Introdução e contexto ético-profissionalA pesquisa cirúrgica em animais utilizados em laboratório tem se expandido nas últimas décadas, mormente em decorrência do melhor suporte anestésico, da sofisticação da infra-estrutura material para monitorização contínua peroperatória e de uma busca incessante por modelos que reproduzam condições mórbidas da espécie humana. Os focos principais destas pesquisas têm sido aprimorar o conhecimento acerca dos mecanismos fisiopatológicos de doenças, empreender ensaios terapêuticos com novos fármacos, estudar marcadores biológicos e avaliar novas técnicas com perspectivas de aplicabilidade na espécie humana. A diversidade de cenários que vinculam as necessidades humanas ao uso de animais fomentou, ao longo da história, reflexões éticas, bioéticas, filosóficas e religiosas direcionadas para pesquisa em animais vertebrados 1,2 . Aristóteles (384-322 a.C.), Galeno (131-201), Francis Bacon (1561-1626), William Harvey (1578-1657), Claude Bernard (1813-1878 representam alguns dos atores que se destacaram neste contexto.Todavia, somente por ocasião da segunda guerra mundial, em face das atrocidades cometidas nos campos de concentração, dentre as quais as cruéis experiências em anima nobile, ocorreu uma conscientização mundial para ações concretas capazes de preservar a vida humana 3 . RESUMO -Avaliaram-se os efeitos da utilização de ciclos alternados de restrição alimentar e realimentação no crescimento do pacu, durante o período de engorda, e a viabilidade desta prática na produção comercial da espécie. Juvenis foram distribuídos em três tanques e submetidos a diferentes manejos alimentares, constituindo os tratamentos: A (alimentado ad libitum), B (restrição alimentar de 4 semanas, realimentado por 9 semanas) e C (6 semanas de restrição alimentar, realimentado por 7 semanas), em um total de 13 semanas por ciclo (4 ciclos experimentais). No final de cada ciclo alimentar, 20 peixes de cada tratamento foram amostrados e os dados biométricos registrados. Os valores obtidos para peso, comprimento total e fator de condição (K) foram submetidos a ANOVA e as médias comparadas pelo teste de Duncan. Os resultados mostraram que o tratamento C é o mais indicado somente para outono/inverno, promovendo maior crescimento, menor custo com ração, baixa conversão alimentar e maior receita líquida parcial. Entretanto, durante as estações mais quentes do ano (primavera/verão) outros programas de alimentação devem ser testados, utilizando-se períodos mais curtos de restrição alimentar. Quando se tratou da produção anual, o tratamento A respondeu melhor em termos de biomassa produzida, apesar do maior gasto com alimentação, visto que, com o aumento da temperatura, o crescimento dos peixes dos demais tratamentos ficou prejudicado.Palavras-chave: avaliação econômica, ciclos de restrição alimentar e realimentação, crescimento, custos com alimentação Evaluation of the Growth and Feeding Costs of Pacu (Piaractus mesopotamicus) Submitted to Alternate Cycles of Feeding Restriction and RefeedingABSTRACT -The present study evaluated the effects of alternating food restriction and refeeding cycles on the growth of pacu, during the growout phase and the viability of this practice in the commercial production of the species. Juvenile fish were distributed in 3 tanks and submitted to different feeding strategies: group A (fed ad libitum daily), B (food restricted to 4 weeks and refed for 9 weeks) and C (food restricted to 6 weeks and refed for 7 weeks) totalizing 13 weeks per cycle (4 experimental cycles). At the end of each feeding cycle, 20 fish per treatment were sampled and the biometric data were registered. Values of weight, total length and condition factor (K) were submitted to ANOVA and the averages were compared by Duncan test. The results showed that the treatment C presented better results only in autumn/winter periods, promoting better growth, lower cost ration, lower food conversion and better partial net income. Otherwise, during the warmer periods of the year (spring/summer) other feeding strategies must be tested, utilizing shorter periods of food restriction. Regarding to the annual production, the treatment A showed higher biomass, although higher feeding costs. With the increase in the temperature, the growth of the fish from the other treatments was delayed.

BackgroundThis study evaluated the effectiveness of treating pseudarthrosis in rats by using bone marrow cell suspensions or cultures of bone marrow mesenchymal stromal cellsMethodsThirty-eight specific pathogen-free (SPF) animals were randomly assigned to four groups: Group 1, Control, without surgical intervention; Group 2 (Placebo), experimental model of femoral pseudarthrosis treated only with saline solution; Group 3, experimental model of femoral pseudarthrosis treated with heterologous bone marrow cells suspension; Group 4, experimental model of femoral pseudarthrosis treated with cultures of heterologous mesenchymal stromal cells from bone marrow. When pseudarthrosis was confirmed by simple radiological studies, digital radiography and histopathology after a 120-day postoperative period, Groups 2, 3 and 4 were treated as above. At 30, 60 and 90 days after the treatment, all animals were evaluated by simple radiological studies, and at the end of the experiment, the animals were assessed by computed axial tomography and anatomopathological and histomorphometric examinations.ResultsInjected cells were detected in the areas affected by pseudarthrosis using scintigraphy within the first 24 hours after their administration. After 60 days, the animals of Group 3 showed callus formation while the animals of Group 4 presented periosteal reaction and had some consolidated areas. In contrast, Group 2 showed a predominance of fibro-osteoid tissue. After 90 days, bone consolidation and remodeling was observed in all animals from Group 3 whereas animals from Group 4 exhibited partial consolidation and those ones from Group 2 persisted with pseudarthrosis.ConclusionThe treatment with heterologous bone marrow cells suspension proved to be effective in the treatment of pseudarthrosis whereas cultures of heterologous bone marrow mesenchymal stromal cells did not show the same potential to aid bone healing.

Purpose:The difficulty to anesthetize small laboratory animals with vaporizer prompted us to go in search of new materials, and create new techniques. The improved equipment of anesthesia we looked for should be low cost, practical, versatile, and its management serve ethical, teaching, and research purposes. Methods: The new components of the equipment were: the vaporizer, the unidirectional valve, the glass cylinder filled with water, the flow guidance y-shape tube, the flow regulators, the mask modifications, and another free airway for emergency occurrence. A test was done with 30 Wistar rats, Rattus norvegicus albinus, divided into three groups with 10 rats for each one. Groups 1, 2 and 3 were anesthetized with Ether, Halothane and Sevoflurane respectively, using the new gadget. The anesthetic induction time, the breathing rhythm alteration during an anesthesia pre-established time (10 minutes), and the recovery time were observed. Results: The equipment enabled an easy handling, and fulfilled a larger safeness and stability during the induction and anesthetic management. The test showed it was possible to make use of several anesthetic agents. Conclusion:The device is effective, and turns the anesthesia procedure into a very easy practice with low-cost. RESUMO Objetivos:As dificuldades evidenciadas no uso de vaporizadores para anestesia em animais de pequeno porte motivaram o aperfeiçoamento e a criação de novos componentes técnicos visando a construção de um aparelho de anestesia de baixo custo, prático e versátil no manuseio e que atenda aos preceitos éticos do ensino e da pesquisa. Métodos: Utilizaram-se 30 ratos da linhagem Wistar, Rattus norvegicus albinus, distribuídos em três grupos. Grupo 1,2 e 3 (n=10, cada), compreenderam o uso do novo aparelho aduzido, respectivamente, do Éter; Halotano e Sevoflurano. Foram verificados o tempo de indução anestésica, as alterações do ritmo ventilatório durante o tempo de manutenção anestésica (pré-estabelecido em 10 min) e o tempo de recuperação anestésica. O aparelho foi construído com dispositivos técnicos novos sobressaindo: o vaporizador, a válvula unidirecional na campânula, o cilindro de vidro com água, o tubo direcionador de fluxo em forma de Y, os reguladores de fluxo, as modificações na máscara e a entrada de ar, independente, para situações emergenciais. Resultados: O aparelho permitiu um fácil manuseio e propiciou uma maior segurança e estabilidade na indução e manutenção anestésicas possibilitando, inclusive, a utilização de vários agentes anestésicos. Conclusão: O aparelho demonstrou ser útil para anestesia em cirurgia experimental, facilitando a realização dos procedimentos de pesquisa e de ensino de forma mais prática, eficiente, segura, e com baixo custo. The experimental model is suitable for the development of a pseudoarthrosis in rats.

The experimental model of IRC is feasible, with animal's survival in middle term which allows the use of this interval like a therapeutic window for testing different approaches in order to repair the kidney damages.

Carbohydrate rich substrates such as lignocellulosic hydrolysates remain one of the primary sources of potentially renewable fuel and bulk chemicals. The pentose sugar d-xylose is often present in significant amounts along with hexoses. Saccharomyces cerevisiae can acquire the ability to metabolize d-xylose through expression of heterologous d-xylose isomerase (XI). This enzyme is notoriously difficult to express in S. cerevisiae and only fourteen XIs have been reported to be active so far. We cloned a new d-xylose isomerase derived from microorganisms in the gut of the wood-feeding beetle Odontotaenius disjunctus. Although somewhat homologous to the XI from Piromyces sp. E2, the new gene was identified as bacterial in origin and the host as a Parabacteroides sp. Expression of the new XI in S. cerevisiae resulted in faster aerobic growth than the XI from Piromyces on d-xylose media. The d-xylose isomerization rate conferred by the new XI was also 72% higher, while absolute xylitol production was identical in both strains. Interestingly, increasing concentrations of xylitol (up to 8 g L−1) appeared not to inhibit d-xylose consumption. The newly described XI displayed 2.6 times higher specific activity, 37% lower KM for d-xylose, and exhibited higher activity over a broader temperature range, retaining 51% of maximal activity at 30 °C compared with only 29% activity for the Piromyces XI.
