Background:
In saliva, neutrophil constitutes the most prominent first-line defense of immune cells against pathogenic microbes. The importance of neutrophils to the host immune systems of neutropenic or patients disabled with regard to their neutrophil function results in a tendency toward serious infections, such as early childhood caries (ECC). The cytoplasmic granules present in neutrophils play a major role in neutrophil-mediated inflammation. Azurophilic granules contain antimicrobial proteins, such as defensin, a human antimicrobial peptide (HNP 1–3). The aim of this study is to analyze the correlation of HNP 1–3 secretion with CD63 expression on the surface of salivary neutrophils.
Materials and Methods:
This study constituted a cross-sectional, analytical observational study. Saliva taken from preschoolchildren between the ages of 4–6 years who had been divided into two groups, i.e., early childhood caries group with decayed, extracted, filled teeth (def-t) index >6 and caries free with def-t = 0, was subjected to a HNP 1–3 secretion test using ELISA assay and an expression test for CD63 by means of a flow cytometry test. Results:
The secretion of HNP 1–3 in the saliva of ECC was higher (172.6 ± 41.64) compared to that of caries-free cases (140.39 ± 31.91), whereas the level of CD63 salivary expression in ECC was lower (2.32 ± 0.57) than in the presence of caries (2.67 ± 0.46).
Conclusion:
In ECC cases, saliva increases HNP 1–3 secretion but decreases CD63 expression on the surface of salivary neutrophils.
Background:Pomegranates fruit extracts have several activities, among others, anti-inflammatory, antibacterial, and antioxidants that have the main content punicalagin and ellagic acid. Pomegranate has the ability of various therapies through different mechanisms. Vascular endothelial growth factor (VEGF) function was to form new blood vessels produced by various cells one of them was macrophages. Platelet-derived growth factor (PDGF) was a growth factor proven chemotactic, increased fibroblast proliferation and collagen matrix production. In addition, VEGF and PDGF synergize in their ability to vascularize tissues. The PDGF function was to stabilize and regulate maturation of new blood vessels. Activities of pomegranate fruit extract were observed by measuring the increased of VEGF and PDGF expression as a marker of wound healing process.Aim:To investigate the potential of pomegranate extracts on the tooth extraction wound to increase the expression of VEGF and PDGF on the 4th day of wound healing process.Materials and Methods:This study used 12 Cavia cobaya, which were divided into two groups, namely, the provision of 3% sodium carboxymethyl cellulose and pomegranate extract. The 12 C. cobaya would be executed on the 4th day, the lower jaw of experimental animals was taken, decalcified about 30 days. The expression of VEGF and PDGF was examined using immunohistochemical techniques. The differences of VEGF and PDGF expression were evaluated statistically using t-test.Results:Statistically analysis showed that there were significant differences between control and treatment groups (p<0.05).Conclusion:Pomegranate fruit extract administration increased VEGF and PDGF expression on post-tooth extraction wound.
Backgrounds: The use of mouthwash daily may influence the mechanical properties of bioactive composite resin as a dental restoration, one of the mechanical properties is surface hardness. Herbal mouthwash can be used to minimize the side effect of mouthwash that contain alcohol. Mauli banana stem and basil leaf extracts can be produced into herbal mouthwash. Methods: Thirty specimens (10mm diameter x 2mm thick; n=5/group) bioactive composite resins were immersed in a mixed solution of Mauli banana and basil leaf extracts with concentration of 25%, 50%, 75 %%, 100%, the control group of chlorhexidine gluconate 0.2% and aquadest for 7 days in 37oC. Measurement of the surface hardness was using the Vickers Microhardness Tester with 100 gf load for 15 seconds. Results: One Way ANOVA and Post Hoc Bonferroni exhibited the significant differences (p <0.05) in hardness values between the specimens that immersed in the mixed solution Mauli banana and Basil leaf extracts, which in the concentration of 100% (4,49 ± 4,61 VHN) compared with 25% (38,20 ± 2,58 VHN), 50% (41,40 ± 3,84 VHN) and 75% concentration (40,40 ± 3,55 VHN). There was no siginificant difference (p >0.05) between specimens immersed in CHX, aquadest and the mixed solution of mauli banana and basil leaf extracts in all concentration. Conclusion: There is no change in bioactive composite resin surface hardness after immersed in the mixed solution of Mauli Banana stem (Musa acuminata) and Basil leaf (Ocimum basilicum) extracts.Keywords: Basil leaf extract, Bioactive resin, Mauli Banana stem extract, Surface hardness
