RESUMOPalavras-chave adicionais: Brachiaria brizantha, Panicum maximum, patógeno.Objetivou-se avaliar a incidência de fungos e nematoides em sementes de Brachiaria sp. e Panicum maximum produzidas nos estados de Mato Grosso do Sul (MS), Mato Grosso (MT), Goiás (GO), Minas Gerais (MG) e São Paulo (SP Com a expansão das pastagens e intensificação da atividade pecuária nos últimos anos, várias doenças de forrageiras começaram a ter importância significativa, especialmente nas regiões centro-oeste e norte do Brasil, causando perdas em produtividade e qualidade de pastagens.Pouco se conhece sobre a qualidade sanitária de sementes forrageiras produzidas e comercializadas no país, o que provavelmente contribui para o aumento da incidência de doenças em áreas de pastagens (6).A associação de patógenos às sementes representa impacto significativo no estande, desenvolvimento das plantas, produção de sementes, persistência das forrageiras no campo e, consequentemente, na produção de leite e carne. The fungi Fusarium oxysporum f. sp. vasinfectum (Fov) and F. oxysporum f. sp. phaseoli (Fop) are important pathogens that cause wilt on cotton and common bean, respectively. In the present study, phylogenetic trees constructed with partial sequences of the translation elongation factor gene and ribosomal intergenic spacer region of Brazilian Fov and Fop strains revealed polyphyletic origin of strains within both formae speciales. This creates an obstacle for the development of specific molecular diagnostic methods and explains the failure of supposedly specific methods described in the literature to correctly discriminate formae speciales of F. oxysporum.
The effect of chemical treatment of wheat seeds (cv. "BR 18 Terena") was studied under laboratory and greenhouse conditions using the following fungicides with the respective doses of i.a./100 kg of seeds: captan (150 g), methilic tioafanate (75 mL), triflumizole (45 g), triticonazole (45 mL) triadimenol (13.5 mL), tolyfluanid (75 g), tebuconazole (5 mL), fludioxonil (5 mL), difeconazole (30 mL) and thiabendazole (30 mL). In laboratory, seeds health and germination were evaluated while under greenhouse conditions were evaluated emergency, velocity of emergency and the height of seedlings. Portanto a utilização de sementes livres do patógeno é uma medida que visa diminuir a fonte de inoculo.De acordo com Machado (7), o melhor método para o controle de doenças é sempre o uso de sementes de boa qualidade. Quando não há disponibilidade dessas sementes em quantidade suficiente, ou se deseja introduzir materiais de procedências duvidosas, ou ainda reduzir o potencial de inóculo primário, é recomendável que as sementes sejam tratadas. No caso de fungos de solo, o tratamento das sementes pode também proteger as plântulas (12).O tratamento químico é o método mais comum de se tratar sementes, sendo de grande valor comercial. O objetivo deste trabalho foi comparar diferentes métodos utilizados para a detecção de Fusarium graminearum em sementes de trigo. Foram empregados 22 tratamentos: papel de filtro (PFC) com congelamento; papel de filtro (PF); PF mais 0,02% de 2,4 - D; meio semi-seletivo (MSS); MSS mais 0,02% de 2,4 - D; MSS mais KCl (-0,8 MPa); KCl, NaCl, manitol e sacarose nos potenciais osmóticos de -0,4, -0,6, -0,8 e -1,0 MPa. O delineamento estatístico empregado foi o inteiramente casualizado, com quatro repetições (duas placas mais substrato com 25 sementes cada/repetição). Não houve diferenças significativas entre os diferentes métodos empregados e o método do PF com congelamento das sementes, considerado o método padrão para o teste de sanidade de sementes de gramíneas, na detecção de F. graminearum em sementes de trigo.
Considerando a grande variabilidade apresentada pelo complexo Diaporthe/Phomopsis, os métodos rotineiros de sanidade de sementes, empregados na detecção do gênero Diaporthe, não são confiáveis visto que, para sua identificação são utilizadas as características morfológicas das colônias que se desenvolvem sobre as sementes. Diante disso, este trabalho teve como objetivo desenvolver iniciadores específicos, bem como um método confiável para detecção e identificação de Diaporthe phaseolorum var. meridionalis (Dphm), em sementes de soja. Amostra de sementes da cultivar IAS5, analisada previamente para sanidade, não portadoras de Diaporthe (SS) foram inoculadas com Dphm (SI), obtendo-se amostras com as seguintes proporções de SI:SS: 1:10, 1:50, 1:100, 1:200 e 1:400. Para a extração de DNA do micélio do patógeno das sementes foi necessário primeiro incubar as sementes por 7 dias, utilizando-se o método do papel de filtro modificado com 2,4 D e, em seguida submetê-las à lavagem em tampão de extração (1% de SDS, 10 mM de TRIS/HCL e 25 mM de EDTA) por 20 min, sob agitação a 100 rpm. Em seguida, alíquotas de 350 miL do sobrenadante foram transferidas para novos microtubos contendo 350 miL de resina de sílica gel da Wizard/Promega permanecendo em contacto com a mesma por 1 minuto. Os DNAs extraídos foram utilizados como molde na reação de PCR com os iniciadores: DphLe (TCG GCC TTG GAA GTA GAA AG) e DphRi (ACT GAA TGC GTT GCG ATT CT) Utilizando-se estes iniciadores, foi possível detectar a presença de D. phaseolorum var meridionalis em sementes de soja, na proporção de uma semente infectada com o patógeno em amostras de 400 sementes (0,25% de incidência), utilizando-se o método do papel de filtro modificado com 2,4 D associado à técnica de PCR.
