Quercetin (QUE) and its water-soluble form сorvitin are medicinally important members of the flavonoid family and the most prominent dietary antioxidants. Numerous pharmacological effects of quercetin include protection against diseases, such as atherosclerosis, myocardial infarction, and cerebrovascular disea ses. Corvitin modulates liver blood flow but its effects on liver lipid metabolism have not been understood. We investigated the influence of corvitin (2.5, 5, 10 mg/kg) on the formation and secretion of cholesterol (Chol), cholesterol esters (EChol), phospholipids (PLs), free fatty acids (FFAs), and triglycerides (TGs) into the bile. Secreted bile was collected during 2.5 h of the experiment. Lipid fractions of the bile were separated by thin-layer chromatography. We defined that administration of corvitin caused a significant (P < 0.05-0.001) increase in levels of all studied lipid components of bile. At a dose of 2.5 and 5 mg/kg corvitin increased bile content of Chol, PLs and FFAs,and at a dose of 10 mg/kg had the largest effect on the total production of EChol and TGs. We concluded that corvitin activates the liver lipids metabolism and processes of bile formation.

Досліджено дію кверцетину (у вигляді модифікованої форми -корвітину) на об'ємну швидкість кровотоку (ОШК) у тканині підшлункової залози, печінки та слизової оболонки шлунка (СОШ) щурів з гострим панкреатитом (ГП), викликаним введенням L-аргініну. ОШК визначали методом кліренсу водню з електрохімічною його генерацією за допомогою полярографа LР-9 (Чехія ВСТУПГострий панкреатит (ГП) або запалення під-шлункової залози -важке захворювання з високим рівнем летальності, патогенез якого й досі досконало не вивчений, тому методів і засобів лікування наразі бракує. Однією з причин, яка не дає змоги більш повно до-слідити патофізіологічний механізм ГП, є складність одержання клінічних даних від пацієнтів на найбільш небезпечних ранніх стадіях хвороби -у перші години чи дні. ГП лишається серйозним захворюванням ще й через ускладнення, які його супроводжують, у першу чергу -це мультиорганні та мульти-системні порушення функцій [1]. Патологічні процеси в підшлунковій залозі викликані не лише активованими панкреатичними фер-ментами та посиленою продукцією медіаторів запалення [2], а й ішемією підшлункової за-лози. Оскільки тканина підшлункової залози не має компенсаторного резерву, вона є дуже чутливою до ішемічного стресу. Порушення кровотоку в залозі провокує інтерстиційні крововиливи, вазоконстрикцію, збільшення проникності судинної стінки і в'зкості крові, зростання взаємодії лейкоцитів із клітинами ендотелію, формування внутрішньосудин-них тромбів, і, як результат, набряк ткани-ни, деструкцію панкреатичної паренхіми і дрібних судин [3,4]. L-аргінінову модель ГП вважають найбільш придатною для до-слідження екстрапанкреатичних ушкоджень органів та їх патомеханізмів. Abstract.One of the synthetic analogues of antidiuretic hormone -desmopressin is used in patients with central diabetes insipidus and in those with coagulation disorders. However, its effects on bile secretion are not fully defined. We investigated the effect of desmopressin on bile formation and determined the role of V 1a vasopressin receptors in the action of desmopressin on choleresis. Rats were injected intraportally with a bolus of desmopressin; the changes of bile flow, the content of free and conjugated cholates were compared with control animal group. Selective antagonist of V 1a receptors was injected 10 minutes before desmopressin treatment and the findings were compared with the results after desmopressin injection alone. Desmopressin increased bile flow, secretion of total cholates like amino acids conjugated, while diminished free bile acids content. Secreted bile volume and conjugated bile acids content were reduced in V 1a receptors antagonist+desmopressin-treated rats. In contrast, free bile acids content was more than the results in desmopressin-treated rats. Desmopressin at concentrations nearly equal to physiological concentrations of natural hormone in blood shows its choleretic effect. Antagonist of V 1а vasopressin receptors modulates desmopressin action. This certifies the role of these receptors in the action of desmopressin on different processes of bile formation.

Water-soluble analogue of quercetin, corvitin is used in patients with myocardial infarction as blocker of 5-lipoxygenase. However, its effects on secretion, lipid content and physico-chemical properties of bile have not been understood yet. We investigated the effect of corvitin, applied in different doses, on the level of bile flow, the content of bile free and esterified cholesterol, phospholipids, triacylglycerols, and free fatty acids. In order to determine stability of the bile colloidal system, we examined the relationship between different lipid components. The rats were injected intraportally with a bolus of corvitin. At doses of 2.5, 5, and 10 mg/kg, the latter increased bile flow and concentration of total cholates, as well as free fatty acids. Corvitin (5 mg/kg) elevated phospholipids and cholesterol content, but at a dose of 10 mg/kg it increased the concentration of bile cholesterol esters and triacylglycerols. Corvitin applied at doses of 2.5 and 10 mg/kg increased total cholates/cholesterol ratio, but at a dose of 10 mg/kg, the drug reduced cholesterol / esterified cholesterol ratio. The results suggest that corvitin exerts choleretic effect and improves stability of bile colloidal system.
