit of MOS in treatment of diarrhoea.
Introduction: The aim of the study was to investigate changes in the serum levels of adiponectin and TNF-α, as well as insulin sensitivity, and to elucidate the possible relationship among the parameters and negative energy balance during the periparturient period of dairy cows. Material and Methods: Thirty primiparous Holstein dairy cows were selected for the study. Blood samples were collected from each cow seven days before the expected calving date, on the calving day, and 7, 14, and 21 days after calving. Blood non-esterified fatty acids (NEFA), β-hydroxybutyric acid (BHBA), glucose, insulin, adiponectin, and TNF-α levels were measured. Revised Quantitative Insulin Sensitivity Check Index (rQUICKI) was calculated using data on NEFA, insulin, and glucose concentrations. Results: When compared to prepartum levels, serum concentration of adiponectin significantly increased on day 21 postpartum. The rQUICKI increased and NEFA levels decreased on day 7 after parturition. Insulin and glucose levels decreased on days 7, 14, and 21 postpartum when compared with prepartum levels. BHBA levels decreased on day 21 and TNF-α concentration also decreased on days 7, 14, and 21 postpartum. Adiponectin levels positively correlated with NEFA during the preparturient period. Negative correlation was detected between adiponectin and rQUICKI on calving day and on 14 th day after parturition. TNF-α concentration positively correlated with glucose levels on day 7 prepartum and on 21 st day postpartum and with rQUICKI on 21 st day postpartum. Negative correlation was detected between adiponectin level and insulin sensitivity. Conclusion: Based on the results of the study, we concluded that adiponectin could possibly increase insulin sensitivity when blood NEFA concentrations are elevated.
Winter dysentery (WD) is a very contagious disease of cattle characterized by profuse diarrhea. The precise etiology of disease is still not fully elucidated. Aim of the presented study was investigation of the causative agents of disease in a herd affected by WD and to evaluate routine haematological and biochemical parameters of cows with WD. This study was carried out on blood and fecal samples collected from 12 cows showing typical signs of disease during an outbreak of WD in Bursa, Turkey. Samples were analysed for the presence of bovine coronavirus (BCoV), Bovine viral diarrhea (BVD), Eimeria oocysts and Campylobacter spp. using ELISA, RT-PCR, flotation technique and culture. Faecal samples were inoculated in HRT cell cultures for virus isolation. Isolated viruses were identified as coronaviruses from 25% of the samples by ELISA, PCR and serum neutralisation tests. Campylobacter spp. was isolated from 33.3% cows with dysentery. BVD and Eimeria oocysts were not detected in any of the samples. Results of the presented study indicates that BcoV along with Compylobacter spp. may be the primary agent of WD in cows. However negative results for BCoV and Campylobacter spp of 58 % of feacal samples indicates that etiology of the disease is still not fully elucidated and pathogens other than BCoV and Campylobacter could also be involved in pathogenesis of the disease
Background: Anomalies and fetal problems are encountered in all animals. Intestinal atresia is a congenital anomaly of both humans and animals. Intestinal atresia has four morphological types, and they can be observed in jejunum, ileum, cecum, colon, rectum and anus (type I: mucosal atresia, type II: atretic ends separated by fibrous cord, type III: atretic ends separated by a “V” shaped mesenteric gap or atretic end coil like an “apple peel”, type IV: multiple atresia). In calves, it is also commonly encountered; it is still a question to be replied about the exact prognosis of the patients with intestinal atresia. Thus, the main objective of the present study was to evaluate peritoneal fluid analysis results of calves with intestinal atresia.Materials, Methods & Results: Twenty-two calves were presented with abdominal distension and lack of defecation. Clinical and radiological findings pointed out the intestinal atresia. Blood samples were collected for routine hematological and serum total protein (TP) analysis. Peritoneal fluids were aspirated with sterile technique from the caudal abdomen, and biochemical features, cell contents and microbiological cultures of the peritoneal fluids were analyzed. Following to preoperative, surgical and necropsy findings, intestinal atresia were diagnosed as type II, III and IV in calves. Neutrophilic leukocytosis was observed in calves with type IV intestinal atresia. Red blood cells and platelet counts were determined in the peritoneal fluids. TP and density values of the peritoneal fluid were high in all cases. These results pointed the presence of the acute infection and peritonitis. Statistical comparison of each parameter showed that there was no significant difference between the types of intestinal atresia. Microbiologically, fecal origin bacteria were cultured in 12 calves.Discussion: In general, four types of intestinal atresia are encountered in calves, and all types are fatal pathologies; thus, surgery should urgently be planned as soon as diagnostic work-ups are completed after birth. Peritoneal fluid analysis is a useful diagnostic choice because changes in the peritoneal fluid parameters help to diagnosis of the intra-abdominal pathology. Intestinal atresia as more common anomaly in Holstein, Jersey and Montafon breed calves. In animals with intestinal atresia, surgical treatment is not usually recommended due to economic reasons and small chance of postoperative success. Long-term survival rate of surgically treated animals depends on the type of intestinal atresia and applied surgical technique. Atresia recti and ani may occur simultaneously with atresia coli, and all atresias (atresia coli, ani, recti or ani et recti) can be successfully treated by surgery. Continuous distention of the large intestinal segments leads to ischemia, necrosis, peritonitis and bowel perforation. Peritoneal fluid analysis should include the classic parameters for diagnosis, but these analyses are often insufficient to identify the etiology or pathogenesis of the disease. An increase in total WBC count and percentage of the neutrophil cells in the peritoneal fluid indicate the acute infectious origin inflammation. Additional parameters in the peritoneal fluid analysis have been established to improve diagnostic precision and specific information. Presence or absence of the bacteria in the peritoneal fluid samples is important to characterize the transudate and exudate. Changings in the peritoneal fluid values does not related to type of the intestinal atresia in calves.
This paper presents the case of a six year-old, male, thoroughbred horse with clinical signs of inappetence, weakness, and incoordination when walking. Clinical examination showed that the horse staggered and leaned to the left side. Feedstuff was present inside and around its mouth. Salivation was increased and there was no reflex at the palpebrae and tongue. The horse had difficulty swallowing and the tone of its tail was reduced. Botulism was diagnosed based on the clinical signs. Antibiotic (ceftiofur) and fluid-electrolyte treatment was commenced. Next day, neostigmin was added to the horse's treatment, and it became recumbent. The horse's palpebral, tongue and tail reflexes returned partially after neostigmine methylsulphate treatment on the same day and it stood up on day four. However, it could not swallow anything during the whole week, so after getting permission from the owner, the horse was euthanized on day 10. Samples of the colonic content and blood serum were sent by courier to the laboratory for toxin neutralization, however, botulinum neurotoxins could not be detected. After that, serum samples from days 6 and 10 were sent to another laboratory for testing for botulinum neurotoxin antibodies by ELISA. Specific antibodies against botulinum neurotoxin type A were measured, indicating a previous, immuno-relevant contact with the toxin. This seroconversion for type A supports the clinical botulism diagnosis. Type A botulism is rarely seen in Europe and has been detected in a horse in Turkey for the first time. The aim of this study was to determine diagnostic value of cELISA in anaplasmosis in clinically suspected animals and to compare the cELISA results with the clinical examination results. For this purpose a total of 720 ruminants (457 cattle, 146 sheep, 117 goat) were examined in terms of clinical signs. Eighty-eight ruminants consisting of 61 cattle, 11 sheep and 16 goat which had the symptoms of anemia, fever, icterus, weakness, depression and lack of appetite were selected for the study. Blood samples were collected from the jugular vein of all clinically suspected animals and serum samples were separated. A commercially available competitive enzyme-linked immunosorbent assay (C-ELISA) kit was used for determine antibodies to Anaplasma species. cELISA based diagnosis revealed that 47 of 88 serum samples (53.4%) were positive for anaplasmosis. In serological examination Anaplasma specific antibodies were determined in 45.9% of cattle, 63.6% of sheep and 56.2% of goats. Seropositivity rate was statistically differ among the age groups of cattle and the highest seropositvity rate was found in <12 month age (P<0.005). However no difference was found in the seropositivity rate of Anaplasma in sheep and goat in relation to age group. From the data obtained in this study it can be concluded that clinical findings are not sufficient criteria for the diagnosis of anaplasmosis and must be supported by serological examination.
