Surface ocean productivity mediates the transfer of carbon to the deep ocean and in the process regulates atmospheric CO 2 levels. A common axiom in oceanography is that large phytoplankton contribute disproportionally to the transfer of carbon to the deep ocean because of their greater ability to escape grazing pressure, build biomass, and sink. In the present study, we assessed the relationship of net community production to phytoplankton assemblages and plankton size distribution in the Sub-Antarctic Zone and northern reaches of the Polar Frontal Zone in the Australian sector of the Southern Ocean. We reanalyzed and synthesized previously published estimates of O 2 /Ar net community oxygen production (NCP) and triple-O 2 isotopes gross primary oxygen production (GPP) along with microscopic and pigment analyses of the microbial community. Overall, we found that the axiom that large phytoplankton drive carbon export was not supported in this region. Mixed-layer-depth-integrated NCP was correlated to particulate organic carbon (POC) concentration in the mixed layer. While lower NCP/GPP and NCP/POC values were generally associated with communities dominated by smaller plankton size (as would be expected), these communities did not preclude high values for both properties. Vigorous NCP in some regions occurred in the virtual absence of large phytoplankton (and specifically diatoms) and in communities dominated by nanoplankton and picoplankton. We also observed a positive correlation between NCP and the proportion of the phytoplankton community grazed by microheterotrophs, supporting the mediating role of grazers in carbon export. The novel combination of techniques allowed us to determine how NCP relates to upper ocean ecosystem characteristics and may lead to improved models of carbon export.

Ultrastructure and large subunit rDNA sequences of Lepidodinium viride reveal a close relationship to Lepidodinium chlorophorum comb. nov. (= Gymnodinium chlorophorum) SUMMARY The ultrastructure of the green dinoflagellate Lepididodinium viride M. M. Watanabe, S. Suda, I. Inouye Sawaguchi et Chihara was studied in detail. The nuclear envelope possessed numerous chambers each furnished with a nuclear pore, a similar arrangement to that found in other gymnodinioids. The flagellar apparatus was essentially identical to Gymnodinium chlorophorum Elbrächter et Schnepf, a species also containing chloroplasts of chlorophyte origin. Of particular interest was the connection of the flagellar apparatus to the nuclear envelope by means of both a fiber and a microtubular extension of the R3 flagellar root. This feature has not been found in other dinoflagellates and suggests a close relationship between these two species. This was confirmed by phylogenetic analysis based on partial sequences of the large subunit (LSU) rDNA gene of L. viride, G. chlorophorum and 16 other unarmoured dinoflagellates, including both the 'type' culture and a new Tasmanian isolate of G. chlorophorum. These two isolates had identical sequences and differed from L. viride by only 3.75% of their partial LSU sequences, considerably less than the difference between other Gymnodinium species. Therefore, based on ultrastructure, pigments and partial LSU rDNA sequences, the genus Lepidodinium was emended to encompass L. chlorophorum comb. nov.

The order Gymnodiniales comprises unarmoured dinoflagellates. However, the lack of sequences hindered determining the phylogenetic positions and systematic relationships of several gymnodinioid taxa. In this study, a monophyletic clade was defined for the species Ceratoperidinium margalefii Loeblich III, Gyrodinium falcatum Kofoid & Swezy, three Cochlodinium species, and two Gymnodinium-like dinoflagellates. Despite their substantial morphotypic differentiation, Cochlodinium cf. helix, G. falcatum and 'Gymnodinium' sp. 1 share a common shape of the acrobase. The phylogenetic data led to the following conclusions: (1) C. margalefii is closely related to several unarmoured dinoflagellates. Its sulcus shape has been observed for the first time. (2) G. falcatum was erroneously assigned to the genus Gyrodinium and is transferred to Ceratoperidinium (C. falcatum (Kofoid & Swezy) Reñé & de Salas comb. nov.). (3) The genus Cochlodinium is polyphyletic and thus artificial; our data support its separation into three different genera. (4) The two Gymnodinium-like species could not be morphologically or phylogenetically related to any other gymnodinioid species sequenced to date. While not all studied species have been definitively transferred to the correct genus, our study is a step forward in the classification of inconspicuous unarmoured dinoflagellates. The family Ceratoperidiniaeceae and the genus Ceratoperidinium are emended.

Toxin analysis of 15 species of Kareniaceae revealed the presence of karlotoxin, KmTx 2, in only a single species (Karlodinium veneficum) but with variable activity in strains from the Swan (KmSwanTx 2‐1, 2.1 pg · cell−1; and KmSwanTx 2‐2, 0.53 pg · cell−1), Huon (KmHuonTx 2, 0.86 pg · cell−1), and Derwent rivers (<0.001 pg · cell−1) in Australia. A newly isolated Southern Ocean species, Karlodinium conicum, contained a novel poorly hemolytic karlotoxin analogue (KmconicumTx, 2.8 pg · cell−1). The hemolytic potency (HD50%) of the Australian karlotoxins were as follows: KmSwanTx 2‐1 (65.9 ± 4.8 ng) and KmSwanTx 2‐2 (63.4 ± 3.7 ng), KmHuonTx 2 (343 ± 4.9 ng), and KmconicumTx (>4,000 ng). Species from the closely related genera Takayama (T. helix, T. tasmanica, T. tuberculata), Karenia (K. asterichroma, K. brevis, K. mikimotoi, K. papilionacea, K. umbella), and Karlodinium (Ka. australe, Ka. antarcticum, Ka. ballantinum, Ka. corrugatum, Ka. decipiens) were all consistently negative for karlotoxin production. Brevetoxin (PbTx) was only detected in K. brevis, and hemolytic activity was only observed in Ka. veneficum strains.
