An experiment was conducted to investigate the effect of moisture stress on morpho-physiological parameters, seed yield and total oil content of thirteen fenugreek genotypes grown under moisture stress at different growth stages. Fenugreek genotypes showed significant differences in plant fresh weight, shoot and root weight, shoot and root length, number of branches, number of pods and seed yield per plant. Chlorophyll content and water potential was found to be reduced under water stress. Genotypes showed variation in canopy temperature under non stress conditions which was narrowed under stress conditions. Water stress at flowering and post flowering stage increased the oil content from a minimum of 3.29% in AFg 6 to a maximum of 5.31 in AM 327-3.

Fenugreek (Trigonella foenum-graecum) is receiving global attention due to rare medicinal properties of significance to human health. Gene banks possess scanty germplasm and very little background information regarding its genetic variability that has hampered its improvement. We investigated the extent of variability among 17 Indian varieties of fenugreek using phenotypic and genetic markers. Multilocus genotyping by ten random amplified polymorphic DNA (RAPD) primers detected an average of intraspecific variations amounting to 64.7% polymorphism in banding patterns. Analysis of molecular variance indicated that a greater proportion of total genetic variation exists within population (91%) rather than among populations. Higher values of Nei's gene diversity (h) and Shannon Information Index (i) and genetic distance analysis validate higher genetic diversity among Indian fenugreek varieties. SNPs at 14 sites of rDNA region revealed further lineages of distinct varieties with main RAPD clusters. The representative sequences of each subgroup and all distinct varieties have been submitted to NCBI database and assigned Gen Accession numbers HM 176640-176649. The measures of relative genetic distances among varieties of fenugreek did not completely correlate with the geographical distances of places of their development. The homogeneous phenotypic markers proved insufficient in exhibiting genetic divergence among fenugreek varieties studied. Eventually, the knowledge of their genetic relationships, DNA bar coding and phylogenies might contribute for the designing of intraspecific crosses between cultivars of this fenugreek collection with potential interest in seed spices breeding programme.

An analysis has been carried out to explore the genetic variation in seventeen selected varieties of fenugreek. Sixteen fatty acids were identified in seed oil. Linoleic acid (18:2; n-6) and -Linolenic acid (18:3; n-3) were the major contributor found in the range of 26-43% in variety Lam Selection. The MUFA content varied from 3.50% in Azad Methi to 19.31 % in Hisar Madhvi. The ratio of n-6 to n-3 fatty acids indicated that fenugreek seed lipids are a good source of PUFA. PCA identified linolenic and oleic acid as the most important traits responsible for variation in presented material and improving quality through breeding. Estimates of genetic distance values showed wide range of variation among the fenugreek genotypes. The Euclidean based UPGMA clustering revealed three real and four singleton clusters. Genetic diversity showed no relationship to geographical origin. Dissimilarities obtained showed a wide variation in oil content and in composition.
