A rapid and convenient biochemometrics-based analysis of several cereal-derived extracts was used to identify n-alkyl(enyl)resorcinols (AR) as antifungals against Fusarium oxysporum. Total AR content and liquid chromatography/mass spectrometry (LC-MS)-based profiles were recorded for each extract, in addition to their antifungal activity, to help integrate these chemical and biological datasets by orthogonal partial least squares regression. In this study, we developed and used a micro-scale amended medium (MSAM) assay to evaluate the in vitro mycelial growth inhibition at low amounts of extracts. Triticale husk-derived extracts had the highest AR content (662.1 µg olivetol equivalent/g dry extract), exhibiting >79% inhibition at the highest doses (10.0–1.0 µg/µL). Correlation of the chemical and antifungal datasets using supervised metabolite profiling revealed that 5-n-nonadecanylresorcinol, 5-n-heneicosylresorcinol, and 5-n-tricosyl-resorcinol were the most active ARs occurring in cereal products from Colombia. Hence, we propose the biochemometrics-based approach as a useful tool for identifying AR-like antifungals against F. oxysporum.
Baccharis es un género de plantas muy numerosa (400-500 especies) altamente distribuidas en el neotrópico, dentro de las que se encuentra B. latifolia (Ruiz & Pav.) Pers. Esta planta presenta diversos usos en medicina tradicional en América Latina, dentro de los cuales se destaca usos analgésicos y antiinflamatorios, los cuales ha sido objeto de estudios adicionales que se han encauzado en su validación. Sin embargo, no hay una conexión importante entre las propiedades medicinales o farmacológicas con la composición química, salvo aquellos casos donde se justifica la actividad con otros estudios donde se aíslan compuestos que poseen actividad. Por tanto, el escenario indica que, pese a que existe información importante, los datos son marginales o separados entre sí, lo que permite justificar que es todavía importante estudios adicionales con fines quimioprospectivos. Por consiguiente, en el presente trabajo se describe, de forma generalizada, la información reportada a la fecha en estudios químicos y biológicos de la familia Asteraceae, del género Baccharis y de la especie B. latifolia, la cual, pese a tener un potencial claro en el neotrópico, es una planta poco conocida y valorada que merece mucha atención.
Para la evaluación de la actividad citotóxica de las especies Conyza trihecatactis y Ageratina vacciniaefolia pertenecientes a la familia Asteareaceae, se recolectaron las partes aéreas de las especies y se obtuvieron extractos completos etanólicos y fracciones con solventes en orden creciente de polaridad. La evaluación de la actividad citotóxica se realizó por el método del MTT sobre líneas celulares tumorales de cáncer de seno murino y humano, y adicionalmente, sobre una línea no tumorigénica de fibroblastos de origen murino. La fracción de diclorometano de C. trihecatactis (CD) presentó la mayor actividad citotóxica con una CI50 de 36,23 µg/mL para 4T1, 47,81 µg/mL para TSA, 46,05 µg/mL para MCF-7 y 70,67 µg/mL en fibroblastos 3T3. A partir de esta fracción se obtuvo una mezcla de flavonoides (CMF), identificados como apigenina e hispidulina, que presentó un marcado efecto citotóxico sobre MCF-7 con una CI50 de 23,50 mg/mL. Las fracciones obtenidas a partir de A. vacciniaefolia presentaron CI50 mayores a 150 µg/mL en las líneas tumorales evaluadas y superior a 180 µg/mL en la línea de fibroblastos 3T3. En la fracción de cloroformo de A. vacciniaefolia (AC) se identificaron cuatro compuestos de tipo terpenoide, que presentaron similitud en el tiempo de retención (tret) y en el espectro de masas con los compuestos aislados e identificados en estudios previos para esta especie.
