Hesperidin has cytotoxic effect on MCF-7/Dox cells with IC50 of 11 µmol/L. Hesperidin did not increased the apoptotic induction combined with doxorubicin. Co-chemotherapy application of doxorubicin and hesperidin on MCF-7/Dox cells showed synergism effect through inhibition of Pgp expression.
The prevalence of degenerative diseases increases with age. Furthermore, various factors tend to trigger cells injury, thereby, causing inflammation. This study, therefore, aims to examine the anti-inflammatory mechanisms of steeping date seeds in middle age women. This is a quasi-experimental design with a pre- and post-test approach used to evaluate the anti-inflammatory effect of 2.5 g of steeped of date palm seed, consumed by 30 healthy middle-aged women per day (in 250 mL water) for 14 days. The final numbers (22 subjects) of recruited women were included in the statistical analysis. Their level of IL-1β, TGF-β, IL-6, TNF-α, IL-12, COX-1, COX-2, and PGE2 were determined using ELISA. The results showed that the expression of IL-1β, TGF-β, COX-1, and COX-2 in women significantly decreased after consuming date palm seed. Steeped of date seed acts as an anti-inflammatory by downregulating the expression of key proinflammatory mediators.
Long-term use of doxorubicin causes cancer resistance due to overexpression of P-glycoprotein (P-gp), a protein that plays a role in cell drug efflux. The purpose of this study is to determine the action of hesperetin in increasing the cytotoxicity of doxorubicin on MCF-7 cancer cells resistant to doxorubicin (MCF-7/DOX) through suppression of P-gp expression. Cytotoxic assay of single and combinational treatment of doxorubicin and hesperetin were performed by using MTT assay. Apoptosis evidence was examined by using double staining with acridine orange and ethidium bromide dyes, while Pgp expression was determined by using immunocytochemistry. Hesperetin reduced cell viability in dose dependent manner. Both MCF-7 ori and MCF-7/DOX cells gave different responses to hesperetin with the IC 50 values of >500μM and 267μM, respectively. Combining treatment of hesperetin and doxorubicin to MCF-7/DOX cells at the dose of 95μM and 230nM increased apoptosis evidence and suppressed P-gp expression. These results suggest that hesperetin enhances the anticancer effect of doxorubicin to resistance MCF-7 cells through suppression of P-gp expression.

Short Communication: The emergence and rise of indigenous thermophilic bacteria exploration from hot springs in Indonesia. Biodiversitas 21: 5474-5481. Indonesia is an archipelagic country located in the pacific ring of fire, and is estimated to cause numerous hot springs spread across the country. In addition, small living microbes have been explored in these locations since 1985. These microbes possess the ability to survive in areas with high temperature (more than 40oC-90oC), and are therefore termed thermophiles. Hence, massive explorations have been conducted on Java island and other unexplored areas at Sumatra to Papua in New Guinea islands. Moreover, a total of 71 hot springs characterized by the presence of thermophilic bacteria have been explored in Indonesia. These investigations ensue with various approaches, including through conventional and microbiological, 16S rRNA, as well as whole-genome sequencing methods. In addition to species exploration, the application of thermophiles has become a topic of interest from 1999, especially based on thermostable enzymes with the capacity to maintain activity at high-temperature conditions. These include amylase, protease, lipase, xylanase, esterase, and cellulase as the most common isolated form, which indicates the existence of significant extractable potentials. Hence, there is a need for further research in terms of both exploration and application purposes.
