Resumo -O objetivo deste trabalho foi indicar caracteres anatômicos em folhas de plantas micropropagadas de abacaxizeiro, visando ao aperfeiçoamento do protocolo de aclimatação. Utilizaram-se plântulas micropropagadas in vitro e aclimatadas por seis e dez meses, apresentando, em média, 3,1, 50,2 e 65 g, respectivamente. A densidade estomática foi determinada na face abaxial da epiderme, nas regiões basal, mediana e apical da folha, usando o delineamento inteiramente casualizado em esquema fatorial de 2x3 (dois ambientes de cultivo e três regiões da folha), em seis repetições. A espessura da hipoderme, parênquimas aqüífero e clorofilado foi determinada na região mediana da folha, usando-se o delineamento inteiramente casualizado com três tratamentos em quatro repetições. A estrutura básica da folha do abacaxizeiro não se modificou, entretanto, ocorreram diferenças na freqüência estomática, no espessamento da cutícula e paredes da epiderme, formato e sinuosidade das paredes das células do tecido aqüífero e presença de células papilosas nos diferentes ambientes de cultivo, indicando plasticidade fenotípica.Termos para indexação: Ananas comosus, ambiente de cultivo, micropropagação, aclimatação. Leaf anatomy of micropropagated pineapple plantsAbstract -The objective of this work was to study leaf anatomy of pineapple plants in order to improve acclimatization protocols. In vitro plantlets weighting an average of 3.1 g and greenhouse plantlets derived from in vitro stock material, after six and ten months of culture, weighting an average of 50.2 and 65 g, respectively, were used. Stomatal density was determined on the abaxial epidermis, at the basal, median, and apical portions of the leaf, using a completely randomized design under 2x3 factorial (two culture environments and three leaf regions) with six replicates. Thickness of the hypodermis, aquiferous and photosynthetic parenchyma were determined at the median portion of the leaf using a totally randomized design with three treatments and four replicates. The basic structure of the pineapple leaf under in vitro conditions did not change. However, stomatal frequency, cuticular and epidermal wall thickening, shape and sinuosity of the cell walls of aquiferous parenchyma and the presence of papillary cells were verified as a result of environmental conditions during culture, indicating phenotypic plasticity.
RESUMOO objetivo do trabalho foi determinar as condições mais favoráveis para a germinação in vitro de sementes e o crescimento inicial de plântulas de mangabeira. Após assepsia, sementes oriundas de frutos maduros foram inoculadas em tubos de ensaio contendo os seguintes tratamentos: T1-15 mL de meio de cultura MS; T2-15 mL de meio de cultura MS + 2,0 g L -1 de carvão ativado; T3-15 mL de meio de cultura ½ MS; e T4-15 mL de meio de cultura ½ MS + 2,0 g L -1 de carvão ativado. Todos os meios de cultura foram gelificados com 0,3 g L -1 de Phytagel® e suplementados com 3,0 g L -1 de sacarose. O experimento foi instalado em delineamento inteiramente casualizado com quatro tratamentos e oito repetições, sendo cada parcela experimental composta de dez tubos de ensaio contendo uma semente cada. Não houve diferença significativa dos tratamentos para a porcentagem de germinação aos 20 dias, que variou de 95 a 100%. Quanto ao comprimento da raiz principal, observou-se que o meio de cultura constituído de ½ MS com 2,0 g L -1 de carvão ativado proporcionou maior crescimento quando comparado com os demais tratamentos. Aos 50 dias, não foi observada a formação de plântulas anormais e nem diferenças significativas do comprimento da parte aérea das plântulas. Entretanto, a diluição em 50% dos sais do meio MS associada à presença de carvão ativado induziu maior crescimento da raiz principal (8,50 cm) quando comparado com meio MS, na presença (6,19 cm) ou ausência (6,00 cm) de carvão ativado. ABSTRACTThe objective of this study was to determine the most favorable conditions for the in vitro germination of mangaba seeds and initial development of plantlets. After asepsis, emerging seeds of mature fruits were inoculated in tubes contend the next treatments: T1-15 mL of MS culture medium; T2-15 mL of MS culture medium + 2.0 g L-1 of activated charcoal; T3-15 mL of ½ MS culture medium; and T4-15 mL of ½ MS culture medium + 2.0 g L-1 of activated charcoal. All the culture medium were gellified with 0.3 g L-1 of Phytagel® and supplemented with 3.0 g L-1 of sucrose. The statistical design was completely randomized with four treatments, eight repetitions and ten seeds by experimental unit. There was not significant difference of the treatments for the germination percentage at twenty days, which varied from 95 to 100%. The ½ MS with 2.0 g L-1 of activated charcoal promoted higher growth the main root when compared with the others treatments. After 50 days, abnormal plantlets were not observed and neither significant difference were verified among the length of the aerial part. However, the dilution in 50% of the MS culture medium associate to the presence of activated charcoal induced higher growth of the main root (8.50 cm) when compared with MS culture medium in the presence (6.19 cm) or absence (6.00 cm) of activated charcoal.
Resumo -O objetivo deste trabalho foi ajustar os protocolos de cultivo in vitro de embriões zigóticos e de aclimatação de plântulas de coqueiro-anão-verde do Brasil de Jiqui (Cocos nucifera L.). Os embriões zigóticos maduros, colocados assepticamente em meio de cultura líquido Y3 suplementado com 27,8 mg L de carvão ativado, apresentaram incremento do desenvolvimento radicular e da parte aérea. Na fase de aclimatação, o substrato composto por areia lavada e pó de casca de coco seco, na proporção de 1:1, proporcionou maior sobrevivência das plântulas (58,33%), maior crescimento da parte aérea (39,08 cm) e maior número de folhas (6,33). Os protocolos estabelecidos para o cultivo in vitro de embriões zigóticos e a aclimatação de coqueiro-anão-verde de Jiqui podem ser utilizados no intercâmbio e conservação de germoplasma.Termos para indexação: Cocos nucifera, germinação in vitro, meios de cultura, regulador de crescimento. In vitro culture of zygotic embryos and acclimatization of green dwarf coconut palmAbstract -The objective of this work was to improve the protocols for culture and acclimatization of green dwarf coconut palm (Cocos nucifera L.). Mature zygotic embryos, cultivated in vitro aseptically in Y3 liquid medium culture, supplemented with 27.8 mg L of activated charcoal, promoted better root and shoot development. In the acclimatization phase, the 1:1 substrate composed of washed sand and coconut coir dust provided higher survival of plantlets (58.33%), larger growth of the aerial part (39.08 cm) and larger number of leaves (6.33). The established protocols for in vitro culture of zygotic embryos and acclimatization of green dwarf coconut palm plantlets can be applied to germplasm exchange and conservation.
Resumo Segmentos nodais estiolados são utilizados para a micropropagação e conservação da identidade genética em várias culturas. O objetivo deste trabalho foi avaliar a multiplicação in vitro de segmentos estiolados para produção de mudas do abacaxi híbrido PE x SC-52. Talos de plântulas produzidos in vitro receberam inóculo em tubos de ensaio contendo o meio de cultura MS e mantidos no escuro por 60 dias, para estiolamento. Foram utilizados três tratamentos, sem reguladores de crescimento, ANA a 1,86 mg/L e AIA a 1,75 mg/L em cinco repetições com dez explantes por repetição. Não houve diferença no número de brotos estiolados por explante entre os tratamentos avaliados. No entanto, aos 35 dias de cultivo, os tratamentos com ANA e AIA apresentavam maior número de nós por broto, sendo o ANA superior aos demais após 60 dias. Brotos estiolados in vitro por 60 dias foram colocados horizontalmente em placas de Petri contendo o meio MS e incubados na presença de luz. Foram usados quatro tratamentos para a regeneração das plântulas, sem reguladores de crescimento; BAP a 2 mg/L; ANA a 1,86 mg/L + BAP a 2 mg/L e CIN a 5 mg/L, em quatro repetições, com sete brotos estiolados, por repetição. O BAP promoveu maior número de plântulas regeneradas por broto e por nó, com uma média de 10,4 plântulas por broto estiolado, aos 30 dias.Termos para indexação: Ananas comosus, cultura in vitro, micropropagação, propagação de plantas. Etiolation and regeneration in the in vitro multiplication of hybrid PE x SC-52 pineappleAbstract Etiolated nodal segments are used for micropropagation and conservation of the genetic identity of several crops. The present study evaluated this method for the multiplication of the pineapple hybrid PE x SC-52. Plantlet stems produced in vitro were inoculated in test tubes containing MS nutrient medium and maintained in darkness for 60 days for etiolation. Three treatments were used with growth-regulator free MS medium; NAA at 1.86 mg/L, and IAA at 1.75 mg/L, with five repetitions and ten explants per repetition. There was no difference among the treatments in the number of etiolated shoots, per explant. However, NAA and IAA, after 35 days, produced a greater number of nodes per shoot, and NAA was the most efficient after 60 days. Etiolated shoots were placed horizontally in Petri dishes containing MS medium and incubated in the presence of light. Four treatments were used for the regeneration of plantlet growth-regulator free MS medium; BAP at 2 mg/L; NAA at 1.86 mg/L + BAP at 2 mg/L, and KIN at 5 mg/L with four repetitions and seven etiolated shoots per repetition. BAP promoted the largest number of regenerated plantlets per shoot and per node, with an average of 10.4 plantlets per shoot after 30 days.

COMUNICAÇÃO CULTIVO INICIAL IN VITRO DE GEMAS AXILARES DE RESUMOA capacidade regenerativa de gemas axilares de abacaxizeiro cv. Pérola (Ananas comosus (L.) Merr.) foi avaliada em meio de cultivo líquido e gelificado, na presença e ausência de luz, com o objetivo de produzir material para subsequentes fases da micropropagação. Os explantes foram estabelecidos, por 60 dias, em meio MS acrescido de 0,63 M de ANA e 2,5 M de BAP. O delineamento experimental foi inteiramente casualizado, constituído com quatro tratamentos (meio líquido com e sem ponte de papel de filtro e meio gelificado com ágar a 6,0 g L -1 e com Phytagel ® a 3,0 g L -1 ), com quatro repetições e cinco explantes por repetição. Diferentes volumes de meio líquido (2,5 mL; 5,0 mL e 10,0 mL) também foram avaliados, usando-se quatro repetições por volume de meio de cultura. Maior número de brotos foi obtido na presença de luz, em meio líquido sem ponte de papel de filtro, onde 80% dos explantes se diferenciaram em brotos. Na ausência de luz, 68,8% a 87,5% dos explantes não regeneraram brotos. Explantes cultivados em volumes de 5,0 mL ou 2,5 mL de meio MS líquido, acrescido de 0,63 M de ANA e 2,5 M de BAP, na presença de luz, constitui um protocolo adequado para cultivo inicial de gemas axilares e regeneração de brotos de abacaxizeiro. Merr.) was evaluated in liquid and semi-solid medium, under light and dark conditions, with the objective of producing materials for subsequent phase of micropropagation. The explants were established for 60 days in basal MS medium, supplemented with 0.63 M NAA and 2.5 M BAP. The statistical design was completely randomized with four treatments (liquid medium with and without filter paper bridge, and semi-solid medium gellified with agar and phytagel®), four repetitions and five explants per experimental unit. Different volumes of liquid medium (2.5 mL, 5.0 mL, and 10.0 mL) were also evaluated in four replications per treatment. More buds were obtained in liquid medium without filter paper bridge, where 80.0% of explants differentiated new shoots. In treatments cultivated in the dark, 68.8% to 87.5% of the explants did not regenerate new shoots. Explants cultivated in volumes of 2.5 mL and 5.0 mL of medium, in the presence of light, were more efficient in the induction and formation of shoots. The cultivation of axillary buds of lateral pineapple sprouts in 2.5 mL or 5.0 mL of MS liquid medium, supplemented with 0.63 M NAA and 2.5 M BAP, in the presence of light, represents an adequate protocol for establishing new pineapple shoots for use in subsequent phases of micropropagation.
