A Streptomyces strain was isolated from soil and the sequence of 1471 nucleotides of its 16S rDNA showed 99% identity to Streptomyces sp. HV10. This newly isolated Streptomyces strain produced an extracellular polysaccharide (EPS) composed mainly of glucose and mannose in a ratio of 1:4.1, as was characterized by Fourier transform infrared spectroscopy (FTIR), HPLC and 1H-NMR. The antioxidant activities of the partially purified MOE6-EPS were determined by measuring the hydroxyl free radical scavenging activity and the scavenging of 2,2-diphenyl-2-picryl-hydrazyl (DPPH) radicals. In addition, the partially purified MOE6-EPS showed high ferrous ion (Fe2+) chelation activity which is another antioxidant activity. Interestingly, 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assays that were colorimetric assays for NAD(P)H-dependent cellular oxidoreductases and a proxy of the number of viable cells, showed that the partially purified MOE6-EPS inhibited the proliferation of the human breast cancer cells (MDA-MB-231). The scratch wound assay showed that MOE6-EPS reduced the migration of mouse breast cancer cells (4T1). This study reports the production of EPS from Streptomyces species with promising antioxidant, metal chelating and mammalian cell inhibitory activities.

Abstract. Elkhateeb WA, Daba GM, Elnahas MO, Thomas PW, Emam M. 2020. Metabolic profile and skin-related bioactivities of Cerioporus squamosus hydromethanolic extract. Biodiversitas 21: 4732-4740.  Being a functional food capable of showing nutritional as well as medicinal properties have great attention. Mushrooms have been proven as leading targets in this field. For this purpose, the edible mushroom Cerioporus squamosus was investigated in this study to evaluate the in vitro skin-related bioactivities of its hydromethanolic extract in terms of enhancing wound healing, and human skin cancer suppression capabilities. Treatment of fibroblast cells (BJ-1) with the hydromethanolic extract of this mushroom at 50 µg/mL enhanced cell migration rates by 71.7% after 24 h of exposure to the extract. Moreover, the same extract exhibited a promising impact on human skin cancer using an epidermoid carcinoma cell line (A431). The gradual increase in C. squamosus hydromethanolic extract concentration caused gradual decrease in the A431 cell viability and proliferation. Maximum effect on reducing the cell viability was obtained at a concentration of 100 µg/mL, where cell viability was 3.7%, and recorded IC50 was 52.6 µg/mL. The metabolic profile of the extract was analyzed by GC-MS, which was performed on its silylated metabolites. Nineteen compounds were detected including sugar alcohols, amino acids, fatty and organic acids. Promising results of this mushroom extract encourage conducting further steps towards using this mushroom as a functional food showing promising bioactivities.

Polypore mushrooms have been used medicinally for thousands of years. Agarikon (Fomitopsis officinalis) is a medicinal polypore mushroom containing a host of pharmacologically active compounds that beneficially affect human health. Agarikon is known for its capability of producing various biologically active compounds with medical applications such as antiviral, antibacterial, anticancer, and anti-inflammatory agents. This review describes the importance of medicinal mushrooms, with a specific focus on Agarikon as an example of a globally commercialized medicinal mushroom.

Toxic metal contamination has serious effects on human health. Crude oil that may contain toxic metals and oil spills can further contaminate the environment and lead to increased exposure. This being the case, we chose to study the bio-production of inexpensive, environmentally safe materials for remediation. Streptomyces sp. MOE6 is a Gram-positive, filamentous bacterium from soil that produces an extracellular polysaccharide (MOE6-EPS). A one-factor-at-a-time experiments showed that the maximum production of MOE6-EPS was achieved at 35 °C, pH 6, after nine days of incubation with soluble starch and yeast extract as carbon sources and the latter as the nitrogen source. We demonstrated that MOE6-EPS has the capacity to remove toxic metals such as Co(II), Cr(VI), Cu(II) and U(VI) and from solution either by chelation and/or reduction. Additionally, the bacterium was found to produce siderophores, which contribute to the removal of metals, specifically Fe(III). Additionally, purified MOE6-EPS showed emulsifying activities against various hydrophobic substances, including olive oil, corn oil, benzene, toluene and engine oil. These results indicate that EPS from Streptomyces sp. MOE6 may be useful to sequester toxic metals and oil in contaminated environments.

The genus Trichoderma is multicultural soil-borne fungi found in different ecosystems. They are highly successful colonizers of their habitats. Genus Trichoderma is capable of dealing with various environments such as compost, agricultural soils, rhizosphere, and waste material. Therefore, different strains of Trichoderma have been applied in agriculture, bioremediation, waste management, and biotechnology. Many Trichoderma species act as biological control agents and plant growth promoters. Additionally, the genus Trichoderma is a new fungal source for the production of cyclosporin A as well as various hydrolytic enzymes with industrial importance.

Metacordyceps neogunnii is an edible mushroom that has been previously known as Cordyceps gunnii. Recently, it has been proven that Metacordyceps neogunnii differs from similar species by having longer asci and wider ascospores and is phylogenetically distinct to related species. Few studies are available to describe bioactivities exerted by Metacordyceps neogunnii extracts which is insufficient to evaluate the potency of this mushroom. Therefore, the current work was conducted to evaluate the in vitro α-Amylase inhibitory ability, wound healing, and anti-human pancreatic cancer activities of Metacordyceps neogunnii 80% methanolic extract. M. neogunnii extract showed no effect on human cancer pancreatic cell line (Paca2). However, the same extract exhibited slight wound healing effect. On the other hand, M. neogunnii extract exhibited good α-amylase inhibitory activity that reached 69.3±0.78%, and IC50 of 32.38±4.0.
