Insects are the most speciose group of animals, but the phylogenetic relationships of many major lineages remain unresolved. We inferred the phylogeny of insects from 1478 protein-coding genes. Phylogenomic analyses of nucleotide and amino acid sequences, with site-specific nucleotide or domain-specific amino acid substitution models, produced statistically robust and congruent results resolving previously controversial phylogenetic relations hips. We dated the origin of insects to the Early Ordovician [~479 million years ago (Ma)], of insect flight to the Early Devonian (~406 Ma), of major extant lineages to the Mississippian (~345 Ma), and the major diversification of holometabolous insects to the Early Cretaceous. Our phylogenomic study provides a comprehensive reliable scaffold for future comparative analyses of evolutionary innovations among insects.
BackgroundNext-generation-sequencing (NGS) technologies combined with a classic DNA barcoding approach have enabled fast and credible measurement for biodiversity of mixed environmental samples. However, the PCR amplification involved in nearly all existing NGS protocols inevitably introduces taxonomic biases. In the present study, we developed new Illumina pipelines without PCR amplifications to analyze terrestrial arthropod communities.ResultsMitochondrial enrichment directly followed by Illumina shotgun sequencing, at an ultra-high sequence volume, enabled the recovery of Cytochrome c Oxidase subunit 1 (COI) barcode sequences, which allowed for the estimation of species composition at high fidelity for a terrestrial insect community. With 15.5 Gbp Illumina data, approximately 97% and 92% were detected out of the 37 input Operational Taxonomic Units (OTUs), whether the reference barcode library was used or not, respectively, while only 1 novel OTU was found for the latter. Additionally, relatively strong correlation between the sequencing volume and the total biomass was observed for species from the bulk sample, suggesting a potential solution to reveal relative abundance.ConclusionsThe ability of the new Illumina PCR-free pipeline for DNA metabarcoding to detect small arthropod specimens and its tendency to avoid most, if not all, false positives suggests its great potential in biodiversity-related surveillance, such as in biomonitoring programs. However, further improvement for mitochondrial enrichment is likely needed for the application of the new pipeline in analyzing arthropod communities at higher diversity.
This study investigates the influence of investment in key intangible resources and capabilities of firms, including intellectual capital (IC), financial capability (FC), and corporate social responsibility (CSR) on sustainable competitive advantage (SCA) and firm performance (FP) in an emerging country, Pakistan. The research model and its pertinent hypotheses are tested on a sample of 329 Pakistani small and medium enterprises (SMEs) by a structural equation model (SEM). The results exhibited a significant influence of the aforesaid factors on FP. However, SCA fully mediates the relationship between FC and FP and between CSR and FP, whereas SCA partially mediates the relationship between IC and FP. Taken together, the findings suggest the role of CSR and FC in indirectly spurring FP through SCA. Despite the limitations of this study, the results have several practical and theoretical implications for owners, managers, and policymakers.
We performed a genome-wide association study with 706 individuals with leprosy and 5,581 control individuals and replicated the top 24 SNPs in three independent replication samples, including a total of 3,301 individuals with leprosy and 5,299 control individuals from China. Two loci not previously associated with the disease were identified with genome-wide significance: rs2275606 (combined P = 3.94 × 10(-14), OR = 1.30) on 6q24.3 and rs3762318 (combined P = 3.27 × 10(-11), OR = 0.69) on 1p31.3. These associations implicate IL23R and RAB32 as new susceptibility genes for leprosy. Furthermore, we identified evidence of interaction between the NOD2 and RIPK2 loci, which is consistent with the biological association of the proteins encoded by these genes (NOD2-RIPK2 complex) in activating the NF-κB pathway as a part of the host defense response to infection. Our findings have expanded the biological functions of IL23R by uncovering its involvement in infectious disease susceptibility and suggest a potential involvement of autophagocytosis in leprosy pathogenesis. The IL23R association supports previous observations of the marked overlap of susceptibility genes for leprosy and Crohn's disease, implying common pathogenesis mechanisms.

Dermatologists should screen for PsA in their patients, especially those with risk characteristics and early signs.
It has been shown that acquired stimulus-response bindings result from at least two types of associations from the stimulus to the task (stimulus-task or stimulus-classification; S-C) and from the stimulus to the motor response (stimulus-response or stimulus-action; S-A). These types of associations have been shown to independently affect behaviour. This finding suggests that they are processed in different pathways or different parts of a pathway at the neural level. Here we test a hypothesis that such associations may be differentially affected by repetition learning and that such effects may be detected by measuring their durability against overwriting. We show that both S-C and S-A associations are in fact strengthened when learning is boosted by increasing repetitions of the primes. However, the results further suggest that associations between stimuli and actions have less durable effects on behaviour and that the durability of S-C and S-A associations is independent of repetition learning. This is an important finding for the understanding of the underlying mechanisms of associative learning and particularly raises the question of which processes may affect flexibility of learning.
The effect of genetic factors is strong in AA, but environmental factors such as infection and psychological stress may still play an important role. Our findings on the genetics of AA are consistent with a polygenic additive mode of inheritance.
