RESUMOA Spirulina platensis tem sido estudada devido a seu alto valor protéico, digestibilidade e por apresentar quantidades significativas de ácidos graxos poliinsaturados, vitaminas, fenólicos e ficocianina, podendo ser utilizada na alimentação humana. A utilização de nutrientes de baixo custo é um fator importante na produção da cianobactéria por possibilitar a redução de custos de processo. Objetivou-se com este trabalho estudar o cultivo mixotrófico da S. platensis por meio da adição de uma fonte orgânica de carbono (glicose) em modo batelada alimentada. Foi utilizado um Planejamento Fatorial Completo 2 3 para o cultivo e as variáveis de estudo foram a concentração de glicose (0,5 gL -1 e 1,0 gL -1 ), a diluição do meio Zarrouk (50% e 75%) e a iluminância (1800 lux e 3000 lux). A concentração celular máxima obtida foi de 5,38 gL -1 com uma velocidade específica máxima de crescimento de 0,0063 h -1 , nas condições de 0,5 gL -1 de glicose, diluição do meio de 75% e iluminância de 3000 lux.Termos para indexação: Biomassa, glicose, iluminância, mixotrófico, Spirulina platensis. ABSTRACTThe cyanobacterium Spirulina platensis has been studied due to its high content (~65%) of highly digestible protein as well as significant amounts of polyunsaturated fatty acids, phenolics, vitamins, minerals and phycocyanin which could be useful in the human nutrition. The use of nutrients of low costs in the cyanobacterium growth could reduce the costs of production. We studied the fed-batch mixotrophic growth of the S. platensis in Zarrouk s medium with glucose (0.5 gL -1 and 1.0 gL -1 ) as carbon source and also investigated the effects of dilution (50% and 75%, with water) and illumination (1,800 lux and 3,000 lux) using a 2 3 factorial design. The maximum celular concentration of 5.38 gL -1 and maximum specific growth rate of 0.0063 h -1 were obtained with a glucose concentration of 0.5 gL -1 , 50% dilution and 1800 lux of illuminance. RESUMOA remoção de metais empregando microrganismos selecionados tem se tornada promissora, já que os mesmos apresentam alta seletividade e taxa de remoção. O presente trabalho teve como objetivo avaliar a remoção de cádmio via bioprocesso submerso por fungos filamentosos. Os microrganismos utilizados foram fungos filamentosos do gênero Aspergillus ssp, identificados por O-3 e O-8, e do gênero Trichoderma sp, identificado como E-19. O meio de cultivo foi composto por 10 % de farelo de trigo, 10 % de solução salina, 4,5 % de extrato de levedura, 2 % de óleo de soja e 10 mg/L ou 20 mg/L de cádmio. Os experimentos foram realizados em erlenmeiers a 30 ºC durante 7 d em agitador orbital. Alíquotas foram coletadas nos tempos inicial e final do bioprocesso para a determinação da remoção do metal. Amostragens nos tempos 0, 1, 2, 3 5 e 7 d foram realizadas para determinação da atividade emulsificante dos meios, a qual indica a produção de biossurfactantes pelos microrganismos. Os fungos O-3 e O-8 apresentaram 100 % de remoção do metal adicionado aos meios de cultivo na fermentação submersa, sendo estas remoções superiores que as obtidas por uma espécie de Thrichoderma (46-49%). Durante os processos de biossorção, foi observada a produção de biossurfactantes pelos microrganismos estudados, demonstrando a importância destes compostos nos processos de remoção de metais de meios líquidos. Palavras-chave: bioprocesso submerso, fungos, remoção de metal ABSTRACTThe removal of metals using selected microorganisms has made promising, since they present high selectivity and rate of removal. The aim of this work was to evaluate the removal of cadmium via submerged bioprocess for filamentous fungi. The microorganisms used were filamentous fungi of the genus Aspergillus spp, identified by O-3 and O-8, and of the genus Trichoderma sp, identified as E-19. The culture medium was composed of 10% wheat bran, 10% of saline solution with micronutrients, 4.5% of yeast extract, 2% of soybean oil and 10 mg/L or 20 mg/L of cadmium. The experiments were performed in erlenmeiers at 30° C for 7 d in a shaker. Aliquots were collected at beginning and end of the bioprocess for the determination of metal removal. Sampling at 0, 1, 2, 3, 5 and 7 d were performed to determine the emulsifying activity of the media, which indicates the production of biosurfactants by microorganisms. The Aspergillus strains showed 100% removal of the metal added to the culture medium in submerged fermentation, superior than Thrichoderma (O-3 and O-8) removals (46-49%). During the process of biosorption, we observed the production of biosurfactants by the microorganisms studied, demonstrating the importance of these compounds in the process of removal of metals from liquid media.
The inappropriate discharge of wastewater containing high concentrations of toxic metals is a serious threat to the environment. Given that the microalga Spirulina platensis has demonstrated a capacity for chromium VI (Cr (VI) biosorption, we assessed the ideal concentration of chromium-containing wastewater required for maximum removal of Cr (VI) and chemical oxygen demand (COD) from the environment by using this microalga. The Paracas and Leb-52 strains of S. platensis, with initial wastewater concentrations of 0%, 12.5%, 25%, and 50%, were cultured in Zarrouk medium diluted to 50% under controlled air, temperature, and lighting conditions. The cultures were maintained for 28 days, and pH, biomass growth, COD, and Cr (VI) were assessed. The wastewater concentration influenced microalgal growth, especially at high concentrations. Removal of 82.19% COD and 60.92% Cr (VI) was obtained, but the COD removal was greater than the Cr (VI) removal in both strains of S. platensis.
RESUMONa confecção deste trabalho se utilizou a metodologia de superfície de resposta para otimizar o efeito do número de Reynolds, tempo de floculação e concentração de hipoclorito de sódio sobre a oxidação/floculação do ferro presente em águas subterrâneas em um sistema de aeração com autoaspiração de ar. O sistema se compunha de um vaso tipo Venturi, acoplado a um tubo de mistura para promover a oxigenação da água através da sucção do ar atmosférico. O mapeamento hidrodinâmico permitiu verificar as condições de operação no qual o sistema apresentou melhor eficiência de sucção de ar e menor consumo de energia, além de compará-las com as melhores condições a campo. Os resultados observados demonstraram que foi possível a remoção de 98,7% do ferro presente (residual ferro de 0,06 mg L -1 ) quando o sistema operou com número de Reynolds no estrangulamento do Venturi de 5,39 x 10 4 , concentrações de hipoclorito de sódio de 38,4 mg L -1 e tempo de floculação 30 min. A metodologia de superfície de resposta foi satisfatória e permitiu otimizar as variáveis operacionais citadas.Palavras-chave: aeração, águas subterrâneas, metodologia de superfície de resposta, remoção de ferro Optimization of auto-aspiration aeration system type Venturi for the treatment of ferruginous water ABSTRACTIn this study the response surface methodology was used to optimize the effect of Reynolds number, flocculation time and sodium hypochlorite concentration on the iron oxidation/flocculation present in groundwaters in an aeration system with air auto-aspiration. This system was composed of a recipient type Venturi coupled to a mixture tube to promote the oxygenation of the water through the suction of the atmospheric air. The hydrodynamic mapping allowed the verification of the operation conditions in which the system presented the best air suction efficiency and energy consumption, and the comparison of the best field conditions. The observed results demonstrated that it was possible to remove 98.7% of present iron (residual iron of 0.06 mg L -1 ) when the system operated with Reynolds number of 5.39 x 10 4 , sodium hypochlorite concentrations of 38.4 mg L -1 and flocculation time of 30 min. The response surface methodology was satisfactory and allowed for the optimization of the mentioned operational variables.
