The purpose of this study was to achieve a better therapeutic efficacy and patient compliance in the treatment for vaginitis. Clotrimazole (1%) has been formulated in a vaginal gel using the thermosensitive polymer Pluronic F127 (20%) together with mucoadhesive polymers such as Carbopol 934 and hydroxypropylmethylcellulose (0.2% for both). To increase its aqueous solubility, clotrimazole was incorporated as its inclusion complex with 1:1 molar ratio with β-cyclodextrin. The inclusion complex was thoroughly characterized using various techniques, including 1 H NMR spectroscopy, FT IR spectrophotometry, differential scanning calorimetry, scanning electron microscopy, phase solubility studies, and determination of stability constant (k 1:1 ). The gelation temperature and rheological behavior of different formulations at varying temperatures were measured. In vitro release profiles of the gels were determined in pH 5.5 citrate buffer. It was observed that complexation with cyclodextrin slowed down the release of clotrimazole considerably. Carbopol 934, on the other hand, was found to interact with β-cyclodextrin, inducing precipitation. As far as rheological properties are concerned, thermosensitive in situ gelling was obtained with formulations containing drug: cyclodextrin complex rather than with free drug. Thus, the optimum formulation for a controlled-release thermosensitive and mucoadhesive vaginal gel was determined to be clotrimazole:β-cyclodextrin 1% with 0.2% hydroxypropylmethylcellulose in Pluronic F127 gel (20%) providing continuous and prolonged release of active material above MIC values.
Tropical hilsa shad, which is an anadromous fish migrating from the sea to spawn in freshwater river, constitutes an important fishery in some Asian and Middle East countries, particularly in Bangladesh. But, historical information on hilsa biology and ecology has confronted the scientists and policymakers with research and management challenges. We have reviewed both the old and recent findings on hilsa fishery to document the status of knowledge and potential gaps, necessary to comprehend for formulating a more effective fishery management plan. Thus, there has been a decline in hilsa catches in the riverine system associated with shift in fish migration routes, indiscriminate harvesting of brood and juvenile fish, and degradation of habitat. Specifically, the riverine hilsa catches peaked in the 1960s, declining thereafter, and became relatively abundant in marine waters since 1990s. Biological data indicated that hilsa goes through multiple reproductive cycles; therefore, a comprehensive understanding of reproductive biology, recruitment by various cohorts, stock abundance and habitats across the life cycle are necessary to accurately impose fishery regulatory measures, such as fishing ban in spawning season in Bangladesh. Moreover, domestication initiative is important for artificial seed production and mariculture development of hilsa that can not only offer economic return to small‐scale farmers but also reduce the growing pressure on capture fishery. Importantly, the arrangement of co‐management is found ideal as fishermen, scientists and managers can work jointly to improve the regulatory processes and to sustain the hilsa fishery over time.
Rapamycin (Sirolimus) is a macrolide lactone with antifungal, immunosuppressant, and antiproliferative actions. The mechanism of rapamycin action involves the inhibition of mTOR and subsequent cytostasis. Rapamycin also prevents angiogenesis in tumors and can prevent cancer cells' resistance to other chemotherapeutic agents. However, very poor water solubility, bioavailability, only slight solubility in acceptable parenteral excipients, chemical instability, and major sequestration (95%) of free rapamycin into the erythrocytes have prevented its development as an anticancer drug. To address these problems, it was attempted to develop liposomal rapamycin delivery systems in this study. Conventional and pegylated liposomes were prepared with various lipid and cholesterol ratios. They were then characterized; these liposomes contained 0.68-0.90 mg of rapamycin per milliliter of liposome suspension. Having suitable particle size, these liposomes successfully retained the entrapped drug. Both types of liposomes were found to be effective; however, conventional liposomes showed better antiproliferative activity against MCF-7 cells than pegylated liposomes. But, pegylated liposome showed better stability than conventional liposomes. In conclusion, the enhanced permeability and retention effects of tumors should provide the opportunity for pegylated liposomal rapamycin to be applied as an intravenous drug-delivery system for targeted delivery to cancer cells, avoiding the major sequestration of free rapamycin into the erythrocytes.
Recently mud crab (Scylla olivacea) has emerged as a potential export commodity. Its farming is increasing rapidly in Bangladesh because of its lucrative price and high demand in international market. However, the farming or fattening of mud crab is totally dependent on capture of wild crablets. A huge quantity of mud crabs is being caught indiscriminately from nature and thus putting intense pressure on its wild stock, which is a major concern of conservation. Imposing ban at peak breeding season and setting a legal capture size are considered effective management tools for safe migration and spawning of berried females. There is contradictory information on breeding season of mud crab in Bangladesh. Thus, it is crucial to identify the peak breeding season of it and to formulate conservation policies to protect the berried crabs. Live crabs were collected year-round from the river adjacent to southwest part of Sundarbans. Although mud crab breeds throughout the year, March-April was identified as the peak breeding season of it. The second peak was observed in August-September. The highest abundance of crablets (1-2 months age) was recorded in May-June, and the lowest in January-February. Immature crablets were found all through the year that proves that mud crab is a continuous breeder. The highest amount of GSI value (11.6) was observed in February-March, which indicates that the breeding is very near. The size at first maturity (M 50 ) was estimated as 95.5 mm CW which lied at size class 91-100 mm (CW). The findings from the study will assist in fixing up the banning time and thus protecting the mother crabs; and will assist in formulating police to conserve and protect juvenile crabs in the sense that the minimum legal capture size of crabs will allow the adults to mate and spawn at least once before their capture.
