The aim of this research was to investigate the antiproliferative and anticholinesterase activities of 11 extracts from 5 Annonaceae species in vitro. Antiproliferative activity was assessed using 10 human cancer cell lines. Thin-layer chromatography and a microplate assay were used to screen the extracts for acetylcholinesterase (AchE) inhibitors using Ellman's reagent. The chemical compositions of the active extracts were investigated using high performance liquid chromatography. Eleven extracts obtained from five Annonaceae plant species were active and were particularly effective against the UA251, NCI-470 lung, HT-29, NCI/ADR, and K-562 cell lines with growth inhibition (GI50) values of 0.04-0.06, 0.02-0.50, 0.01-0.12, 0.10-0.27, and 0.02-0.04 µg/mL, respectively. In addition, the Annona crassiflora and A. coriacea seed extracts were the most active among the tested extracts and the most effective against the tumor cell lines, with GI50 values below 8.90 µg/mL. The A. cacans extract displayed the lowest activity. Based on the microplate assay, the percent AchE inhibition of the extracts ranged from 12 to 52%, and the A. coriacea seed extract resulted in the greatest inhibition (52%). Caffeic acid, sinapic acid, and rutin were present at higher concentrations in the A. crassiflora seed samples. The A. coriacea seeds contained ferulic and sinapic acid. Overall, the results indicated that A. crassiflora and A. coriacea extracts have antiproliferative and anticholinesterase properties, which opens up new possibilities for alternative pharmacotherapy drugs.
ABSTRACT. It is estimated that 60% of anticancer drugs are derived directly or indirectly from medicinal plants. Schinus terebinthifolius Raddi (Anacardiaceae) is traditionally used in Brazilian medicine to treat inflammation, ulcers, and tumors. Because of the need to identify new antimutagenic agents and to determine their mechanism of action, this study evaluated the chemopreventive activity of the methanolic extract from leaves of S. terebinthifolius (MEST) in Allium cepa cells and in Swiss mice analyzing different protocols of MEST in association with DNA-damaging agents. The antigenotoxic and antimutagenic aspects in peripheral blood were evaluated using the comet and micronucleus assays, respectively. The percentage of damage reduction was used to compare the A. cepa and mice results. Our results showed for the first time that MEST can act as a chemopreventive compound that promotes cellular genome integrity by desmutagenic and bioantimutagenic activities in vegetal and animal models. This finding may therefore have therapeutic applications that can indirectly correlate to the prevention and/or treatment of the degenerative diseases such as cancer.
The aim of this study was to evaluate the phenolic and flavonoids contents and the antioxidant and antitumoral activity of leaf and calyx methanolic extracts from Hibiscus sabdariffa (roselle) cultivated with poultry litter and organosuper under three modes of application. The antioxidant potential of fruit pulp and seeds of extracts of the Annona coriacea, and A. sylvatica (Annonaceae) were investigated, as well contents total phenolics, flavonoids, condensed tannins and ascorbic acid. Was used to determine the antioxidant activity the 1,1-diphenyl-1-picrylhydrazyl free radical (DPPH), β-carotene bleaching and ABTS radical cation method. The total phenol, total flavonoid, condensed tannin, and ascorbic acid contents were measured spectrophotometrically. In this study, the pulp and seeds of the fruits were extracted using methanol/water (8:2) for maceration. The seed extracts of A. coriacea demonstrated a moderate antioxidant effect with free radical scavenging activity of 31.53%, by the DPPH test, 51.59% by the β-carotene bleaching test and 159.50 µM trolx/g of extract in the ABTS assay. We found that the hydromethanolic seed extract of A. coriacea had high total phenol (147.08 ± 4.20 mg of GAE/g of extract) and flavonoid (131.18 ± 2.31 mg of QE/g of extract) content. Spodoptera frugiperda (J.E. Smith) (Lepidoptera: Noctuidae) causes significant losses in corn crops and necessitates the use of alternative control strategies, such as the application of bioinsecticides. We report the effect of methanolic leaf extracts of Annona dioica, Annona cacans, and Annona coriacea on the development and reproduction of S. frugiperda. A quantitative analysis was carried out to determine the total concentration of phenolics, flavonoids, and condensed tannin (CT) in leaf extracts. Corn leaves were immersed in a 1% methanolic leaf extract solution and fed to second instars of S. frugiperda. Leaf disks dipped in the synthetic insecticide Connect® (Bayer CropScience Ltda) composed of a neonicotinoid (imidacloprid) and a pyrethroid (β-cyfluthrin), which are harmful to S. frugiperda, was used as positive control. Distilled water was used as a negative control treatment. The leaf extract of A. coriacea decreased larval survivorship, arrested pupal development, and affected the weight gain of S. frugiperda. A. dioica also affected larval survivorship, but its effects were more pronounced for the adult stage, as fecundity, fertility, egg hatchability, and embryonic development were severely affected. Leaf extracts from A. cacans had no effect on S. frugiperda. The leaf extracts of A. dioica and A. coriacea showed a higher content of flavonoids and phenols, respectively. Our results indicated that both A. dioica and A. coriacea have the potential for development as botanical insecticides.
RESUMO:O trabalho teve como objetivos avaliar a morfometria dos frutos e sementes de carobinha (Jacaranda decurrens subsp. symmetrifoliolata) e o efeito de temperaturas na germinação de sementes armazenadas em ambiente sem controle de temperatura e umidade relativa. O estudo foi realizado no Laboratório de Sementes da UFGD, Dourados, MS. Em amostra de 120 frutos e de 550 sementes foi determinado: o comprimento, a largura, a espessura, o número de sementes por fruto, e a massa dos frutos. A germinação foi estudada após três períodos de armazenamento das sementes (120, 480 e 720 dias) a temperatura ambiente e sob quatro temperaturas de germinação: três constantes (18, 25 e 30ºC) com luz contínua e sob condição de tempera alternada, 20-30ºC, com fotoperíodo de 12 horas. O delineamento experimental foi inteiramente ao acaso, em arranjo fatorial 3 x 4, com quatro repetições. As variações nas medidas de frutos e sementes representam indício da alta variabilidade genética populacional. As sementes armazenadas por 120 dias e submetidas à germinação na temperatura de 25ºC resultaram em 69,4% de germinação e índice de velocidade de germinação -IVG, de 0,983. Sob temperaturas de germinação de 30ºC, constante, e de 20-30ºC, alternada, obtiveram-se 63,9% e 63,8% de germinação e 0,486 e 0,413 de IVG, respectivamente. Sob temperatura de 18ºC a germinação foi lenta e menor, 27,7% e IVG de 0,119. As sementes armazenadas por 480 e 720 dias não germinaram. Com base no comportamento fisiológico das sementes é possível classificar Jacaranda decurrens subsp. symmetrifoliolata como espécie intermediária (entre ortodoxa e recaucitrante).Palavras-chave: planta medicinal, Cerrado, biometria de sementes e frutos, germinação ABSTRACT: Fruit and seed morphometry and germination of "carobinha" (Jacaranda decurrens subsp. symmetrifoliolata) seeds after storage. This study aimed to evaluate the morphometry of fruits and seeds of Jacaranda decurrens subsp. symmetrifoliolata ("carobinha") and the effect of temperature on the germination of seeds that were stored in environment without controlled temperature and relative humidity. The study was carried out at Seed Laboratory of Federal University of Grande Dourados, Mato Grosso do Sul, Brazil. For a sample of 120 fruits and 550 seeds, the following variables were determined: length, width and thickness, besides the number of seeds per fruit and fruit biomass. Germination was studied after three seed storage periods (120, 480 and 720 days) at environment temperature and four germination temperatures: three constant temperatures (18, 25 and 30ºC) with continuous light, and one alternating temperature, 20-30ºC, with 12-hour photoperiod. RESUMO:O objetivo deste trabalho foi avaliar o desenvolvimento inicial da guavira (Campomanesia adamantium) cultivada sob diferentes doses de fósforo e nitrogênio. O experimento foi realizado em casa de vegetação, da Universidade Federal da Grande Dourados, em Dourados-MS. Foram estudadas cinco doses de fósforo (20, 120, 200, 280 e 380 kg ha -1 ), na forma de superfosfato simples e cinco doses de nitrogênio (6, 36, 60, 84 e 114 kg ha -1 ), na forma de sulfato de amônio. Os tratamentos foram definidos utilizando-se a matriz experimental Plan Puebla III, dando origem a nove combinações, respectivamente, de doses de fósforo e de nitrogênio (kg ha -1 ): 280 e 84; 280 e 36; 120 e 6; 120 e 36; 120 e 84; 200 e 60; 380 e 84, 20 e 36 e 280 e 114. O delineamento experimental utilizado foi blocos casualizados, com quatro repetições. A unidade experimental foi composta por cinco vasos, com uma planta por vaso. A colheita das plantas foi feita aos 270 dias após o transplante (DAT). Sob as maiores doses de P e N utilizadas observaram-se as maiores alturas de plantas (38,12 cm), aos 261 DAT; o maior número de folhas por planta (54), aos 186 DAT; a maior massa seca de folhas (5,68 g planta -1 ), a maior área foliar (610 cm 2 planta -1 ), a maior massa seca de raiz (6,2 g planta -1 ) e o maior número de ramos (3 planta -1 ) aos 270 DAT. O teor de clorofila foi em média de 36 ICF. Recomenda-se o uso de 380 kg ha -1 de fósforo e de 114 kg ha -1 de nitrogênio para o melhor desenvolvimento inicial da guavira.
