Ammonia in gastric juice is considered a potential biomarker for Helicobacter pylori infection and as a factor contributing to gastric mucosal injury. High ammonia concentrations are also found in patients with chronic renal failure, peptic ulcer disease, and chronic gastritis. Rapid and specific methods for ammonia detection are urgently required by the medical community. Here we present a method to detect ammonia directly in gastric juice based on Fourier transform infrared spectroscopy. The ammonia dissolved in biological liquid samples as ammonium ion was released in air as a gas by the shifting of the pH equilibrium of the ammonium/ammonia reaction and was detected in line by a Fourier transform infrared spectroscopy system equipped with a gas cell for the quantification. The method developed provided high sensitivity and selectivity in ammonia detection both in pure standard solutions and in a simulated gastric juice matrix over the range of diagnostic concentrations tested. Preliminary analyses were also performed on real gastric juice samples from patients with gastric mucosal injury and with symptoms of H. pylori infection, and the results were in agreement with the clinicopathology information. The whole analysis, performed in less than 10 min, can be directly applied on the sample without extraction procedures and it ensures high specificity of detection because of the ammonia fingerprint absorption bands in the infrared spectrum. This method could be easily used with endoscopy instrumentation to provide information in real time and would enable the endoscopist to improve and integrate gastroscopic examinations.
Mikro-Bestimmung* der Oxalsäure im Harn und Blut. Mit 2 Figuren Im Text. Im Verfolg biologischer Überlegungen sind in den letzten Jahren zahlreiche Verfahren zur Bestimmung von Oxalsäure im Harn und Blut angegeben worden. Ilda Schindler 1 ) hat auf der Suche nach einer geeigneten Methode das Schrifttum über die Biochemie der Oxalsäure gesammelt und einige Verfahren nachgearbeitet. Paul B. Müller 2 ) würdigt kritisch die wichtigsten Arbeiten über Bestimmung und Biologie der Oxalsäure bei Mensch und Tier. Für alles Nähere verweisen wir auf diese Arbeiten. Die quantitative Erfassung der Oxalsäure im Harn und Blut ist deshalb so schwierig, weil kleine Mengen Säure neben vielen Begleitstoffen zu isolieren sind. Fast alle Bearbeiter benützen die Fällbarkeit des Calciumoxalates. Über das Vorgehen dabei ist man sich noch nicht einig. Von den zahlreichen Vorschlägen sagte uns das Verfahren von Dodds und Gallimore 3 ) besonders zu. Wir wollten diese Vorschrift für noch kleinere Mengen Harn und Blut verwenden. Beim Nacharbeiten stellten wir fest, daß für die Gewinnung eindeutiger Ergebnisse eine ganze Reihe von Bedingungen einzuhalten sind. Schließlich haben wir die Vorschrift von Dodds und Gallimore verlassen und glauben ein neues, nach jeder Sichtung gesichertes Mikroverfahren ausgearbeitet zu haben. Wir geben die Vorarbeiten (277 Versuche) hier in gedrängter Kürze, schildern aber das neue Verfahren ausführlich. Harn, Serum oder Gesamtblut werden enteiweißt. Aus dem Filtrat wird die Oxalsäure im neuen Mikroextraktionsapparat mit Äther ausgezogen, nach dem Abdampfen des Äthers als Kalksalz gefällt uud abzentrifugiert. Aus dem Fällungsgemisch wird die Oxalsäure in einer besonderen Apparatur verestert, der Äthylester abdestilliert, verseift und die Säure neuerdings als Kalksalz gefällt Nach kurzer Behandlung mit W*asserdampf in schwefelsaurer Lösung wird die Oxalsäure mit Permanganat titriert. B. Grundlagen zur Oxalsäure-Bestimmung. a) Die einzelnen Versuehsphasen.
-Nach Erkalten fielen 0,l g Di-pyrazol des Succinyl-di-(acetessigesters) (VII) als feines Krystallpulver aus. Each Waschen mit wenig eiskaltem Allrohol zeigten sie den richtigen Smp. 155-157O. Das Praparat war identisch mit dem oben beschriebenen Produkt.Das alkoholische Filtrat wurde mit etwas Wasser ausgespritzt und das emulgierte 61 durch Erwiirmen in Losung gebracht. Nach Stehen uber Nacht krystallisierten 0,3 g Pyrazol-pyrazolon des Succinyl-malonester-acetessigesters (IX), das nach dreimaligem Umlosen aus wassrigem Alkohol fast farblose Xadelchen vom Smp. 114-115'' bildete. Die Substanz gibt eine violette Eisen(II1)-chlorid-Reaktion. 3,938 mg Subst. gaben 9,565 mg CO, und 1,980 mg H,O 2,292 mg Subst. gaben 0,235 om3 N, (18O, 746 mm) C,,H,O& Ber. C 66,38 H 5,78 N 11,47;/, Gef. ,, 66,28 ,, 5,63 ,, 11,800/, Eine Anreicherung des Succinyl-di-(acetessigesters) konnte auch durch fraktionierte Fallung mit Kupferacetat erreicht werden. Eine Losung von 5 g des oben beschriebenen 0 1 9 (Gemisch) in 5 om3 Alkohol wurde tropfenweise unter Reiben mit konz. wassriger Kupferacetatlosung versetzt, bis sich die schon blaue Kupferverbindung abzuscheiden begarm. Dann gab man weitere 2 cm3 Kupferacetat zu, saugte ab und wnsch aus. Durch Zerlegung erhielt man 1,3 g gelbliches 01, das mit Phenylhydrazin in Essigsaure zu einer gleichfalls oligen Verbindung umgesetzt wurde. Diese wurde mit 5 cm3 konz. Sodalosung unter Erwarmen verrieben, die uberstehende Losung abdekantiert und mit Wasser nachgespiilt. Beim Erkalten wurde das 01 fest. Each Losen in 8 cm3 warmem Alkohol krystallisierten 0,6 g farblose kleine Prismen vom Smp. l56O, identisch mit den1 Di-pyrazol des Succinyl-di-(acetessigesters) (VII). Die physiko-chemische Bestimmung des Vitamin D erfolgt heute fast ausschliesslich spektrometrisch durch Messung der Lichtabsorption des Vitamin D im U. V. bei 2650 A oder kolorimetrisch unter Zugrundelegung der Caw-Price-Reaktion. I n Naturprodukten wird die spektrometrische Bestimmungl-s) des Vitamin D im U. V. durch zahlreiche Begleitstoffe gestort, so dass diese Methode nur fur ganz reine Praparate in Frage kommt. Auch die kolorimetrische Bestimmungsmethode mit SbC13g-11) ist fur allgemein analytische Zwecke noch zu wenig spezifisch.Das gleiche gilt auch fur andere Farbreaktionen, -\vie z.
