An electrical method to measure extracellular bioelectrical activity in vitro is presented. This method exploits the Helmholtz capacitive double-layer established at the electrode surface. Small extracellular voltage variations in the order of μVs induce through the double-layer capacitor a displacement current that is measured. This current is then enhanced by a gain factor proportional to the electrode capacitance. In addition, when measurements are carried out at low frequencies in current mode the electrode contribution to the noise can be minimized. The performance of the electrodes and the method is demonstrated using zebrafish hearts and glioma cell cultures. We propose that this electrical method is an ideal tool to measure in vitro slow and temporally synchronized events that are often involved in long range intracellular signaling

An impedance based electronic tongue was developed and used to discriminate honey of different botanic origin. The e-tongue presented here is based on the small-signal frequency response of the electrical double-layer established between the honey solution and an array of four different sensing units composed by gold, carbon, indium-tin-oxide, and doped silicon. The ability of the e-tongue to discriminate honey of different floral origins was demonstrated by distinguishing honey from Bupleurum and Lavandula pollen prevalence. The honey fingerprint obtained with the e-tongue was validated by parallel melissopalenogical analysis and physico-chemical methods. It is demonstrated that the e-tongue is very sensitive to changes on the honey electrical conductivity. Small differences in electrical conductivity are introduced by the presence of ionisable organic acids and mineral salts. Moreover, we propose that the sensitivity of the tongue to changes in electrical conductivity can be explored to probe other complex liquid substances.

Ultra-sensitive electrodes for extracellular recordings were fabricated and electrically characterized. A signal detection limit defined by a noise level of 0.3–0.4 μV for a bandwidth of 12.5 Hz was achieved. To obtain this high sensitivity, large area (4 mm2) electrodes were used. The electrode surface is also micro-structured with an array of gold mushroom-like shapes to further enhance the active area. In comparison with a flat gold surface, the micro-structured surface increases the capacitance of the electrode/electrolyte interface by 54%. The electrode low impedance and low noise enable the detection of weak and low frequency quasi-periodic signals produced by astrocytes populations that thus far had remained inaccessible using conventional extracellular electrodes. Signals with 5 μV in amplitude and lasting for 5–10 s were measured, with a peak-to-peak signal-to-noise ratio of 16. The electrodes and the methodology developed here can be used as an ultrasensitive electrophysiological tool to reveal the synchronization dynamics of ultra-slow ionic signalling between non-electrogenic cells.

Astrocytes are neuroglial cells that exhibit functional electrical properties sensitive to neuronal activity and capable of modulating neurotransmission. Thus, electrophysiological recordings of astroglial activity are very attractive to study the dynamics of glial signaling. This contribution reports on the use of ultra-sensitive planar electrodes combined with low noise and low frequency amplifiers that enable the detection of extracellular signals produced by primary cultures of astrocytes isolated from mouse cerebral cortex. Recorded activity is characterized by spontaneous bursts comprised of discrete signals with pronounced changes on the signal rate and amplitude. Weak and sporadic signals become synchronized and evolve with time to higher amplitude signals with a quasi-periodic behavior, revealing a cooperative signaling process. The methodology presented herewith enables the study of ionic fluctuations of population of cells, complementing the single cells observation by calcium imaging as well as by patch-clamp techniques.

The transient electrical behavior of an electrode/electrolyte interface based on an electrode micro-structured with gold protruding mushroom shapes is measured and modeled. The application of a voltage pulse train with changing frequency generates a corresponding train of current pulses with an offset current that approaches a saturation limit exponentially. The transient behavior of the electrical current was accurately reproduced using the Nernst-Planck-Poisson model and the geometric parameters of the mushroom shape. Spatiotemporal patterns of the electric field demonstrate that the current transient is caused by a slow kinetics of charges accumulating at the gold/electrolyte interface and under the micro-mushroom cap. The accumulation of charges establishes an internal electrical potential that drives a Faradic current through the mushroom/electrolyte interface. The aim of this work is to understand how this charge dynamics in the vicinity of the three-dimensional electrode surfaces is established and how it can be minimized to prevent damage to cells and tissues under electrical stimulation.
