Gastric cancer is classified into intestinal and diffuse types, the latter including a highly malignant form, linitis plastica. A two-stage genome-wide association study (stage 1: 85,576 SNPs on 188 cases and 752 references; stage 2: 2,753 SNPs on 749 cases and 750 controls) in Japan identified a significant association between an intronic SNP (rs2976392) in PSCA (prostate stem cell antigen) and diffuse-type gastric cancer (allele-specific odds ratio (OR) = 1.62, 95% CI = 1.38-1.89, P = 1.11 x 10(-9)). The association was far less significant in intestinal-type gastric cancer. We found that PSCA is expressed in differentiating gastric epithelial cells, has a cell-proliferation inhibition activity in vitro and is frequently silenced in gastric cancer. Substitution of the C allele with the risk allele T at a SNP in the first exon (rs2294008, which has r(2) = 0.995, D' = 0.999 with rs2976392) reduces transcriptional activity of an upstream fragment of the gene. The same risk allele was also significantly associated with diffuse-type gastric cancer in 457 cases and 390 controls in Korea (allele-specific OR = 1.90, 95% CI = 1.56-2.33, P = 8.01 x 10(-11)). The polymorphism of the PSCA gene, which is possibly involved in regulating gastric epithelial-cell proliferation, influences susceptibility to diffuse-type gastric cancer.

The syntheses of (+ )-5 3H-eudesma-3,11 -diene [1], (-)-5 ß -eudesmane-4ß, 11-diol [2], and (+ )-5ß -1 5 -4 , 11-diol [15] were carried out by an unambiguous procedure starting from -santonin. The diene 1 was identical with a natural product which was isolated previously as a termite defense substance. Although structure 2 was recently proposed for a new eudesmane-1,4-diol [A] isolated from Pluchea arguta, synthetic 2 and its C-4 epimer, 15, were not identical with this natural eudesmane-4,11-diol when their physical and spectroscopic parameters were compared. The structure of this natural eudesmane-4,11-diol has been revised from 2 to 3 (7 ßH-eudesmane-4 , 11-diol) as a result of the analysis of the Hand ,3Cnmr spectra of related natural and unnatural eudesmane derivatives.

[Purpose] The aim of this study was to determine the reference values for the chair stand test (CST) in healthy older Japanese people. [Methods] Relevant research articles for the 5-repetition chair stand test (CS-5) and the 30-second chair stand test (CS-30) were identified by electronic database and manual searching. Research articles involving healthy Japanese people aged 60 years and older were included in a meta-analysis. Weighted means of the CS-5 and CS-30 were estimated by the random effect model as the reference values for the CST. Further, the effects of age and sex on the reference values were analyzed by a meta-regression analysis. [Results] Seven articles (21 data) and three articles (14 data) were included in the meta-analyses for the CS-5 and CS-30, respectively. The reference value for the CS-5 was estimated as 8.50 sec [95% confidence interval (CI): 7.93–9.07]; age and sex were not associated with this reference value. The reference value for the CS-30 was estimated as 17.26 times [95%CI: 15.98–18.55], and age was significantly associated with this value. [Conclusion] When the CS-5 and CS-30 are used to evaluate elderly Japanese people, the reference values for the CS-5 and CS-30 determined in this study would be useful indices.
