The bioguided fractionation of the crude extracts of Lychnophora passerina (Mart. ex DC) Gardn., L. pinaster Mart. and L. trichocarpha Spreng. led to the isolation of four compounds active against the trypomastigote forms of Trypanosoma cruzi: the sesquiterpene lactones goyazensolide (1), eremantholide C (2), and lychnopholide (3) and a new caryophyllene derivative, which was named lychnophoic acid (4).
ACTIVITY -GUIDED ISOLATION OF CONSTITUENTS OF UNONOPSIS LINDMANII -ANNONACEAE, BASED ON THE BRINE SHRIMP LETHALITY BIOASSAY. Extracts obtained from leaves, seeds and bark of Unonopsis lindmanii were evaluated by means of Brine Shrimp Lethality test (BSL). Through bioassay-guided chromatographic fractionation, liriodenine, an oxoaporphine alkaloid, was isolated from the bark extracts as the bioactive compound. ARTIGO INTRODUÇÃOMuitos laboratórios de Produtos Naturais têm inserido dentro de suas rotinas de isolamento, purificação e elucidação estrutural, diversos ensaios biológicos simples, no intuito de selecionar e monitorar o estudo fitoquímico de extratos de plantas na procura de substâncias bioativas 1 . Dentre estes bioensaios, encontra-se a toxicidade sobre Artemia salina (TAS), que se caracteriza por ser de baixo custo, rápido e não exigir téc-nicas assépticas. Inúmeros constituintes bioativos têm sido obtidos de extratos vegetais utilizando este teste na monitoração de estudos fitoquímicos 2,3 . Artemia salina é um microcrustáceo de água salgada que é utilizado como alimento vivo para peixes, sendo seus ovos facilmente encontrados em lojas de aquaristas. A simplicidade do bioensaio TAS favorece sua utilização rotineira, podendo ser desenvolvido no próprio laboratório de fitoquímica 2 . Diversos trabalhos tentam correlacionar a toxicidade sobre Artemia salina com atividades como antifúngica, viruscida e antimicrobiana 4 , parasiticida 5 , tripanossomicida 6 , entre outras. McLaughlin e colaboradores 2,3,7-10 têm utilizado sistematicamente este bioensaio na avaliação prévia de extratos de plantas conhecidas como antitumorais. As frações ou substâncias ativas são posteriormente testadas em diferentes culturas de célu-las tumorais, obtendo-se uma boa correlação. .Várias espécies da família Annonaceae foram estudadas empregando o bioensaio TAS, juntamente com outros ensaios biológicos [7][8][9][10] . Porém, nenhuma espécie do gênero Unonopsis havia sido quimicamente investigada utilizando monitoramento por bioensaio.A literatura contém poucos dados sobre a composição quí-mica do gênero Unonopis, que é um dos 27 gêneros da família Anonaceae. Apenas as espécies U. Stipitata e U. guaterioides foram motivo de publicação, sendo que da primeira foram obtidos alcalóides do tipo bisaporfínico, azafluorenona a oxaporfínicos 11 , enquanto na segunda a presença de policarpol 12 foi detectada por cromatografia em camada delgada. Algumas utilizações empíricas do gênero Unonopsis foram descritas por Schultes, que menciona o uso de plantas desse gênero pelos índios da Floresta Amazônica no tratamento de demência senil. Sixteen extracts, obtained from eight Brazilian plants of Annonaceae family, were screened for their antibacterial activity: Xylopia frutescens, X. aromatica, X. amazonica, X. benthamii, Annona ambotay, A. crassiflora, A. muricata and A. cherimolia. Amongst the investigated extracts, six showed antibacterial activity against at least one of the tested organisms at the concentration of 100 microg/mL. The most active extracts were those prepared from X. frutescens, X. amazonica, and A. ambotay. A phytochemical screening showed the presence of anonaceus acetogenins in some active extracts. Eleven diterpenoids were also tested for comparison purposes. Six were natural products, previously isolated from Xylopia sp. (kaurenoic, frutoic, xylopic, 15beta-hydroxy-kaurenoic and trachylobanic acids plus kaurenol) and five were derivatives of such compounds, obtained by esterification or reduction reactions. Trachylobanic acid showed antibacterial activity against B. subtilis and S. aureus.
