During dark-induced senescence isovaleryl-CoA dehydrogenase (IVDH) and D-2-hydroxyglutarate dehydrogenase (D-2HGDH) act as alternate electron donors to the ubiquinol pool via the electron-transfer flavoprotein/electron-transfer flavoprotein:ubiquinone oxidoreductase (ETF/ETFQO) pathway. However, the role of this pathway in response to other stresses still remains unclear. Here, we demonstrated that this alternative pathway is associated with tolerance to drought in Arabidopsis. In comparison with wild type (WT) and lines overexpressing D-2GHDH, loss-of-function etfqo-1, d2hgdh-2 and ivdh-1 mutants displayed compromised respiration rates and were more sensitive to drought. Our results demonstrated that an operational ETF/ETFQO pathway is associated with plants' ability to withstand drought and to recover growth once water becomes replete. Drought-induced metabolic reprogramming resulted in an increase in tricarboxylic acid (TCA) cycle intermediates and total amino acid levels, as well as decreases in protein, starch and nitrate contents. The enhanced levels of the branched-chain amino acids in loss-of-function mutants appear to be related to their increased utilization as substrates for the TCA cycle under water stress. Our results thus show that mitochondrial metabolism is highly active during drought stress responses and provide support for a role of alternative respiratory pathways within this response.
Despite the fundamental importance of nicotinamide adenine dinucleotide (NAD+) for metabolism, the physiological roles of NAD+ carriers in plants remain unclear. We previously characterized the Arabidopsis thaliana gene (At1g25380), named AtNDT2, encoding a protein located in the mitochondrial inner membrane, which imports NAD+ from the cytosol using ADP and AMP as counter-exchange substrates for NAD+. Here, we further investigated the physiological roles of NDT2, by isolating a T-DNA insertion line, generating an antisense line and characterizing these genotypes in detail. Reduced NDT2 expression affected reproductive phase by reducing total seed yield. In addition, reduced seed germination and retardation in seedling establishment were observed in the mutant lines. Moreover, remarkable changes in primary metabolism were observed in dry and germinated seeds and an increase in fatty acid levels was verified during seedling establishment. Furthermore, flowers and seedlings of NDT2 mutants displayed upregulation of de novo and salvage pathway genes encoding NAD+ biosynthesis enzymes, demonstrating the transcriptional control mediated by NDT2 activity over these genes. Taken together, our results suggest that NDT2 expression is fundamental for maintaining NAD+ balance amongst organelles that modulate metabolism, physiology and developmental processes of heterotrophic tissues.
The agriculture sector has historically been a major source of greenhouse gas (GHG) emissions into the atmosphere. Although the use of synthetic fertilizers is one of the most common widespread agricultural practices, over-fertilization can lead to negative economic and environmental consequences, such as high production costs, depletion of energy resources, and increased GHG emissions. Here, we provide an analysis to understand the evolution of cereal production and consumption of nitrogen (N) fertilizers in Brazil and to correlate N use efficiency (NUE) with economic and environmental losses as N2O emissions. Our results show that the increased consumption of N fertilizers is associated with a large decrease in NUE in recent years. The CO2 eq. of N2O emissions originating from N fertilization for cereal production were approximately 12 times higher in 2011 than in 1970, indicating that the inefficient use of N fertilizers is directly related to environmental losses. The projected N fertilizer forecasts are 2.09 and 2.37 million ton for 2015 and 2023, respectively. An increase of 0.02% per year in the projected NUE was predicted for the same time period. However, decreases in the projected CO2 eq. emissions for future years were not predicted. In a hypothetical scenario, a 2.39% increase in cereal NUE would lead to $ 21 million savings in N fertilizer costs. Thus, increases in NUE rates would lead not only to agronomic and environmental benefits but also to economic improvement.
Resumo: O presente artigo analisou o papel desempenhado pelos sistemas agroflorestais (SAF's) como medida adaptativa às mudanças climáticas no Brasil. Foi desenvolvido um modelo de efeito de tratamento (Propensity Score Matching), que permitiu identificar os principais determinantes do uso de SAF's e se municípios nos quais essa técnica é utilizada são menos vulneráveis às mudanças climáticas. Os resultados indicaram que variáveis socioeconômicas (propriedade da terra, opções de financiamento, acesso a informações e assistência técnica) e agronômicas (disponibilidade de recursos hídricos e qualidade do solo) influenciam a adoção de SAF's nos municípios brasileiros. As condições climáticas (temperatura e precipitação) também têm participação importante no emprego desses sistemas, o que confirma seu papel de estratégia adaptativa. Concluise também que os SAF's têm potencial de melhorar o desempenho agrícola brasileiro, já que o valor da terra tende a ser maior em municípios onde esses sistemas são utilizados. Desse modo, os SAF's podem tornar o setor agropecuário menos exposto aos efeitos negativos das mudanças climáticas, tanto no presente quanto em cenários futuros.
Responses of leaf gas exchange, fluorescence emission, chlorophyll concentration, and morpho-anatomical features to changes in photosynthetic photon flux density (PPFD) were studied in three wild ornamental species of Passiflora L. to select sun and shade species for landscaping projects. Artificial shade was obtained with different shading nylon nets, under field conditions, which allowed the reduction of 25, 50, and 75% of global radiation, along with a control treatment under full sunlight. For Passiflora morifolia the highest mean values of light-saturated net photosynthetic rate (P Nmax ) and light compensation point (LCP) were observed at 50 and 25% shade, respectively, while the highest values of dark respiration rate (R D ) and apparent quantum yield (α) were observed at 75% shade. For Passiflora suberosa litoralis the highest value of P max was observed at full sunlight. The highest mean values for P max , R D , and LCP for Passiflora palmeri var. sublanceolata were obtained at 25% shade. The highest values of net photosynthetic rate (P N ) for P. morifolia, P. palmeri var. sublanceolata, and P. suberosa litoralis were 21.09, 16.15, and 12.36 μmol(CO 2 ) m -2 s -1 , observed at 50 and 75% shade and full sunlight, respectively. The values of the minimal chlorophyll fluorescence (F 0 ) were significantly different in P. suberosa litoralis and P. palmeri var. sublanceolata, increasing with the increase of the irradiance. In contrast, the values of maximum photochemical efficiency of PSII (F v /F m ) were significantly different only in P. suberosa litoralis, being higher at 75%, progressively reducing with the increase of PPFD levels. The total concentration of chlorophyll (Chl) was higher in shaded plants than in the ones cultivated in full sunlight. On the other hand, the values of Chl a/b ratio were reduced in shaded plants. A significant effect of shade levels on leaf area (LA) and specific leaf area (SLA) was found for the three species, whose highest mean values were observed at 75% shade. The thickness of foliar tissues was significantly higher for the three species at full sunlight and 25% shade. These results suggested that P. morifolia and P. palmeri var. sublanceolata appeared to be adapted to moderate shade conditions. P. suberosa litoralis presented higher plasticity to greater variation of the irradiance levels, while the photoinhibition was one of the limiting factors for this species at full sunlight.
