The glycosylation of the 4-methoxypyrazolo[3,4-d]pyrirnidine (5) anion with I-halo-2-deoxyribose 6 in MeCN/TDA-I gives N*-deoxynucleoside 9 (29 %)together with N'-isomer 7 (48%) and its anomer 8 (6%) [7). The G( -o-anomer 8 is not formed and the yield of the B-D-anomer 7 increased to 62 % when dimethoxyethane is used as solvent and [18]crown-6 as catalyst. Employing l-halo-2,3-dideoxyribose 10 instead of halogenose 6, the 2',3'-dideoxynucleosides 12 and 14 were formed which were desilylated (+ lb and 2b) and converted into the ddI and ddA derivatives 3b (c7z81dd), 15b (C72*A&), and 17 (c~z'A',,). Contrary to 7-deazdpurine nucleotides, the triphosphates of 3b and 15b showed no appreciable activity against HIV-reverse transcriptase.
Blood-protein adducts are used as dosimeter for modifications of macromolecules in the target organs where the disease develops. The functional groups of cysteine, tyrosine, serine, lysine, tryptophan, histidine and N-terminal amino acids are potential reaction sites for isocyanates. Especially the N-terminal amino acids, valine and aspartic acid of hemoglobin and albumin, respectively, are reactive towards electrophilic xenobiotics. To develop methods for the quantification of such blood-protein adducts, we treated 4-chlorophenyl isocyanate (I) with the tripeptide L-valyl-glycyl-glycine (Za) and with single amino acids yielding N-[(4-chlorophenyl)carbamoyl]valyl-glycyl-glycine (3a). N-[(4-~hlorophenyl)carbamoyl]valine (3b). N-[(4-~hlorophenyl)carbamoyl]aspartic acid (3c), N -[(4-chlorophenyl)carbamoyl]glutamic acid (3d), N-acetyl-S-[(4-chlorophenyl)carbamoyl]cysteine (3e), and Nacetyl-O-[(4-~hlorophenyl)carbamoyl]serine (30, Na-acetyl-N'-[(4-chlorophenyl)carbamoyl]Iysine (3g).For several chemicals, it was shown that blood-protein adducts are good dosimeters of exposure and dosimeters for the target dose. The hydrolysis of the N-terminal adducts of isocyanates release hydantoins which can be separated from the rest of the protein and analyzed using GC/MS or HPLC. This was achieved with 3a. The released hydantoin could be analyzed using GC/MS. We propose to analyze the N-terminal adducts of isocyanates with blood protein to distinguish between arenamine and arylisocyanate exposure.
Die [2 + 21-Cycloaddition der Thioketene 1 af an die Isocyanate 2a-f liefert als Hauptprodukt 4-Thioxo-2-azetidinone 3, die sich zu 4-Imino-2-thietanonen 5 isomerisieren konnen. In Konkurrenzreaktionen werden 2,4-Azetidindione 8, N-Sulfonylamide 9 und 3H-1,2,4-Dithiazole 15 gebildet. Mit Chlorsulfonylisocyanat (20b) reagieren die Thioketene 1 zu N-unsubstituierten 4-Thioxo-2-azetidinonen 22. Je nach Thioketen 1 und Reaktions-Tlirung resultieren auch die Verbindungen 23-26. Die Konstitution von 15b und 23a wurde durch Rontgenstrukturanalyse bestimmt.Die Cycloaddition von zwei Heterocumulen-Molekulen bietet einen bequemen und variablen Zugang zu Vier-und Sechsring-Heterocyclen*). Unter den mittlerweile in groDer Vielfalt zuganglichen Thioketenen3) ist bisher nur der Bis-(trifluormethy1)-Vertreter mit anderen Heterocumulenen umgesetzt worden4). Wir berichten jetzt uber die Reaktion von Dialkylthioketenen rnit Isocyanaten ". Darstellung von 4-Thioxo-2-azetidinonen 3Hauptprodukte der Umsetzung von Dialkylthioketenen 1 rnit Isocyanaten 2 sind in der Regel 1: 1-Addukte. Die Konstitution von 4-Thioxo-2-azetidinonen 3 wird durch die IR-und 13C-NMR-spektroskopischen Daten (Tab. 4) nahegelegt und konnte fur 3i durch eine Rontgenstrukturanalyse bewiesen werden 5! Die Ausbeuten an 3 spiegeln die Reaktivitat von 1 und 2 wider (Tab. 1). So erfordert zunehmender sterischer Anspruch der Substituenten R', R' in 1 hohere Reaktionstemperaturen und fiihrt so verstarkt zu Konkurrenzreaktionen (s. unten); die sterisch extrem gehinderten Thioketene 1 g, h bilden keine Addukte 3 mehr. Die Bereitschaft der Isocyanate 2 zur Reaktion mit 1 nimmt in der Reihenfolgea b und korreliert so mit der Elektrophilie der Isocyanate. Damit reagiert in der Cycloaddition von 1 mit 2 das Isocyanat als typisches Heterocumulen, also elektrophil, wahrend die Thioketene hier als Nucleophil auftreten. In competing reactions, 2,4-azetidinediones 8, N-sulfonylamides 9, and 3H-1,2,4-dithiazoles 15 are formed. Thioketenes 1 react with chlorosulfonyl isocyanate (2Ob) to give N-unsubstituted 4-thioxo-2-azetidinones 22. Depending on the thioketene 1 and the reaction conditions, compounds 23-26 also result. The constitutions of 15b and 23a were determined by X-ray structural analyses. erhoht. Die basenkatalysierte Alkoholyse der NS‐Bindung in N‐Sulfonyl‐thioxoazetidinonen 1 tritt leicht ein; der entstehende Sulfonsäureester alkyliert dann das freigesetzte 2−. Unter verschiedenen Bedingungen durchgeführte Alkylierungsreaktionen an 2 zeigen, daß Ladungs‐ und thermodynamische Kontrolle den Angriff am Stickstoff zu 5 gegenüber der Reaktion am Schwefel zu 4 begünstigen. Neben einfachen Alkyl‐Resten (4a–c, 5a–c) lassen sich auch funktionalisierte Alkyl‐(4d, e, g, m, n, o, 5d, e, g, i, m–o), Aryl‐ (5p) und Acyl‐Gruppen (5q) einführen sowie Additionsreaktionen (5r, s) erreichen. Komplexe Hydride reduzieren die Carbonyl‐Gruppe in 1, 5 zu 8–10, aber den CN+‐Teil im Salz 6a zum β‐Lactam 11. Die Anellierung eines Fünfrings zu 12 gelingt über die 1,3‐dipolare Cycloaddition von Fumarsäure‐dimethylester an das in situ freigesetzte Azomethin‐Ylid 6d.
