ABSTRACTγ -oryzanol, a phytochemical, has antioxidant activities and potential health benefits. In this study, we aimed to develop a method for extracting high amounts of γ -oryzanol from rice bran and for verifying its stability in different solvents. Of the 5 methods under investigation, the one that yielded the highest γ -oryzanol content was further improved by using a central composite rotational design (DCCR -2 4 , including 8 tests in axial conditions and 4 central points, totaling 28 trials). The parameters under study were temperature, time, volume, and the ratio of hexane:isopropanol. High γ -oryzanol content (13.98 mg . g -1 ) was obtained by using the following extraction conditions: 40° C, 40 min, and 75 mL hexane:isopropanol (1:3). The limits of detection and quantification of the method were 0.9 µg g -1 and 31 µg g -1 , respectively. The instrumental precision was 0.004%, the repeatability (CV r ) was 9.4%, and the recovery was 111.7 ± 17.7%. γ -oryzanol is more stable in isopropanol than in hexane, in which 100% concentration could be maintained after refrigeration for 72 days.Index terms: Antioxidant, spectrophotometer, stability, phytochemistry. Esse estudo objetivou desenvolver um método para extração de alta quantidade de γ -orizanol de farelo de arroz e verificar sua estabilidade em diferentes solventes. Dentre os cinco métodos estudados, o que teve maior rendimento em γ -orizanol foi melhorado, usando um delineamento composto central rotacional (DCCR -2 4 , incluindo 8 experimentos no ponto axial e 4 no ponto central, totalizando 28 experimentos). As variáveis testadas foram temperatura, tempo, volume e proporção de hexano: isopropanol usados na extração. Elevado conteúdo de γ -orizanol (13,98 mg g -1 ) foi obtido quando as condições de extração foram 40° C, 40 min e 75 mL de hexano:isopropanol (1:3). This study aimed at evaluating the potential of the peroxidase (PO) enzyme to reduce ochratoxin A (OTA) levels and its application to grape juice. Both commercial PO and PO extracted from rice bran were evaluated, respectively, regarding their activity towards OTA in a model system. The affinity between PO and OTA was verified by the Michaelis-Menton constant and the maximum velocity parameters, resulting in 0.27 μM and 0.015 μM min −1 for the commercial enzyme, and 6.5 μM and 0.031 μM min −1 for PO extracted from rice bran, respectively. The lowest residual OTA levels occurred when 0.063 U mL −1 of the enzyme was applied. Under these conditions, the OTA reduction was 41% in 5 h for the commercial enzyme, and 59% in 24 h, for PO extracted from rice bran. When the extracted PO, with the activity of 0.063 U mL −1 , was applied to whole grape juice, the OTA levels decreased to 17%, at 24 h. The capacity shown by PO for reducing OTA levels was confirmed in whole white grape juice, as a model system. This study may assist the wine industry to offer healthier products and add value to rice bran.
RESUMOO trigo durante seu beneficiamento gera o farelo, um material lignocelulósico, que constitui aproximadamente 28% do grão. Este material representa uma valiosa fonte de hemicelulose (23,7%), lignina (3,6%) e celulose (11%), o que lhe confere a característica de possuir baixa digestibilidade e difícil aproveitamento para fins diversos. Uma alternativa para disponibilizar os componentes dessa matriz é a degradação microbiana da celulose ou utilização de um complexo enzimático contendo celulases produzidas por bactérias ou fungos. Neste trabalho, foi avaliado o tempo de ação de complexo enzimático celulolítico, produzido durante a fermentação em estado sólido por Rhizopus oryzae, a fim de aumentar a digestibilidade proteica de farelo de trigo. O complexo extraído do farelo e casca de arroz foi aplicado no farelo de trigo sob condições ótimas (1 hora a 50 ºC e 100 rpm ) promoveu um aumento de 35,4 % na digestibilidade protéica.Palavras-chave: Farelo de trigo, celulases, hidrólise. INTRODUÇÃOOs materiais lignocelulósicos, como o farelo de trigo, caracterizam-se pela variabilidade do conteúdo de proteínas e baixa digestibilidade, quando comparado com outras fontes proteicas, o que dificulta sua utilização como alimento no seu estado original. O emprego de tratamentos prévios permite aumentar a sua digestibilidade e disponibilizar os nutrientes presentes nestas matrizes. A maior dificuldade para o aproveitamento dos resíduos lignocelulósicos está representada pela barreira física formada pela lignina, o que também impede o aproveitamento da celulose nativa 1 e de outros compostos ligados a ela para diversos fins industriais.A degradação microbiana da celulose ou utilização de um complexo enzimático contendo carboidrases, em especial as celulases produzidas por um espectro de bactérias e fungos, é um caminho aplicável para recuperar estes materiais carbonados 2 para uso tecnológico. O objetivo deste trabalho foi avaliar o tempo de ação de um complexo enzimático celulolítico, produzido durante a fermentação em estado sólido por Rhizopus oryzae, a fim de aumentar a digestibilidade proteica de farelo de trigo, visando aplicá-lo em formulações alimentícias.
