Cold induces expression of Cor/Lea genes, in particular of Wcs120 and Wdhn13. The level of the expression at low temperatures was related to the frost tolerance (FT) of wheat cultivars. We studied the expression in five cultivars very different in the FT, at relatively high temperature of 17 and 25°C, respectively. Significant correlations at the 1% level were found at 17°C between the expression level and FT, characterized by correlation coefficients 0.92** and 0.97** for Wcs120 and Wdhn13, respectively. These preliminary results could be used for screening of frost tolerant lines.

Abstract:The sequences of the closely linked barley dehydrin genes dhn4 and dhn7 are both known to vary allelically. Here we associated allelic constitution at dhn4/7 with frost sensitivity across a panel of 30 diverse barley varieties. The combined presence of a 6 bp insertion in exon1 of dhn4 and a 30 bp deletion in exon1 of dhn7 was restricted to six-rowed winter and intermediate varieties characterised by relatively higher frost tolerance (12 genotypes; LT 50 from -14.2°C to -15.6°C). The alternative combination was present with one exception (sixrowed winter variety Alissa) only in spring and two-rowed winter varieties (17 genotypes; LT 50 from -10.0°C to -14.3°C). The genetic linkage between dhn4 and dhn7 identified e.g. in Dictoo and Morex varieties was verified by segregation analysis of F 2 plants from a cross between two genotypes carrying different allelic combination of dhn4 and dhn7 genes (two-rowed spring variety Akcent × six-rowed winter variety Okal). The potential of the former allelic combination as a marker for enhanced frost tolerance was tested in a sample of F 5 derivatives of a cross between the two-rowed winter type variety Monaco (Akcent allele combination) and the six-rowed winter type variety Okal. Plants with the Okal allele combination showed significantly higher frost tolerance than those with the alternative growth habit. The effect of ear type on frost tolerance was insignificant.

Šlechtění odrůd tolerantních vůči suchu je stále důležitější z hlediska zlepšení výnosů a zdravotního stavu rostlin zvláště u hospodářsky významných plodin. Je založeno na hodnocení a účinné selekci tolerantních genotypů. Hodnocení citlivosti rostlin vůči suchu však není jednoduché, vzhledem ke komplexnímu charakteru této vlastnosti. Cílem této práce bylo porovnat stresové reakce pěti různě citlivých genotypů ječmene (Tadmor, Malz, Amulet, Bojos a Jersey) vystavených stresu suchem pomocí molekulárně biologické metody založené na hodnocení aktivity dvou dehydrinových genů (Dhn1 a Dhn4) a ověřit účinnost této metody nejen v laboratorních podmín-kách, ale i v nádobovém pokusu, kdy byla regulována pouze zálivka. Přesnější hodnocení bylo prováděno na rostlinách pěstovaných v kultivačním boxu hydroponicky v podmínkách fyziologického sucha (-0,3 MPa). Transkripční aktivita obou genů byla hodnocena v prů-běhu 14 dnů ve vzorcích z listových tkání v závislosti na citlivosti vůči stresovým podmínkám. U tolerantnějších genotypů (Tadmor a Malz) byla zaznamenána nižší ztráta vody z pletiv a vyšší exprese obou genů. Srovnatelné výsledky pro jednotlivé odrůdy byly získány i v ná-dobovém pokusu. Opět se projevila závislost mezi aktivitou těchto genů a citlivostí odrůd k suchu, zvláště ve fázi metání a plnění zrna. U testovaného souboru odrůd byla dále hodnocena aktivita obou genů v závislosti na působení exogenně aplikovaného fytohormonu ABA. Ukázalo se, že tímto fytohormonem byla aktivována exprese pouze genu Dhn4 a výrazné zvýšení jeho exprese bylo pozorováno pouze u odrůdy Malz. Srovnáním výsledků ze všech tří experimentů jsme ověřili nejen vhodnost použití této metody jako nástroje pro stanovení tolerance ječmene vůči suchu, ale u tolerantnějších odrůd Tadmor a Malz jsme zaznamenali dva odlišné mechanismy aktivace stresových obranných reakcí, které by mohly být způsobeny rozdílnou mírou zapojení fytohormonu ABA v jejich regulaci. Breeding of drought-tolerant varieties, which is steadily gaining in importance for improving crop yields and health condition of economically important crops, is based on evaluation and efficient selection of tolerant genotypes. The assessment of plant sensitivity to drought is not simple owing to the complex character of this trait. We compared the stress responses of five different barley genotypes with different sensitivity (Tadmor, Malz, Amulet, Bojos and Jersey) exposed to drought stress by using a molecular biological method based on evaluating the activity of two dehydrin genes, Dhn1 and Dhn4. The efficiency of the method was verified in laboratory conditions and also in a pot experiment in which only watering was regulated. More detailed evaluation was done on plants grown hydroponically in a cultivation box under conditions of physiological drought (-0.3 MPa). Transcription activity of both genes, as dependent on stress sensitivity, was evaluated over 14 days in leaf tissue samples.
