Nitrones 9 and 10, formed by treatment of the corresponding aldehydes with N-substituted hydroxylamines, are converted into the cis fused bicyclic compounds 11 and 12, respectively. While this intramolecular cycloaddition as a rule occurs der Z-Konfiguration vorliegen, kann es aber auch zur Reaktion aus der E-Konfiguration kommen'), wobei sich die Anordnung von R2 relativ zu R3/R4 umkehren wurde. Grundsatzlich lassen sich durch 1,3-dipolare Cycloadditionen zwischen Nitronen und 1,Zdisubstituierten Alkenen Isoxazolidine mit drei Chiralitatszentren aufbauen (1). Entsprechendes gilt fur die intramolekulare 1,3-dipolare Cycloaddition zwischen Nitron-und Alkengruppierung (Gl. 2). Hier ist prinzipiell die Bildung der beiden stereoisomeren cis-und trans-verknupften Produkte 2 und des regioisomeren uberbruckten Produkts 3 moglich. Die grundlegenden Arbeiten von LeBel') ergaben entscheidende Unterschiede zwischen den Verbindungen l a und 1 b. Wegen der Kurze der verbindenden Kette in l a ist in diesem Falle die Reaktion uber einen 2-exo-Ubergangszustand so stark begunstigt, dafi sie wie bei der Stammverbindung (R' = Me, R2 = H) auch bei den meisten anderen Verbindungen dieses Typs hoch regio-und stereoselektiv unter Bildung cis-verknupfter 3-Oxa-2-azabicyclo[3. Enantiomerically Pure 3,?-Dioxa-2-azabicyclo[3.3.0]octanes by Intramolecular 1,3-Dipolar Cycloaddition of NitronesStarting with chiral non-racemic esters of lactic acid 7 and mandelic acid 8, respectively, chiral nitrones 4 have been prepared. They spontaneously undergo an intramolecular cycloaddition which proceeds with asymmetric induction by the chiral center yielding exclusively the bicyclic compounds 5 in enantiomerically pure form. Reductive opening of the isoxazolidine ring affords tetrahydrofuran derivatives 13 with an y-amino alcohol group. Reaction of 13 with (-)-camphanoyl chloride gives the esters 14. Their 'H-NMR spectra reveal a diastereomeric purity of at least 98%.Fur Nitrone vom Typ der 2-substituierten 5-Hexen-1-Abtrennung von 6 -mit hohem EnantiomerenuberschuS imin-N-oxide 1 sind die Ubergangszustande der intramo-zu erhalten. In all diesen Fallen verlauft die intramolekulare Cycloaddition rnit hoher Diastereoselektivitat; es entstehen ausschlieI3lich oder uberwiegend bicyclische Verbindungen vom Typ 2 mit dem Rest R1 in exo-Position4).Wahrend Heteroatom-substituierte 6-Hepten-l-imin-Noxide in Analogie zu den einfachen 6-Hepten-1-imin-N-oxiden bei ihrer intramolekularen Cycloaddition in der Regel Gemische aus cis-und trans-anellierten und iiberbriickten Produkten ergeben, entstehen aus Nitronen 4 (R1 = H) und ihren Schwefelanalogen regio-und stereoselektiv ausschliel3lich cis-verknupfte anellierte Produkte 5! Wir erwarteten daher bei der intramolekularen Cycloaddition von 2-substituierten Nitronen 4 die Bildung der cis-anellierten bicyclischen Verbindungen 5 und 6 mit deutlicher Bevorzugung von 5. Ausgehend von chiralen, nicht-racemischen Verbindungen sollte es auch moglich sein, 5 -gegebenenfalls nach 4 5 n 6Voraussetzung dafur ist allerdings, daI3 die Darstellung der chiralen Nitrone 4 und ihre intramolekulare Cycloaddition ohne nennenswerte Racemisierung ablauft. Eine Racemisierung von Nitronen mit C-2 als chiralem Zentrum ware immerhin iiber die entsprechende tautomere Hydroxyenamin-Form moglich. Diese laDt sich zwar nur in Ausnahmefallen direkt nachweisen6), ihr intermediares Auftreten wird aber durch die leichte Bildung von ,,Nitron-Dimeren" durch Cycloaddition zwischen Nitron-und Hydroxyenamin-Form angezeigt 6d).
