As atividades amilolítica e pectinolítica de 45 isolados de Alternaria solani, provenientes de diferentes hospedeiros, foram estimadas por meio da difusão enzimática em meio sólido específico e mensuração do halo de degradação do substrato. Todos os isolados degradaram pectina. Apenas 17 isolados apresentaram atividade amilolítica, sendo nove isolados provenientes de batateira. Somente o isolado AS18 se destacou como bom produtor de ambas as enzimas. Uma vez que a atividade pectinolítica foi mais evidente, avaliou-se a influência de pectinases na agressividade de A. solani ao tomateiro. Para isso, cinco isolados (2 de berinjela, 2 de tomateiro e 1 de batateira) contrastantes quanto à produção de pectinases foram selecionados para testes em folíolos destacados e plantas inteiras. Quatro isolados foram utilizados no teste em folíolos destacados (AS6, AS7, AS12 e AS26), e constatou-se haver variabilidade patogênica. A correlação obtida entre o tamanho das lesões e a atividade pectinolítica foi de r = 0,963 (P = 0,087). Cinco isolados (AS6, AS7, AS12, AS25 e AS26) foram inoculados em plantas inteiras de tomate. Os isolados não diferiram quanto ao número de lesões/cm² de área foliar, porém variaram em agressividade. Houve correlação (r = 0,916; P = 0,042) entre a atividade de pectinases e o índice de doença, sugerindo possível papel para as enzimas pécticas durante a infecção de A. solani em tomateiro. É provável que as diferenças no perfil enzimático dos isolados estejam associadas ao hospedeiro original de onde os mesmos foram obtidos. Os resultados reforçaram evidências de especificidade por hospedeiro em populações de A. solani.
A produção de pectinases de 40 isolados de Colletotrichum gloeosporioides, obtidos de Stylosanthes spp. em diferentes regiões produtoras, foi estimada por meio de difusão enzimática em meio específico e mensuração do halo de degradação de pectina. Em todos os isolados havia atividade pectinolítica, sendo divididos em cinco grupos distintos. Na maior parte dos isolados notou-se moderada atividade de pectinases. Em geral, isolados de Camapuã-MS tiveram menor diversidade genética para a produção de enzimas pécticas. Com base na atividade pectinolítica, isolados do patógeno foram selecionados e inoculados em genótipos de S. capitata (GC1081, GC1084, GC1086 e GC1094), S. scabra (GC1496) e S. macrocephala (GC1511), visando analisar a influência das pectinases na agressividade ao hospedeiro. Inicialmente, instalou-se ensaio de agressividade com os isolados CG656, CG657, CG687, CG706 e CG707, sendo constatada agressividade diferenciada apenas quando inoculados em S. scabra. Enquanto CG657, CG706 e CG707 foram os mais agressivos, CG656 foi o menos agressivo. Outro teste de agressividade avaliou os isolados CG768, CG769, CG772, CG775, CG779 e CG781. Estes se diferenciaram quanto à agressividade ao S. capitata (GC1081 e GC1094) e S. macrocephala (GC1511). Em geral, CG768 comportou-se como pouco agressivo, enquanto que CG772, CG775 e CG781 como os mais agressivos. Para ambos, nos ensaios de agressividade, não se evidenciou aumento da severidade da antracnose com o aumento da atividade pectinolítica. Nas condições analisadas, a agressividade de C. gloeosporioides ao estilosantes não foi influenciada pelas enzimas pécticas.
Ensaios foram conduzidos para verificar a presença, o número de cópias e de sítios de integração de pAN7-1 no genoma de mutantes de M. grisea I-22 com patogenicidade alterada em arroz. Foram analisados T41, T93, T251 (gerados por mutagênese REMI) e T108 (oriundo de mutagênese convencional), os quais exibiram diferentes fenótipos mutantes. O DNA total desses mutantes foi submetido à reação em cadeia de polimerase (PCR) e às análises de hibridização com o vetor (Southern blot). A presença de pAN7-1 no genoma de todos os mutantes foi confirmada por PCR. Segundo as análises de Southern blot, T41 exibiu duas integrações do vetor, ambas na forma de cópia única. No genoma de T93 também foram detectados dois sítios de inserção de pAN7-1, um dos quais envolvendo múltiplas cópias do vetor. Os resultados indicaram a presença de apenas uma cópia do vetor em um único sítio nos genomas de T108 e T251. O padrão de integração em T251 foi o único a sugerir a ocorrência de evento REMI. As diferenças quanto ao tamanho dos fragmentos com homologia a pAN7-1 refletiram a possível aleatoriedade dos eventos de integração no genoma de M. grisea.
