Corn is one of the main grasses used to produce human or animal food. One of the main problems with the use of corn is the presence of mycotoxins, where aflatoxin B 1 is one of the most harmful for human and animal health. Chemical methods such as the addition of antifungals and sequestrants are used to control this contaminant in food; however, these methods can leave dangerous residues. The aim of this research was to evaluate the effectiveness of irradiation with gamma rays on corn for the control of aflatoxin B 1 . For this purpose, three treatments were compared: treatment 1, application of gamma radiation, treatment 2, application of antifungal and treatment 3, combination of gamma radiation and application of antifungal. Corn without exposure to any control of aflatoxin was used as a witness group. Irradiation doses of 2, 6 and 10 kGy were tested, and the dose of 6 kGy was selected as the best since it lowered aflatoxin B 1 more effectively. The corn analyzed in this study was stored during 45 days at 23˚C, and every 15 days. We determined the concentration of aflatoxin B 1 , presence or absence of pathogenic microorganisms and insects, and we evaluated the physicochemical characteristics of the grain. Then, the treatments were compared and treatment 1, only irradiation with gamma rays, proved to be significantly more effective in the reduction of aflatoxin B 1 and the total inhibition of the survival of the insects during the 45 days of storage which allowed better preservation of the protein. than 13%. The cost production of a bag of 40 kg of feed was also estimated in USD 22.56; which is 1.80% greater than the cost corresponding to the conventional process that uses mycotoxin sequestrants.
Essential oils are liquid mixtures of volatile compounds extracted from plants. Their quality is usually controlled via gas chromatography (GC), although with limitations when adulterants are nonvolatile substances. The essential oils of lavender (Lavandula angustifolia Mill.), peppermint (Mentha piperita L.), patchouli (Pogostemon cablin Benth), and their adulterated versions were measured by GC coupled to flame ionization detector (GC‐FID) and Raman spectroscopy. Canola oil, a nonvolatile substance, was used as the adulterant. The adulterated essential oils contained 1%, 3%, 5%, 10%, 15%, and 20% (v/v) of canola oil. Chromatograms of the adulterated essential oils containing 20% (v/v) of canola oil showed decrements in peak areas of the essential oil components, compared with peaks of the pure essential oils. The highest decrements were observed for the adulterated essential oil of patchouli. In general, detection of adulterated essential oils by simple visual inspection of the Raman features was difficult, due to slight differences observed in the spectra. Principal Components Analysis (PCA) allowed achieving a good spectral discrimination between pure and adulterated essential oils. These results suggest that Raman spectroscopy can overcome limitations of GC‐based methods, thus becoming an interesting alternative and complementary technique for quality control of essential oils.
Solanum phureja es una variedad andina de papa con un alto potencial de exportación, gracias a sus propiedades nutricionales, organolépticas y funcionales; sin embargo, su tiempo de vida en percha es muy reducido. Esta investigación tuvo como objetivo evaluar el tratamiento de este tubérculo con radiación gamma para retardar la brotación. Se empleó un diseño factorial 4×2 para seleccionar la mejor combinación de dosis (0, 80, 120 o 160 Gy) y tiempo después de la cosecha (uno o cinco días) del tratamiento. Asimismo, se utilizó un diseño factorial 3×2 para seleccionar la mejor combinación de dosis de clorprofam (CIPC) (0, 20 o 30 mg kg-1) y tiempo después de la cosecha (uno o cinco días); este es un tratamiento químico común para retrasar la germinación de la papa. El mejor tratamiento físico correspondió a la irradiación con una dosis de 120 Gy, cinco días después de la cosecha; produjo una pérdida de peso de 16.5%, una pérdida de firmeza de 17.2%, ausencia de brotes y 4.0% de pudrición después de 40 días de almacenamiento a 19 ° C y 60% de humedad relativa. El mejor tratamiento químico correspondió a la aplicación de 30 mg de CIPC kg-1 a los cinco días de la cosecha, y produjo una pérdida de peso del 17,4%, una pérdida de firmeza del 21,0 %, un 8,0% de podredumbre y la aparición de brotes después de 25 días. En las papas de la muestra control (sin ningún tratamiento), la pérdida de peso fue del 23,7%, la pérdida de firmeza del 20,9%, la pudrición del 7,0% y la aparición de brotes a los 6 días. Los análisis sensoriales (apariencia, color, textura y flavor) y proximal (contenido de humedad, cenizas, proteína, extracto etéreo, fibra cruda y carbohidratos totales) revelaron que los tratamientos aplicados no afectaron significativamente las características organolépticas y nutricionales de las papas. Estos resultados sugieren que la irradiación de chaucha amarilla permite incrementar su vida útil en aproximadamente 33 días, conservando su calidad.
La mosca de la fruta (Anastrepha fraterculus) afecta a varios cultivos ecuatorianos con potencial de exportación. En la actualidad, para el monitoreo de esta plaga, se emplea un cebo importado que tiene un alto costo. La presente investigación tuvo como objetivo formular cebos atrayentes de mosca de la fruta que puedan reemplazar al cebo comercial. Como materia prima se empleó torta de soya, torta de palmiste, sangre bovina y suero de leche; cada material fue irradiado con una dosis de 20 kGy, en una fuente de cobalto-60, como pretratamiento. Luego, la proteína se extrajo y se hidrolizó con una solución de bromelina de 0,025 UA/mL, a pH 7,0 y 50 °C, durante 30 min. Los cebos fueron formulados con proteína hidrolizada, melaza, agua y bórax y fueron colocados en trampas McPhail. La evaluación en campo se llevó a cabo en cultivos de chirimoya (Annona cherimola) y guayaba (Psidium guajava). En el proceso enzimático se alcanzaron grados de hidrólisis entre el 19,16 y 26,64 %. Por electroforesis SDS-PAGE, se determinó que los hidrolizados proteicos contenían péptidos con pesos moleculares entre 5 y 20 kDa. Se encontró que el cebo de proteína hidrolizada de palmiste y el cebo comercial fueron estadísticamente iguales en la cantidad de moscas atrapadas, mientras que el de lactosuero presentó un mayor índice MTD (moscas capturadas por trampa, por día). Los cebos formulados podrían ser una alternativa más económica que el cebo importado para el monitoreo de la mosca de la fruta en el Ecuador.
Los inhibidores de proteasas vegetales con frecuencia son proteínas de baja masa molar, presentes en tejidos de almacenamiento y aéreos de las plantas. Su expresión se induce en respuesta al daño por insectos y microorganismos patógenos y el contenido de estos inhibidores puede ser alto en órganos de cereales y leguminosas. Bajo la premisa de que la combinación de técnicas de purificación de proteínas de baja y alta resolución permite incrementar la actividad específica de inhibidores de proteasas presentes en tejidos vegetales, el objetivo de este estudio fue aislar, purificar y caracterizar bioquímicamente inhibidores de papaína a partir de semillas de amaranto (Amaranthus caudatus) y frijol (Phaseolus vulgaris). El diseño experimental fue completamente al azar para seleccionar las técnicas de baja resolución que incrementan significativamente (p≤0.05) el grado de purificación de los extractos tratados. En todos los estudios se hicieron tres repeticiones y las mediciones se realizaron por triplicado. A partir de harinas de semillas de amaranto y frijol se obtuvieron extractos proteicos crudos que después se ultrafiltraron en membranas de 10 kDa. Las fracciones de baja masa molar se purificaron por cromatografía de afinidad en papaína-glioxil-sepharosa 6B-CL. El extracto crudo de amaranto presentó actividad inhibidora de 1.177 ± 0.067 mU·mL-1 y el de frijol 1.556 ± 0.542 mU·mL-1. Los extractos con purificación parcial de amaranto y frijol se purificaron por cromatografía de afinidad en papaína-glioxil-sepharosa 6B-CL, hasta 137.5 y 79.1 veces, respectivamente. La masa molar estimada fue 7.50 kDa para el inhibidor de amaranto y 8.20 kDa para el de frijol. El inhibidor de amaranto mostró inhibición competitiva con Ki de 0.872 µM, mientras que la del frijol fue no competitiva y con Ki de 0.058 µM. Esto evidencia la presencia de inhibidores de cisteíno proteasas en las semillas de amaranto y frijol.
