Resumo -O objetivo deste trabalho foi identificar e quantificar a nematofauna fitopatogênica de sementes de forrageiras, destinadas aos mercados interno e externo. Phytopathogenic nematofauna of commercial seeds of tropical foragesAbstract -The objective of this work was to identify and quantify the phytopathogenic nematofauna of forage seeds destined for internal and external markets. Seeds of Brachiaria brizantha (cvs. Marandu and Xaraés), B. decumbens cv. Basilisk, Panicum maximum (cvs. Massai, Mombaça and Tanzania), and Stylosanthes capitata and S. macrocephala, components of cv. Estilosantes Campo Grande, were examined. The extractions were made by centrifugal flotation in solution of sucrose with kaolin. Aphelenchoides and Ditylenchus genera were detected in forage grasses. The highest number of Aphelenchoides spp. specimens was observed in seeds of cv. Marandu, followed by seeds of cvs. Xaraés, Basilisk, and Tanzania. Regarding Ditylenchus spp. population density, higher values were detected in seeds of cvs. Marandu, Xaraés, and Basilisk. The results suggest that two or more species of Aphelenchoides and Ditylenchus are associated to forage grass seeds destined for Brazilian market. Phytopathogenic nematodes are not detected in Estilosantes Campo Grande seeds, neither in seeds of cvs. Marandu, Mombaça and Tanzania destined for exportation.
Resumo AbstractThe sanitary quality of 26 lots commercial seeds of tropical forages, produced in different regions (2004-05 and 2005-06) was analyzed. The lots were composed of seeds of Panicum maximum ('Massai', 'Mombaça' e 'Tanzânia') and stylo ('Estilosantes Campo Grande' -ECG). Additionally, seeds of two lots of P. maximum for exportation were analyzed. The blotter test was used, at 20ºC under alternating light and darkness in a 12 h photoperiod, for seven days. The Aspergillus, Cladosporium and Rhizopus genus consisted the secondary or saprophytes fungi (FSS) with greatest frequency in P. maximum lots. Protoplastos são ferramentas biológicas importantes para pesquisas em fungos filamentosos, sendo empregados intensamente em transformação genética. O isolamento de protoplastos de Magnaporthe grisea foi facilitado com Novozym 234, contudo, este complexo enzimático encontra-se indisponível no mercado. Assim, objetivou-se comparar a eficiência de enzimas líticas disponíveis comercialmente na obtenção de protoplastos de M. grisea. Paralelamente, analisaram-se estabilizadores osmóticos, tempos de digestão e freqüência de regeneração. Maior produção de protoplastos foi obtida com o uso simultâneo de Lysing Enzymes e Cellulase Onozuka R-10. O uso de 10 ou 15 mg de cada complexo enzimático, em 3 mL de estabilizador osmótico, resultou em maior liberação de protoplastos. O melhor estabilizador osmótico foi MgSO4 1,2 M / NaH2PO4 0,01 M, pH 5,8, seguido por MgSO4 0,8 M / NaH2PO4 0,01 M, pH 5,8. O isolamento de protoplastos foi monitorado a cada 60 minutos, atingindo o máximo após incubação por 3 a 6 horas. No entanto, maior freqüência de regeneração (19,4%) foi registrada para protoplastos obtidos após 3 horas de hidrólise enzimática.

Os resultados demonstraram existir correlação positiva entre os valores de incidência e severidade da mela.A crescente demanda por forrageiras tropicais tem impulsionado o desenvolvimento da indústria sementeira no Brasil. Atualmente, o país é considerado o maior produtor, consumidor e exportador de sementes (3). A maior parte da produção de sementes (95%) tem sido destinada ao comércio interno e o restante exportado, principalmente para países da América do Sul e Central (12). Marandu the path ogen occu rred in final phase of culture, while in cv. Xaraes the honeydew was detected in phase of intense bloom. Constant increases of disease intensity were observed in two genotypes. The honeydew occurred in 64% of panicles of cv. Marandu and in 81% of panicles of cv. Xaraes; about 20% and 18% of flowers/seeds, respectively, was affected. The results verified high positive correlation between the values of incidence and severity of disease.
