The lipid, FA, and sterol composition of the New Zealand green lipped mussel (NZGLM, Perna canaliculus) and of the Tasmanian blue mussel (TBM, Mytilus edulis) were compared using TLC-FID and GC-MS. The respective mussel species were obtained from three different sites in both New Zealand (NZ) and Tasmania. Lipid class distribution of both mussel species was characterized by a high proportion of phospholipid (PL, 57-79%) and TG (10-25%), FFA (7-12%), and sterols (ST, 12-18%). The NZGLM had higher proportions of TG, FFA, and ST (P < 0.01), whereas the TBM had a higher proportion of PL (P < 0.01). There were higher proportions of total PUFA, saturated FA, n-3 FA, and hydroxy and nonmethylene-interrupted FA (P < 0.05) in the TBM compared with the NZGLM. The major FA in the NZGLM were 16:0 (15-17%), 20:5n-3 (14-20%), and 22:6n-3 (11-17%). The same FA dominated lipids in the TBM, although there were significantly higher proportions of 16:0 (P = 0.000) and 22:6 n-3 (P = 0.003) and lower proportions of 20:5n-3 (P = 0.0072) in the TBM. A novel PUFA, 28:8n-3, was detected in both mussels with higher amounts in the TBM, which probably reflects a greater dietary contribution of dinoflagellates for this species. Cholesterol was the dominant sterol in both mussels. Other major sterols included brassicasterol, 22-methylcholesterol, trans-22-dehydrocholesterol, and desmosterol. There were significant differences (P < 0.05) between the NZGLM and TBM for 12 of the 20 sterols measured. Six sterols showed significant site differences for the NZGLM, and 10 for the TBM. The differences in the FA and sterol composition between the two species may be due to the diet of the NZGLM being more diatom-derived and the diet of the TBM having a greater dinoflagellate component.

The lipid class, fatty acid, and sterol composition of eight species of ichthyotoxic marine gymnodinioid dinoflagellate (Karenia, Karlodinium, and Takayama) species was examined. The major lipid class in all species was phospholipid (78%-95%), with low levels of triacylglycerol (TAG; 0%-16%) and free fatty acid (FFA; 1%-11%). The common dinoflagellate polyunsaturated fatty acids (PUFA), octadecapentaenoic acid (OPA 18:5x3), and docosahexaenoic acid (DHA 22:6x3), were present in all species in varying amounts (14%-35% and 8%-23%, respectively). The verylong-chain PUFA (VLC-PUFA) 28:7x6 and 28:8x3 were present at low levels (<1%), and the ratio of these fatty acids may be a useful chemotaxonomic marker at the species level. The typical dinoflagellate sterol dinosterol was absent from all species tested. A predominance of the 4-methyl and 4-desmethyl D 8 (14) sterols in all dinoflagellate species included 23-methyl-27-norergosta-8(14),22-dien-3b-ol (Karenia papilionacea A. J. Haywood et Steid, 59%-66%); 27-nor-(24R)-4a-methyl-5a-ergosta-8(14),22-dien-3b-ol, brevesterol, (Takayama tasmanica de Salas, Bolch et Hallegraeff 84%, Takayama helix de Salas, Bolch, Botes et Hallegraeff 71%, Karenia brevis (C. C. Davis) G. Hansen et Moestrup 45%, Karlodinium KDSB01 40%, Karenia mikimotoi (Miyake et Kominami ex Oda) G. Hansen et Moestrup 38%); and (24R)-4a-methyl-5a-ergosta-8(14),22-dien-3b-ol, gymnodinosterol, (K. mikimotoi 48%, Karenia umbella de Salas, Bolch et Hallegraeff 59%, Karlodinium veneficum (D. L. Ballant.) J.Larsen 71%-83%). In Takayama species, five steroid ketones were identified, including for the first time the 3-keto form of brevesterol and gymnodinosterol. These results indicate a biochemical link between sterol and steroid ketone biosynthesis, suggesting that selected dinoflagellates can make a significant contribution to ketones in marine sediments. The presence of steroid ketones, specific sterols, and fatty acids, and the ratio of VLC-PUFA may prove to be a useful chemotaxonomic tool for distinguishing between morphologically similar species. The relative levels of the PUFA, OPA, and DHA, coupled with the potential inhibitory action of D 8(14) sterols, may provide an insight into the ichthyotoxicity of these bloom-forming dinoflagellates.

We examined the lipid class and fatty-acid composition of the southern rock lobster, Jasus edwardsii, phyllosomas larvae and puerulus stage to improve understanding of their nutrition in relation to aquaculture. Lipid is critical in the nutrition of larval crustaceans, including lobsters. Specimens were from Tasmanian waters, Australia, and North Island, New Zealand, waters. Analyses were by TLC-FID and capillary GC and GC-MS. Phyllosomas larvae and nektonic pueruli were low in storage lipid (triacylglycerol), and phospholipid was the major lipid class. Sterol, mainly cholesterol, was the next most abundant class. The ratio of the essential omega-3 fatty acid eicosapentaenoic acid (EPA) to the omega-6 fatty acid arachidonic acid (AA) was lower in newly hatched phyllosomas (1.2-1.3) than in other phyllosomas (stages III-XI; 2.8-6.7) and pueruli (3.8). Ratios of the omega-3 fatty acid DHA (docosahexaenoic acid) to EPA were also lower in newly hatched phyllosomas (0.5) than in later-stage phyllosomas (1.5-2.1) and pueruli (1.2). We have followed up these compositional data by successfully enriching the live diet (Artemia) of early phyllosomas with AA, EPA and DHA. This dietary manipulation has achieved ratios of these key polyunsaturated fatty acids similar to those of wild phyllosomas. These findings will be of significance to the future of rock-lobster aquaculture.
