To improve the efficiency of topical ocular drug administration, we focused on development of a nanoparticles loaded contact lens to deliver the hydrophobic drug over a prolonged period of time. The cross-linked nanoparticles based on PCL (poly ε-caprolactone), 2-hydroxy ethyl methacrylate (HEMA), and poly ethylene glycol diacrylate (PEG-DA) were prepared by surfactant-free miniemulsion polymerization. The lens material was prepared through photopolymerization of HEMA and N-vinylpyrrolidone (NVP) using PEG-DA as the cross-linker. Effects of nanoparticles loading on critical contact lens properties such as transparency, water content, modulus and ion and oxygen permeabilities were studied. Nanoparticles and hydrogel showed high viability, indicating the absence of cytotoxicity and stimulatory effect. Drug release studies revealed that the hydrogel embedded with nanoparticles released the drug for a period of 12 days. The results of this study provide evidence that nanoparticles loaded hydrogels could be used for extended delivery of loteprednol etabonate and perhaps other drugs.
Polycaprolactone-tricalcium phosphate (PCL-TCP), a new composite scaffold, has been shown to facilitate early revascularization and speed up bone regeneration process. The objective of this study was to evaluate the effect of PCL-TCP seeded with mesenchymal stem cells (MSCs) on healing of the vertical bone critical sized defect in dog's mandible. Bone marrow aspirate from dog humerous was cultured and the stemness of the cells was examined by differentiation staining methods and flow cytometric analysis. Third passage subculture cells (5 × 10⁵ cells) were loaded on 20 × 10 × 10 mm³ and incubated for 48 h. The presence of MSCs in the pores was evaluated by scanning electron microscope. Bilateral mandibular premolar teeth were extracted in four dogs and the buccal and lingual bone plates were reduced to make a vertical defect. Cell-loaded scaffolds were fixed in right side and left side received pure PCL-TCP scaffolds as a control side defects. Histomorphometric analysis after 8 weeks of the scaffold implantation showed higher amount of lamellar bone in the test side (48.63%) than control side (17.27%) (p < 0.05).The results suggest that PCL-TCP may be an appropriate scaffold for loading MSCs in bone regeneration.
Currently, mesenchymal stem cells (MSCs) are used in veterinary clinical applications. Bone marrow and adipose tissue are the most common sources of stem cells derived from adult animals. However, cord blood which is collected non-invasively is an alternative source of stem cells other than bone marrow and adipose tissue. Moreover, high availability and lower immunogenicity of umbilical cord blood (UCB) haematopoietic stem cells compared to other sources of stem cell therapy such as bone marrow have made them a considerable source for cell therapy, but MSCs is not highly available in cord blood and their immunogenicity is poorly understood. In this study, the cells with spindle morphology from 7 of 9 bovine UCB samples were isolated and cultured. These mesenchymal stromal cells were successfully differentiated to osteocytes, chondrocytes and adipocytes. In addition, Oct-4 and SH3 were determined by RT-PCR assay. It is the first report of isolation, culture, characterization and differentiation of bovine umbilical stem cells.
