Hydrogel patches are some of the most effective dressings for wound healing. In this study, the Gantrez® S-97 (Gan)/xyloglucan (XG) hydrogel patches were formulated by using a full central composite design (CCD). The optimized hydrogel patches consisted of 17.78% w/w of Gan and 0.1% w/w of XG. Honey and D. bulbifera extract were loaded in the Gan/XG hydrogel patches. The physical properties of the hydrogel patches, including water content, water absorption, rate of water vapor transmission, and mechanical properties, were examined. The D. bulbifera extract/honey-loaded patch exhibited a higher value of water absorption, tensile strength, and elongation than the honey-loaded patch and the unloaded patch, respectively. The biological activities of the patches were also investigated. All hydrogel patches protected wounds from external bacterial infection. The D. bulbifera extract/honey-loaded patch exhibited stronger antioxidant activity than the honey-loaded patch and the unloaded patch. Besides, all the hydrogel patches with concentrations of 0.5–2.5 mg/mL showed that they were nontoxic to fibroblast cells. The combination of D. bulbifera extract and honey in the patch affected fibroblast proliferation. In addition, all Gan/XG hydrogel patches significantly induced recovery of the scratch area. Therefore, the Gan/XG hydrogel patches could be candidates as wound dressings.

The objective of this study was to optimize fabrication variables that affected desirable properties of dressings. Boesenbergia rotunda extract incorporated PVA hydrogels for wound dressings were fabricated by freeze-thaw method. The fabrication variables including PVA concentration (15, 17.5 and 20 % w/w), freeze-thaw cycle (2, 3 and 4 cycles) and extract loading (30, 40 and 50 % w/w) were studied and optimized. Effects of variables on the hydrogel wound dressing properties were determined by using Box-Behnken design and response surface method. Hydrogel properties such as tensile strength, elongation at break, Young’s Modulus, water content, swelling and erosion were measured and used as the designed responses. From statistical data analysis (p <0.05), the polynomial quadratic equation which indicated the significant effects of fabrication variables on the hydrogel properties was generated. In conclusion, desirable B. rotunda extract loaded PVA hydrogel dressing was favorably designed. The optimized PVA concentration, freeze-thaw cycle and extract loading were 17.07 % w/w, 3.86 cycles and 50 % w/w, respectively.

Objective: The objective of this study was to investigate the antimicrobial activity of Phellinus linteus against skin infectious pathogens, Staphylococcus epidermidis ATCC12228 and Propionibacterium acnes DMST 14916. Methods:Fungal fruiting bodies were extracted with 95% ethanol and ethyl acetate, and then, vaporized. The antimicrobial activities were determined by the disc diffusion method against Propionibacterium acnes DMST 14916 and Staphylococcus epidermidis ATCC12228 skin infectious pathogens. A minimum inhibitory concentration (MIC) and a minimum bactericidal concentration (MBC) for those crude extracts were determined. Finally, the chemical profile of crude extract was determined by using thin layer chromatography and GC-MS. Results:The result demonstrated that the ethanolic extraction had more active fractions with an MIC of 0.5 mg/ml against the growth of Propionibacterium acnes DMST 14916 and Staphylococcus epidermidis ATCC12228 and also showed a minimum inhibitory concentration (MBC) at a concentration of 1.0 mg/ml, while ethyl acetate-based solvents failed to develop on TLC according to Retention factor (Rf) values of 0.71-0.76. The GC-MS was applied to investigate the chemical profile of crude extract of Phellinus linteus, revealing a component of hexadecanoic acid and 9, 12-octadecadienoic acid. Conclusion:Phellinus linteus fruiting body extracts have great potential as antimicrobial compounds against Propionibacterium acnes DMST 14916 and Staphylococcus epidermidis ATCC12228. Thus, they can be used in the treatment of infectious diseases caused by bacterial pathogens.

Jasminum sambac (J. sambac) has been used as a medicinal herb with wound healing property. This experimental research proposed to evaluate the effect of J. sambac flower extracts on fibroblast cell viability and film characteristics which maybe benefit in development of wound dressing film. The testing extract of J. sambac flowers were prepared as filtration extract (F), compression of the residue flower extract (C), combination of filtration and compression extract (FC) and ethanolic extract (E). The testing extracts were evaluated the effect on fibroblast cell viability using resazurin reduction assay. NIH 3T3 fibroblast cells were treated with variable concentration of J. sambac extracts for 24, 48 and 72 h. prior to evaluate cell viability. The results demonstrated that the F and FC extract of J. sambac flower at concentration of 31.25-500 µg/ml did not induce cytotoxicity on NIH 3T3 fibroblast cells given non-cytotoxic effect. The cell viability seemed to increase at 48 h of treatment with concentration of 31.25-250 µg/ml. Meanwhile, C and E extract induced cytotoxicity on NIH 3T3 cells by reduction of cell viability. The F and FC non-cytotoxic extracts were selected to examine ability of film formation without film-based compounds and assessed physical and mechanical characteristics of the prepared films. The experimental results revealed F and FC extracts could form film after incubation at 60°C. The better physical and mechanical characteristics of the films was presented in FC film which the prepared film revealed smooth surface and better absorption and elongation. Furthermore, gelatin was blended to FC extract for assess improvement of film properties. The results revealed that gelatin-blended FC extract film gave better mechanical characteristics of the film. Non-cytotoxicity of the FC extract of J. sambac flowers on fibroblast cells and appropriate physical and mechanical characteristics of FC film may point to the usefulness of J. sambac flower extracts in herb-based film for wound dressing.
