The effects of selection on genome variation were investigated and visualized in tomato using a high-density single nucleotide polymorphism (SNP) array. 7,720 SNPs were genotyped on a collection of 426 tomato accessions (410 inbreds and 16 hybrids) and over 97% of the markers were polymorphic in the entire collection. Principal component analysis (PCA) and pairwise estimates of F st supported that the inbred accessions represented seven sub-populations including processing, large-fruited fresh market, large-fruited vintage, cultivated cherry, landrace, wild cherry, and S. pimpinellifolium. Further divisions were found within both the contemporary processing and fresh market sub-populations. These sub-populations showed higher levels of genetic diversity relative to the vintage sub-population. The array provided a large number of polymorphic SNP markers across each sub-population, ranging from 3,159 in the vintage accessions to 6,234 in the cultivated cherry accessions. Visualization of minor allele frequency revealed regions of the genome that distinguished three representative sub-populations of cultivated tomato (processing, fresh market, and vintage), particularly on chromosomes 2, 4, 5, 6, and 11. The PCA loadings and F st outlier analysis between these three sub-populations identified a large number of candidate loci under positive selection on chromosomes 4, 5, and 11. The extent of linkage disequilibrium (LD) was examined within each chromosome for these sub-populations. LD decay varied between chromosomes and sub-populations, with large differences reflective of breeding history. For example, on chromosome 11, decay occurred over 0.8 cM for processing accessions and over 19.7 cM for fresh market accessions. The observed SNP variation and LD decay suggest that different patterns of genetic variation in cultivated tomato are due to introgression from wild species and selection for market specialization.

Settling of the potato aphid,Macrosiphum euphorbiae, on feeding membranes was deterred by methanolic leaf rinses ofLycopersicon pennellii, or of its F1 with tomato,L. esculentum. The active compounds in theL. pennellii rinsates were identified as 2,3,4-tri-O-acylglucoses bearing short to medium chain length fatty acids. These compounds are localized in the glandular exudate of the type IV trichomes and may accumulate to levels in excess of 400 μg/cm(2). In choice assays, purified glucose esters fromL. pennellii reduced aphid settling at concentrations as low as 25 μg/cm(2); at concentrations of 150 μg/cm(2) or more, all aphids avoided treated areas. Glucose esters were also active in deterring aphid settling in no-choice assays. At 100 μg/ cm(2), these esters resulted in increased levels of mortality after 48 hr.

Acylsugars are secondary metabolites exuded from type IV glandular trichomes that provide broad-spectrum insect suppression for Solanum pennellii Correll, a wild relative of cultivated tomato. Acylsugars produced by different S. pennellii accessions vary by sugar moieties (glucose or sucrose) and fatty acid side chains (lengths and branching patterns). Our objective was to determine which acylsugar compositions more effectively suppressed oviposition of the whitefly Bemisia tabaci (Gennadius) (Middle East—Asia Minor 1 Group), tobacco thrips, Frankliniella fusca (Hinds), and western flower thrips, Frankliniella occidentalis (Pergande). We extracted and characterized acylsugars from four S. pennellii accessions with different compositions, as well as from an acylsugar-producing tomato breeding line. We also fractionated the acylsugars of one S. pennellii accession to examine the effects of its components. Effects of acylsugars on oviposition were evaluated by administering a range of doses to oviposition sites of adult whiteflies and thrips in non-choice and choice bioassays, respectively. The acylsugars from S. pennellii accessions and the tomato breeding line demonstrated differential functionality in their ability to alter the distribution of whitefly oviposition and suppress oviposition on acylsugar treated substrates. Tobacco thrips were sensitive to all compositions while western flower thrips and whiteflies were more sensitive to acylsugars from a subset of S. pennellii accessions. It follows that acylsugars could thus mediate plant-enemy interactions in such a way as to affect evolution of host specialization, resistance specificity, and potentially host differentiation or local adaptation. The acylsugars from S. pennellii LA1376 were separated by polarity into two fractions that differed sharply for their sugar moieties and fatty acid side chains. These fractions had different efficacies, with neither having activity approaching that of the original exudate. When these two fractions were recombined, the effect on both whiteflies and thrips exceeded the sum of the two fractions’ effects, and was similar to that of the original exudate. These results suggest that increasing diversity of components within a mixture may increase suppression through synergistic interactions. This study demonstrates the potential for composition-specific deployment of acylsugars for herbivore oviposition suppression, either through in planta production by tomato lines, or as biocides applied by a foliar spray.

A family of biodegradable, biocompatible, water soluble cationic polymer precursor, arginine‐based unsaturated poly (ester amide) (Arg‐UPEA), is reported. Its incorporation into conventional Pluronic diacrylate (Pluronic‐DA) to form hybrid hydrogels for a significant improvement of the biological performance of current synthetic hydrogels is shown. The gel fraction (Gf), equilibrium swelling ratio (Qeq), compressive modulus, and interior morphology of the hybrid hydrogels as well as their interactions with human fibroblasts and bovine endothelial cells are fully investigated. It is found that the incorporation of Arg‐UPEA into Pluronic‐DA hydrogels significantly changes their Qeq, mechanical strength, and interior morphology. The structure–property relationship of the newly fabricated hybrid hydrogels is studied in terms of the chemical structure of the Arg‐UPEA precursor, i.e., the number of methylene groups in the Arg‐UPEA repeating unit. The results indicate that increasing methylene groups in the Arg‐UPEA repeating unit increases Qeq and decreases the compressive modulus of hydrogels. When compared with a pure Pluronic hydrogel, the cationic Arg‐UPEAs/Pluronic hybrid hydrogels greatly improve the attachment and proliferation of human fibroblasts on hydrogel surfaces. A bovine aortic endothelial cells (BAEC) viability test in the interior of the hydrogels shows that the positively charged hybrid hydrogels can significantly improve the viability of the encapsulated endothelial cell over a 2 week study period when compared with a pure Pluronic hydrogel.

Mortality of the potato aphid, Macrosiphum euphorbiae (Thomas), on Lycopersicon pennellii (Corr.) D'Arcy and its F1 hybrid with Lycopersicon esculentum Mill. was significantly greater than that on L. esculentum. Physical entrapment was not the sole mechanism of resistance in L. pennellii since few late instar aphids were found trapped in the sticky glandular exudate of the type IV trichomes; entrapment could, however, affect survival of early instars. Aphid settling on L. pennellii was dramatically less than that on L. esculentum, suggesting that starvation may have contributed to high mortality. Compared to L. esculentum, aphid feeding behavior on L. pennellii and the F1 was characterized by a delay in the time to first probe, a reduction in the number of probes, and a decrease in the total proportion of time spent feeding. Removal of the glandular exudate of the type IV trichomes from L. pennellii resulted in a decrease in preprobe time and an increase in both the number of probes and the percent of time spent probing. Transfer of glandular trichome exudate of L. pennellii to leaflets of L. esculentum resulted in an increase in resistance as measured by these three parameters. Zusammenfassung Saugverhalten der Kartoffelblattlaus beeinflusst durch Drüsentrichome der wilden Tomate Die Absterberate der Kartoffellaus, Macrosiphum euphorbiae Thomas, auf Lycopersicon pennellii (Corr.) D'Arcy, sowie auf der Kreuzung L. esculentum Mill. und L. pennellii, war deutlich grösser als auf L. esculentum. Das mechanische Verfangen der Läuse war nicht der Hauptgrund der Resistenz von L. pennellii. Wenige tote Läuse wurden in dem klebrigen Sekret der Typus IV Trichome gefunden. Auf L. pennellii siedelten sich die Läuse in viel geringerer Zahl an als auf L. esculentum. Dies führte zum Schluss, dass Verhungern eine der Ursachen der hohen Mortalität der Läuse war. Im Vergleich zum Saugverhalten auf L. esculentum war das Saugverhalten auf L. pennellii, wie auch auf F1, durch Folgendes gekennzeichnet 1) Verspätung des ersten Stichversuchs, 2) Verminderung der Stichversuche pro Zeiteinheit und 3) Verminderung des Zeitanteils, der zum Saugen verwendet wurde. Die Entfernung des Sekrets der Typus IV Trichomen auf L. pennellii verursachte 1) eine kürzere Zeitspanne vor dem ersten Stichversuch, 2) eine Vergrösserung der Anzahl Stichversuche pro Zeiteinheit, 3) eine Verlängerung der Saugzeit. Die Uebertragung des Sekretes von L. pennellii auf Blätter von L. esculentum verbesserte deren Resistenz gegen Blattläuse gemessen mit den genannten drei Kriterien.
