Infection of cattle with lumpy skin disease virus (LSDV) is very important from the aspect of livestock production. Although it can cause significant economic losses, available serological assays are still not sufficiently efficient and reliable. A 3-day VNT was performed using Madin-Darby bovine kidney (MDBK) cell line and LSDV isolated from clinically infected cow to improve serological diagnostics of lumpy skin disease (LSD). In total, 325 cattle sera samples were tested in order to compare the performances of VNT and ELISA. Tested samples originated from 125 cows before the occurrence of LSD in the Republic of Serbia and 200 tested samples originated from vaccinated cows. Sera samples from vaccinated cows were collected starting from the vaccination day to 4 months after vaccination. In 7 different time intervals after vaccination sampling was carried out in 20 cows originating from one herd and in 3 different time intervals in 20 cows originating from a different herd each time of sampling. Out of 200 samples from vaccinated cows, antibodies against LSDV were detected in 68 (34%) samples by VNT, and in 60 (30%) samples by ELISA. No positive finding was detected by VNT in samples collected before the occurrence of LSD in Serbia, while one positive finding was detected in the same samples by ELISA. The first presence of antibodies in vaccinated cattle was detected by both tests 20 days after vaccination, and the largest number of animals with antibodies against LSDV was detected 30 days after vaccination. Comparing the results obtained by VNT and ELISA, it was calculated that kappa index was 0.913. The specificity of VNT and ELISA was 100% and 99.2%, respectively. VNT is simpler to perform compared to the recommended virus neutralization test by the OIE and can improve LSD serological diagnostics with additional sensitivity testing.
The aim of this study is to summarize the data on the occurrence, presence and prevalence of some zoonotic flaviviruses, which have been actively circulating in the territory of the Republic of Serbia during past decade. The genus Flavivirus, family Flaviviridae,encompasses vector-transmitted viruses with zoonotic potential. The genus is comprised of more than 70 viruses transmitted to humans by mosquitoes and ticks. Some of those are associated only with human diseases (such as yellow fever and dengue fever), whereas others can cause diseases in both humans and animals. Animals are major reservoirs and primary hosts for the latter group. The virus transmission to other animal species or humans occurs via diverse vectors – mostly mosquitoes and ticks. A range of these virus species is widely distributed worldwide, especially in tropical and sub-tropical climatic zones. Some representatives of these species have only recently been identified at the territory of Republic of Serbia. This paper gives an evidence on the presence and distribution of West-Nile virus, tick-borne encephalitis (TBE) virus and Usutu virus that were conducted within the study in Serbia. The research pointed out the presence of recently detected West-Nile virus and Usutu virus as well as the current circulation of tick-borne encephalitis virus, for which only limited serological evidence on the presence of virus-specific antibodies in humans was available so far. In the aspect of public health, the full understanding of the importance of identification and prevalence of different flaviviruses still require further comprehensive entomologicalacarological, seroepidemiological, clinical and virological research.
The results of the Serbian national integrated West Nile virus (WNV) surveillance program conducted in 2018 and funded by the Serbian Veterinary Directorate are presented. The WNV surveillance program encompassed the entire territory of Serbia and was conducted by the veterinary service in collaboration with entomologists and ornithologists. The objective of the program was early detection of WNV circulation in the environment and timely reporting to the public health service and local authorities to increase clinical and mosquito control preparedness. The program was based on the detection of WNV presence in wild birds (natural hosts) and mosquitoes (virus vectors) and on serological testing of sentinel horses (WNV-specific IgM antibodies). The season 2018 was confirmed to be the season of the most intensive WNV circulation with the highest number and severity of human cases in Serbia ever reported. The most intense WNV circulation was observed in the northern and central parts of Serbia including Vojvodina Province, the Belgrade City area, and surrounding districts, where most positive samples were detected among sentinel animals, mosquitoes and wild birds. The majority of human cases were preceded by the detection of WNV circulation during the surveillance. The WNV surveillance program in 2018 showed satisfactory results in the capacity to indicate the spatial distribution of the risk for humans and sensitivity to early detection of WNV circulation in the environment.
During July 2018, the first clinical case of neurological West Nile virus (WNV) infection was reported in a Belgian sports mare in Belgrade, Serbia. Typical symptoms, such as hypersensitive skin reaction, disorientation, weakness, ataxia and the loss of equilibrium were reported. Detection of WNV IgM antibodies by commercial ELISA in the serum samples of the diseased mare strongly indicated acute infection. The ELISA positive results were confirmed by VNT. Hematological and biochemical parameters were in the reference range. The only finding was a minor lymphopenia. WNV RNA was not detected by RT-qPCR in the blood sample extracted seven days after the disease had broken out. The horse improved clinically in two weeks while other horses at the same premises remained asymptomatic. The clinical, serological, biochemical and molecular analyses applied confirmed the first clinical case of neuroinvasive WNV infection in horses in Serbia. The West Nile virus has been circulating in Serbia in the last decade in mosquitoes, birds, and horses, but no evidence of equine WNV clinical cases were registered so far.
Od 2014 godine nekoliko epizootija izazvanih visoko patogenim virusom avijarne infl uence podtipa H5N8, izbilo je u Aziji, Evropi i severnoj Americi. Ovaj tip virusa infl uence uzrokovao je infekcije domaće živine i divljih ptica. Globalnom širenju virusa doprinele su intrakontinentalne i interkontinentalne migracije ptica. Prvi slučaj visoko patogene avijarne influence podtipa H5N8 u Srbiji detektovan je kod labudova (Cygnus olor),lociranih na rukavcu Dunava (područje Koviljskog-Petrovaradinskog rita) u novembru 2016.godine. Tokom ove epizootije, veći broj bolesnih i uginulih labudova pronađen je oko staništa migratornih ptica. Pokazalo se da su labudovi tokom epizootije bili visoko osetljiva i najviše pogođena vrsta ptica virusom infl uence podtipa H5N8. Oboleli labudovi su ispoljavali neurološke simptome, uključujući tortikolis, inkoordinaciju i ataksiju. Uovom radu su predstavljeni rezultati postmortalnog pregleda labudova inficiranih virusom infl uence. Ispitivanje je rađeno na leševima deset uginulih labudova. Najdominantnije patološke promene izazvane virusom influence podtipa H5N8 bile su nekroze pankreasa, petehijalna krvarenja u subepikardijumu i masnom tkivu mezenterijuma. Kod većine uginulih labudova bila je prisutna kongestija pluća.
Sheeppox (SPP), goatpox (GTP), and lumpy skin disease (LSD) are economically significant pox diseases of ruminants, caused by sheeppox virus (SPPV), goatpox virus (GTPV), and lumpy skin disease virus (LSDV), respectively. SPPV and GTPV can infect both sheep and goats, while LSDV mainly affects cattle. The recent emergence of LSD in Asia and Europe and the repeated incursions of SPP in Greece, Bulgaria, and Russia highlight how these diseases can spread outside their endemic regions, stressing the urgent need to develop high-throughput serological surveillance tools. We expressed and tested two recombinant truncated proteins, the capripoxvirus homologs of the vaccinia virus C-type lectin-like protein A34 and the EEV glycoprotein A36, as antigens for an indirect ELISA (iELISA) to detect anti-capripoxvirus antibodies. Since A34 outperformed A36 by showing no cross-reactivity to anti-parapoxvirus antibodies, we optimized an A34 iELISA using two different working conditions, one for LSD in cattle and one for SPP/GTP in sheep and goats. Both displayed sound sensitivities and specificities: 98.81% and 98.72%, respectively, for the LSD iELISA, and 97.68% and 95.35%, respectively, for the SPP/GTP iELISA, and did not cross-react with anti-parapoxvirus antibodies of cattle, sheep, and goats. These assays could facilitate the implementation of capripox control programs through serosurveillance and the screening of animals for trade.
In the Northern part of Serbia, which is part of Pannonian Basin, mute swan (Cygnus olor) population has notably increased over the last few decades. Like other birds from Anatidae family, mute swans are a host of numerous endoparasite species. The aim of this study was to acquire the data on identification and prevalence of endoparasites in mute swans in the Republic of Serbia, as that information is lacking. Individual faecal samples of sixty-eight adult mute swans were examined for the presence of endoparasites. Coprological examination was performed using flotation and sedimentation technique with saturated ZnSO4 solution. The samples were collected from December 2016 to March 2017, during epizootic of highly pathogenic avian influenza H5N8 in the Republic of Serbia. Dead mute swans were collected from twelve different locations in the Northern part of Serbia. Altogether, 39.7% of the examined fecal samples contained different parasites. The endoparasitic fauna was divers and included 3 species of nematodes, 2 cestodes species, one trematode and one protozoan species. Nematodes were the most prevalent helminthes and among these the most frequent nematode species found was Heterakis dispar (17.6%), followed by Echinuria uncinata (5,8%) and Ascaridia spp. (4,4%). Four mixed infections were found, where double infection was the most prevalent. No endoparasites were found in faecal samples of mute swans collected from the locations Sombor and Titel. As this study covers a small population of mute swans, more detailed studies need to be carried out on a larger population in order to gain an insight into the diversity and prevalence of endoparasites in the Republic of Serbia.
Surgical sterilization (ovariohysterectomy) is one of the most common surgical procedures in small veterinary practice because it represents the safest way to control the population of cats. Th e aim of this study is to compare postoperative recovery in three groups of cats (estrous, anestrous and gravid cats) aft er performed ovariohysterectomy using combination of xylasine/ketamine anesthesia and, on the basis of obtained data, determine whether there are some signifi cant diff erences in postoperative recovery in these three groups of cats. Th is study included 45 cats that were divided into three groups. First group consisted of 15 cats in estrous, second group consisted of 15 cats in anestrous and the third group consisted of 15 gravid cats. Ovariohysterectomy was performed in private veterinary clinic. Anesthetic protocol was the same for all cats, that is, combination of xylasine and ketamine at appropriate doses according to body weight. Parameters that were followed through postoperative course were appetite, body temperature, appearance of the wound and activity. Data was collected on the first and third day aft er surgery. Analysis of the collected data shows relatively uniform results among groups of cats considering all followed parameters. Results in this study did not show any signifi cant diff erences in response to xylasine/ketamine anesthesia among cats of diff erent reproductive status through monitoring of postoperative course.
