A hybrid system based on lignocellulosic biomass gasification and syngas fermentation represents a second-generation biorefinery approach that is currently in the development phase. Lignocellulosic biomass can be gasified to produce syngas, which is a gas mixture consisting mainly of H2, CO, and CO2. The major challenge of biomass gasification is the syngas’s final quality. Consequently, the development of effective syngas clean-up technologies has gained increased interest in recent years. Furthermore, the bioconversion of syngas components has been intensively studied using acetogenic bacteria and their Wood–Ljungdahl pathway to produce, among others, acetate, ethanol, butyrate, butanol, caproate, hexanol, 2,3-butanediol, and lactate. Nowadays, syngas fermentation appears to be a promising alternative for producing commodity chemicals in comparison to fossil-based processes. Research studies on syngas fermentation have been focused on process design and optimization, investigating the medium composition, operating parameters, and bioreactor design. Moreover, metabolic engineering efforts have been made to develop genetically modified strains with improved production. In 2018, for the first time, a syngas fermentation pilot plant from biomass gasification was built by LanzaTech Inc. in cooperation with Aemetis, Inc. Future research will focus on coupling syngas fermentation with additional bioprocesses and/or on identifying new non-acetogenic microorganisms to produce high-value chemicals beyond acetate and ethanol.
Crude glycerol is the main by-product of the biodiesel manufacturing industry (10% w/w). Its use as a substrate in microbial fermentations is a concrete strategy to efficiently address its market surplus. In this study, the conversion of crude glycerol to citric acid, a key biochemical in the emerging bioeconomy, by a wild-type yeast Yarrowia lipolytica DSM 8218 was modelled using the Response Surface Methodology. The model relates C/N mass ratio and crude glycerol concentration to maximize the citric acid yield in flask scale using two different N sources, yeast extract and ammonium sulphate. Under the optimal conditions (yeast extract, C/N 141, glycerol 33 g/L), the conversion yield was 0.249 g/g. The optimal conditions were used for up-scaling a fed-batch fermentation in a 2 L bioreactor highlighting a metabolic shift from mannitol to citric acid when high stirring rates were applied (800 rpm). In these conditions, a morphic transition from pseudo-mycelial form to round-shaped yeast-like cells was observed too.
Microbial lipids are renewable platforms for several applications including biofuels, green chemicals, and nutraceuticals that can be produced from several residual carbon sources. Lignocellulosic biomasses are abundant raw materials for the production of second-generation sugars with conversion yields depending on the quality of the hydrolysates and the metabolic efficiency of the microorganisms. In the present work, wheat straw pre-treated by steam explosion and enzymatically hydrolysed was converted into microbial lipids by Lipomyces tetrasporus DSM 70314. The preliminary optimization of the enzymatic hydrolysis was performed at the bench scale through the response surface methodology (RSM). The fermentation medium and set-up were optimized in terms of the nitrogen (N) source and carbon-to-nitrogen (C/N) ratio yielding to the selection of soy flour as a N source and C/N ratio of 160. The bench scale settings were scaled-up and further optimized at the 10 L-scale and finally at the 50 L pilot scale bioreactor. Process optimization also included oxygen supply strategies. Under optimized conditions, a lipid concentration of 14.8 gL−1 was achieved corresponding to a 23.1% w/w lipid yield and 67.4% w/w lipid cell content. Oleic acid was the most abundant fatty acid with a percentage of 57%. The overall process mass balance was assessed for the production of biodiesel from wheat straw.
The competition for freshwater between agricultural, industrial, and civil uses has greatly increased in Mediterranean basin characterized by prolonged dry seasons. The aim of this study was to evaluate biomass production and the potential ethanol production of promising “no-food” herbaceous crops irrigated with low quality water at different ETc restitutions (0%, 50 and 100%). The research was carried out, in 2011 and 2012, in an open field near the full-scale constructed wetland (CW) municipal treatment plant located in the Eastern Sicily (Italy). The CW effluent has been applied in a experimental irrigation field of Vetiveria zizanoides (L.) Nash, Miscanthus x giganteus Greef et Deu. and Arundo donax (L.). Physical, chemical and microbiological analyses were carried out on wastewater samples collected at inlet and outlet of CW and pollutant removal efficiencies were calculated for each parameter. Bio-agronomical analysis on herbaceous species were made with the goal to evaluate the main parameters such as the plant dimension, the growth response and the biomass production. Biomass dry samples were processed with a three-step chemical pretreatment, hydrolysed with a mix of commercial enzymes and next fermented to obtain the yield of ethanol production. Average TSS, COD and TN removal for CW were about 74%, 67% and 68%, respectively. Although the satisfactory Escherichia coli removal, about 3.5 log unit for both beds on average, CW didn’t achieve the restrictive Italian law limits for wastewater reuse. As expected, irrigation was beneficial and the full ET replenishment increase the biomass productivity as compared to the other two treatment. The mean productivity of Vetiveria zizanoides and Myscanthus x giganteus were about 9, 26 and 38 t ha–1 and 3, 7 and 12 t ha–1 respectively in 0%, 50% and 100% ETc restitutions. Arundo donax gave higher values of dry biomass (78 t ha–1 in 100% ETc restitution in 2011 season), and potential ethanol production (about 3,744 kg ha–1). These results suggest the interest in the use of constructed wetland effluents for the irrigation of energy crops to obtain second generation ethanol, particularly in semiarid regions such as the Mediterranean area.
