We report for the first time the efficient use of accelerated solvent extraction (ASE) for extraction of ricin to analytical purposes, followed by the combined use of sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE), Matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS), and MALDI-TOF MS/MS method. That has provided a fast and unambiguous method of ricin identification for in real cases of forensic investigation of suspected samples. Additionally, MALDI-TOF MS was applied to characterize the presence and the toxic activity of ricin in irradiated samples. Samples containing ricin were subjected to ASE, irradiated with different dosages of gamma radiation, and analyzed by MALDI-TOF MS/MS for verification of the intact protein signal. For identification purposes, samples were previously subjected to SDS-PAGE, for purification and separation of the chains, followed by digestion with trypsin, and analysis by MALDI-TOF MS/MS. The results were confirmed by verification of the amino acid sequences of some selected peptides by MALDI-TOF MS/MS. The samples residual toxic activity was evaluated through incubation with a DNA substrate, to simulate the attack by ricin, followed by MALDI-TOF MS/MS analyses.
Gas chromatography simultaneously coupled to flame photometric detector, nitrogen phosphorus detector and mass spectrometer (GC/FPD/NPD-MS) was used to identify nitrogen and sulfur mustards, their precursors and degradation products in samples of water, soil, organic solvent and a PVC floor. Extraction techniques and derivatization with BSTFA were employed during sample preparation. Fast screening of the samples was possible due the high sensibility and selectivity of FPD and NPD for sulfur and nitrogen compounds. Mass spectra of the identified chemicals presented match factors equal or bigger than 90% when compared to OPCW databases.Keywords: Mustard gas; gas chromatography; mass spectrometry; chemical weapons. ResumoCromatografia gasosa acoplada simultaneamente a detectores fotométricos de chama, de nitrogênio e fósforo e espectrômetro de massas (CG/DFC/DNP-EM) foi utilizada na identificação de mostardas de enxofre e de nitrogênio, seus precursores e produtos de degradação em amostras de água, solo, solvente orgânico e um piso de PVC preparadas de forma controlada. Técnicas de extração e derivatização com BSTFA foram utilizadas para possibilitar as análises das substâncias por cromatografia gasosa. O uso simultâneo de vários detectores em um só equipamento possibilitou rapidez nas análises. DFC e DNF apresentam boa seletividade e sensibilidade para compostos contendo enxofre e nitrogênio, permitindo a seleção inicial dos picos cromatográficos de possíveis compostos de interesse. Todos os espectros de massa das substâncias identificadas apresentaram índices de similaridades iguais ou superiores a 90% quando comparados aos existentes nas bases de dados da OPAQ.Palavras-chave: Gás mostarda; cromatografia gasosa; espectrometria de massas; armas químicas.
This article presents a brief review of the literature on ricin. This toxin is considered as a case of vulnerability in Brazilian responsibilities related to The Chemical Weapons Convention (CWC). Ricin is a glycoprotein present in seeds of castor beans (Ricinus communis L). It is very lethal and considered a chemical weapon. Brazil is the third world place in production of castor beans. Tons of residues with high quantities of ricin are produced every year. In this paper, examples of the ricin in terrorist attacks are remembered. It also mentions aspects on the action mechanism in the human body. The importance of the fast detection and identification is emphasized with description of methods used by first emergency responders and specialized chemical laboratories. Ricin detoxification has military interest and also may aggregate value to castor beans residues. ResumoEste artigo apresenta uma mini-revisão da literatura sobre a ricina, abordando a vulnerabilidade que essa toxina representa para o Brasil em relação à Convenção para Proibição de Armas Químicas (CPAQ). A ricina é uma glicoproteína presente na mamoneira (Ricinus communis L.), planta endêmica no território brasileiro. Devido a sua elevada toxicidade é considerada uma arma química. O Brasil é o terceiro maior produtor mundial de mamona, gerando todos os anos toneladas de resíduos contendo alto teor de ricina. Esse trabalho aborda aspectos do mecanismo de ação dessa substância no organismo além de citar casos da literatura internacional sobre o uso dessa substância em ações terroristas. A importância de métodos de detecção e de identificação rápidos e confiáveis é enfatizada com descrição de técnicas utilizadas tanto por equipes de resposta a emergências, quanto por laboratórios de química especializados. Os processos de destoxificação de ricina, além de possuírem importância militar, podem agregar valor econômico aos resíduos da agroindústria mamoneira. Por fim, é abordado o desenvolvimento de vacinas para imunização contra o envenenamento por ricina.Palavras-chave: Ricina; mamona; armas químicas; OPAQ.
This work aims to determine the metabolic profile of athletes, relating metabolites present in the human body to variables such as age, gender and physical effort to which they were submitted. Chromatographic and spectrometric methods (such as GC-MS and HPLC-MS) are used to separate, identify and quantify metabolites in serum and plasma samples. The obtained data are analyzed using multivariate statistics, using principal component analysis (PCA) and partial least squares regression for discriminant analysis (PLS-DA). In this way, the metabolites identified in the samples can be correlated with the studied variables, such as age, gender and physical effort. The results show the potential of metabolomics as an important tool to be used in the study of a variety of unknowns that may interfere in athlete's performance. ResumoEste trabalho procura determinar o perfil metabólico de atletas, relacionando metabólitos presentes no corpo humano a variáveis como idade, sexo e esforço físico a que foram submetidos. Métodos cromatográficos e espectrométricos (como CG-EM e CLAE-EM) são usados para separar, identificar e quantificar metabólitos em amostras de soro sanguíneo e plasma. Os dados obtidos são analisados por meio de estatística multivariada, usando análise de componentes principais (PCA) e mínimos quadrados parciais para análises de discriminantes (PLS-DA). Dessa forma, pode-se correlacionar os metabólitos identificados nas amostras às variáveis estudadas, como a idade, o sexo e o esforço físico dos atletas. Os resultados mostram o potencial da metabolômica como uma importante ferramenta a ser usada no estudo de uma variedade de incógnitas que podem interferir no desempenho dos atletas.
