The ornamental flower sector has growing over the past years worldwide with potential for further expansion. Among the ornamental plants, Passiflora species have been gaining ground in the market, mainly in European and North American countries. However, the market aiming the use of these species in ornamentation is still poorly explored. The inclusion of passion flower in the list of ornamental plants is related to the peculiar characteristics of the flower as it is complex structure, capacity of flowering all year long and also by the abundance and exuberance of the leaves, which in many species adds an ornamental value. Among the biotechnological tools for the production of ornamental plants, tissue culture has been outstanding in the cloning of elite genotypes, with high phytosanitary quality and large scale production. In addition, it offers possibilities of producing new varieties with characteristics peculiar to the market of ornamental plants. The diversity of wild Passiflora opens perspectives to the conservation, market and production of ornamental Passiflora cultivars.
RegeneRação in vitRo deRESUMO Passiflora miniata é uma espécie silvestre nativa da Amazônica Meridional, com potencial ornamental devido à beleza de suas flores de coloração vermelha intensa. Relatos na literatura sobre a espécie ainda são insipientes. O objetivo do presente estudo foi induzir a regeneração de novo pela via organogênica a partir de embriões zigóticos maduros de P. miniata. Os embriões zigóticos foram isolados e cultivados no meio de MS com a adição dos reguladores de crescimento 6-Benziladenine (BA), Thidiazuron (TDZ) e Cinetina (CIN). A regeneração de novo nos embriões zigóticos ocorreu de forma direta e/ou indireta. Um percentual de 80% dos explantes cultivados na presença de BA apresentaram organogênese pela via direta e 20% pela indireta, com TDZ 60% foram regenerados pela via direta e 40% pela indireta. Já nos tratamentos com CIN 58% do explantes apresentaram regeneração por organogênese direta e 42% pela indireta. O desenvolvimento dos primórdios das brotações iniciou-se com a formação das estruturas organogênicas que posteriormente se diferenciaram em multibrotações. As maiores médias no número de brotos produzidos foram na concentração de 0,75 mg L -1 de BA com 40,0 de brotos por explantes, sendo TDZ e CIN na concentração de 0,50 mg L -1 com média de 7,2 e 3,6, respectivamente. ABSTRACT In vitro regeneration of Passiflora miniata MastPassiflora miniata is a wild species native to the Southern Amazon, with ornamental potential due to the beauty of its flowers of intense red coloration. Reports in the literature about the species are still insipid. The aim of the present study was to induce the regeneration of P. miniata by the de novo organogenesis from mature zygotic embryos. The zygotic embryos were isolated and cultivated into the MS medium with the addition of 6-Benzyladenine (BA), Thidiazuron (TDZ) and Kinetin (KIN) growth regulators. The de novo regeneration from the zygotic embryos occurred directly and indirectly. A percentage of 80% of the explants cultivated in the presence of BA had direct organogenesis and 20% by the indirect way, with TDZ 60% were regenerated by the direct and 40% by the indirect way. Regarding the treatments with KIN, 58% of the explants had regeneration by direct and 42% by the indirect organogenesis. The development of shoot primordia initiated with the formation of organogenic structures that later differentiated into multi-shoots. The highest mean number of shoots (40.0 shoots per explants) was obtained on 0.75 mg L -1 BA. Conversely, using 0.50 mg L -1 TDZ or KIN, the highest number of shoots were 7.2 and 3.6, respectively.
RESUMOComo a grande maioria das plantas cultivadas, a cultura do sorgo está sujeita a diferentes variações ambientais. Assim, a resposta fotoperiódica típica de dia curto e de altas taxas fotossintéticas, está relacionada a um bom crescimento, desenvolvimento, e germinação do pólen. O trabalho teve por objetivo estimar a viabilidade polínica, sob diferentes tipos de corantes e a caracterização do grão de pólen, quanto a sua viabilidade e tamanho. Foram analisadas 250 grãos de pólen/lâmina, com 30 repetições, em microscópio óptico modelo Primo Star Carl Zeiss., tendo imagens capturadas por meio de um microscópio fotômico binocular (Leica ICC 50) acoplado a um computador e no software LAZ EZ V1. 7.0, para caracterização quanto a sua morfologia de diâmetro equatorial (DE) e diâmetro polar (DP), pelo software ImageJ. As médias da viabilidade polínica foram submetidas à análise de variância (ANOVA), no software SISVAR®, pelo teste Tukey à 5% de probabilidade. As análises de grãos de pólen do híbrido BRS 330, utilizando três diferentes corantes para estimar a viabilidade polínica, mostrou uma alta taxa de pólen viável, acima de 94%, sendo que a orceína acética apresentou o maior percentual de pólens viáveis (96.24%), seguido do Alexander (94.68%) e por último o Lugol (94.04%). A média para a relação diâmetro equatorial por diâmetro polar foi maior para os polens viáveis com valor de 0.94, determinando assim a classificação do pólen em relação ao seu formato, sendo este oblato-esferoidal, já os grãos de pólen inviáveis apresentaram média de 0.80 denominado assim de suboblato.
