Objectives: A correlation between vitamin D deficiency and primary Sjögren's Syndrome (pSS) has already been described. The limited data have been reported regarding the pathological relevance of vitamin D in primary Sjögren's syndrome. In this study, the peripheral blood mononuclear cells were cocultured with 1,25-dihydroxyvitaminD3 to determine the modulatory effect of vitamin D3 on T and B lymphocyte phenotypes in pSS.Method: Venous blood samples were collected from 11 in treatment phase and 9 drug-naive pSS patients. Peripheral blood mononuclear cells (PBMC) were isolated and separately cultured in the presence and absence of 1,25-dihydroxyvitaminD3 (10mM) for 5 days of culture period. Lymphocyte proliferation was analyzed for CFSE signaling via flow cytometry. CD3+CD4+ cells were analyzed for intracellular IFN- and IL-17 expressions. CD19+IgD cells were analyzed for CD38 and CD27 expressions to evaluate naïve and memory B cell subsets. Culture supernatants were analyzed for the IFN-, IL-17 and IL-10 cytokine secretions via flow cytometry.Results: 1,25-dihydroxyvitaminD3 significantly decreased Th lymphocyte proliferative responses in drug-naïve (p<0.005) and treated pSS patients (p<0.05), and B lymphocyte proliferation in drug-naïve pSS PBMC cultures (p<0.01) compared to mononuclear cell cultures alone. 1,25-dihydroxyvitamin D3 significantly decreased IFN- and IL-17 secreting Th cells in both drug naïve (p<0.005 and p<0.01, respectively) and treated subjects (p<0.05 and p<0.05, respectively) by increasing FoxP3 expressing CD4+CD25+ Treg cell frequency. Plasma B lymphocytes significantly reduced in the presence of 1,25-dihydroxyvitaminD3 in drug naïve pSS (p<0.001) and treated patients (p<0.05) mononuclear cell cultures compared to PBMC cultures alone. Total memory B cell subsets significantly increased with 1,25-dihydroxyvitaminD3 in drug naïve pSS when compared with PBMC cultures alone (p<0.005). IFN- and IL-17 cytokine levels in culture supernatants significantly reduced (p<0.05 and p<0.01, respectively) in drug naïve pSS patients' PBMC cultures with 1,25-dihydroxyvitaminD3, and IL-10 levels significantly enhanced in both drug-naïve (p<0.01) and treated pSS patients' PBMC cultures (p<0.01) in the presence of 1,25-dihydroxyvitaminD3. Conclusion:In conclusion, 1,25-dihydroxyvitaminD3 regulated immune responses in both treated and drug-naïve pSS patients, but have a more pronounced modulatory effect on mononuclear cell responses in drug-naive pSS patients.
Prostat, rahim ağzı, kolorektal ve karaciğer kanserlerinin tedavileri sıklıkla yaşam kalitesinin düşmesine neden olan cerrahi ve/veya radyoterapi tedavilerini içermektedir. Kanser tedavisinde kullanılmak için potansiyel doğal antikanser kaynakların araştırılması dünyada devam vermektedir. Bu çalışma, Türkiye'den elde edilen okaliptüs balının farklı konsantrasyonlarının seçilen kanser hücre hatları üzerindeki sitotoksisitesini araştırmayı amaçlamıştır. Bu amaçla, PC3 insan prostat kanseri hücre hattı, HeLa rahim ağzı kanseri hücre hattı, CaCo2 kolorektal kanseri hücre hattı ve HuH7 karaciğer kanseri hücre hatları kullanılmıştır. Hücre hatlarına 48 ve 72 saat boyunca çeşitli konsantrasyonlarda (%0.78 -%50 arası) okaliptüs balı uygulamasını takiben, sitotoksisite tayini için, hücre canlılık testi (MTT) yapılmış ve okaliptüs balının her bir konsantrasyonunda kanser hücrelerinin yüzde canlılık oranları belirlenmiştir. Okaliptus balı uygulaması ile hücre hatlarının canlılıklarını %80 ve daha fazla kaybettikleri konsantrasyonlara ve bu konsantrasyonlarda hücrelerin canlılık oranlarına bakıldığında, %6.25 okaliptus balı konsantrasyonunda HeLa hücre hattının 48 ve 72 saatlerinde sırasıyla %7.6 ve %7.7, CaCO2 hücre hattının 72 saatinde %20.7; %12.5 okaliptus balı konsantrasyonunda HuH7 hücre hattının 48 ve 72 saatlerinde sırasıyla % 5.3 ve % 5.1, PC3 hücre hattının 72 saatinde %5.2, CaCO2 hücre hattının 48 saatinde %20.4; %25 okaliptus balı konsantrasyonunda PC3 hücre hattının 48 saatinde %7.7 canlılık oranları gösterdikleri görülmüştür.
Objectives: In this study, we aimed to investigate the differentiation potential of dental follicle mesenchymal stem cells (MSCs) in the synovial fluid (SF) niche of early-onset or end-stage rheumatoid arthritis (RA). Patients and methods: Between May 2020 and January 2021, six patients (1 male, 5 females; mean age: 57.5±11.2 years; range, 49 to 65 years) who were diagnosed with RA with the indication of SF aspiration were included in the study. The third passage dental follicle stem cells (DFSCs) were cocultured with fresh SF samples of end-stage or early-onset RA patients in micromass culture system for 21 days. SF samples were analyzed for secreted cytokines. Chondrogenic markers (CD49e, CD49f) were analyzed in DFSCs, gene expression analysis was performed for the expressions of Col I, Col II, Aggrecan and Sox-9, and histochemical analysis was performed by staining three-dimensional pellets with anti-collagen II antibody. The neutralization assay was performed with anti-interleukin (IL)-6, anti-interferon-gamma (IFN-g), and anti-IL-1beta(b). Results: The high levels of IL-1b and IL-6 were observed in end-stage RA patients’ SF samples compared to the early-onset patients (p<0.05). The CD49e and CD49f expressions in DFSCs were significantly higher in the SF samples of end-stage RA patients (p<0.05). Also, the Col II, Sox-9 and Aggrecan messenger ribonucleic acid (mRNA) expressions increased in the DFSCs, when cultured with end-stage RA patients’ SF samples (p<0.01). Collagen-II expression in histochemical analysis of micromass pellets was higher in the DFSCs cultured with end-stage RA patients’ SF samples. The neutralization of IL-6 significantly decreased the CD49e and CD49f expressions (p<0.05). Conclusion: The high levels of IL-6 in SF niche of end-stage RA patients were found to differentiate DFSCs toward chondrogenesis. Based on these findings, DFSCs can be used as a new cell-based treatment in RA patients for the cartilage damage.
Bor, biyolojik açıdan önemli bir elementtir ve doğada tek başına bulunmayıp, diğer elementlerle bileşikler halinde yer alır. Özellikle oksijen içeren molekülleri tercih eder. "Autoinducer 2" gibi hücresel haberleşmede görevli olan molekülden, bortezomib gibi anti-kanser ajanı olarak kullanılan moleküle kadar, geniş yelpazede, içerisinde bor bulunan çeşitli biyove sentetik bor içeren bileşikler son derece önemlidir. Bunların yanında son yıllarda, yeni bir ilaç sınıfı olarak da tanımlanmaya aday, "bor içeren biyoaktif bileşikler" dikkat çekicidir. Bu bileşikler, özellikle anti-mikrobiyal direncin alarm verdiği şimdiki zaman diliminde ve yakın gelecekte, insan sağlığı açısından başvurabileceğimiz moleküler silahlardan biri olmaya namzettir. Birçok bor içeren bileşik elde edilmiş ve bu bileşikler antibakteriyel etkileri açısından birçok bilim insanı tarafından belli bir seviyeye kadar araştırılmıştır. Çalışmalar sonucunda, bor içeren bileşiklerin bazılarının, medikal alanda antibiyotik olarak kullanılmaları önerilmiştir. Bazı bakterilerin spesifik koşullar altında, doğal ortamlarında yüksek seviyede bor içeren metabolitler sentezlediği bilinmektedir. Aplasmomisin, boromisin, tartrolon ve borofisin doğal kaynaklardan elde edilen bor içeren antibiyotiklerdir. Sentetik olarak üretilen, bor içeren biyoaktif bileşikler olan bortezomib, tavaborol, vaborbaktam, benzaksoborol, akoziborol, iksazomib ve krizaborol ise bazı hastalıkların tedavisinde kullanılmaktadır. Bor, birçok canlı grubunda çeşitli fonksiyonları olan bir elementtir. Aynı zamanda ilaç dizaynı açısından da özel bir potansiyele sahiptir. Bu nedenle, bu makalede, bor içeren bazı biyoaktif bileşiklerin potansiyel kullanımlarını değerlendirmek, etki mekanizmalarını anlamak ve moleküllerin yapısal çeşitliliğini incelemek hedeflenmiştir.
Öz Bakterilerde gen fonksiyonlarının keşfedilmesi ve fenotipik çalışmalarının yapılabilmesi için mutant suşların oluşturulması gerekmektedir. Bazı durumlarda, birden fazla gen açısından mutant olan suşlar oluşturulmalıdır çünkü bazı spesifik aktiviteler birden fazla gen tarafından kodlanmaktadır. P1 fajı, Escherichia coli'de, ikili, üçlü veya çoklu mutantların oluşturulmasında başvurulan, genelleştirilmiş bir transdüksiyon fajıdır. Bu faj, gelişimi sırasında, E. coli kromozomunun parçalarını (her bir partikülde genomun yaklaşık % 2'si olmak üzere), rastgele bir şekilde faj kafasına paketleyebilmektedir. E. coli DNA'sını içeren fajlar, içerdikleri DNA parçalarını, alıcı bakteri hücrelerine enjekte edebilmektedir. Verici ve alıcı DNA bölgeleri arasındaki benzerlik ve seçici bir belirteç yardımıyla, transdüktanlar'ın elde edilmesi mümkün olmaktadır. Transdüksiyon için tercihen, P1 fajının, virülan bir türevi (P1vir) kullanılmaktadır, çünkü bu form, bakterinin genomuna entegre olamamaktadır. Fajın bu özelliğinin kullanılmasıyla geliştirilen tekniklerden birisi, P1 transdüksiyon tekniği olarak adlandırılmaktadır. Rekombinasyon teknolojisinde kullanılan bu teknik, E. coli genomuna ait DNA parçalarının, farklı suşlar arasında taşınmasını sağlamaktadır. Bu çalışmada, optimize ettiğimiz P1 transdüksiyon metodolojisinin, deneysel akış ve yeterli detaylarla birlikte, Türk bilim camiasına, ana dilimizde sunulması amaçlanmıştır. Bunun için, KEIO koleksiyonundan elde edilen dört adet tekli gen mutantları kullanılmıştır. İlk olarak, kanamisin (Kan) duyarlı bir alıcı suş oluşturmak için, her bir mutant suştan, Kan gen kasetleri çıkarılmıştır. Kan R kasetlerine sahip diğer dört adet KEIO mutant suşu, donör suşlar olarak kullanılmış ve P1 fajı bu suşlar ile kültür edilerek, lizatlar elde edilmiştir. Bu lizatlar ve alıcı suşlar daha sonra, lizatlardaki fajların içerdikleri DNA parçalarını, alıcı suş genomuna entegre edebilmesi için, transdüksiyona tabii tutulmuştur. Kanamisin ile seleksiyon sonucu, yeni mutant suşlar olan ikili mutantlar elde edilmiştir. Sonuçta, isimleri tsgAugpA, yhdTompA, ugpAmdtG ve aroGmalF olan, dört adet ikili mutant oluşturulmuştur. Deneysel akıştaki ilgili protokoller ve sonuçlara ait detaylar sunulmuş ve tartışılmıştır.
