Targeted Metabolite Profiling-Based Identification of Antifungal 5-n-Alkylresorcinols Occurring in Different Cereals against Fusarium oxysporum

Abstract: A rapid and convenient biochemometrics-based analysis of several cereal-derived extracts was used to identify n-alkyl(enyl)resorcinols (AR) as antifungals against Fusarium oxysporum. Total AR content and liquid chromatography/mass spectrometry (LC-MS)-based profiles were recorded for each extract, in addition to their antifungal activity, to help integrate these chemical and biological datasets by orthogonal partial least squares regression. In this study, we developed and used a micro-scale amended medium (MS… Show more

“…In vitro antifungal activity, directly tested on F. oxysporum mycelial growth sensitivity, was assessed through a 12-well plate amended-medium method 39 using a F. oxysporum isolate from Cape gooseberry. Five concentrations (between 10 and 300 μg/mL) were separately used to evaluate the antifungal activity of all synthesized 5 and 6 series compounds.…”

“…Antifungal activity evaluation of QREs was performed by measuring the growth halo of the phytopathogen F. oxysporum with the presence of the extracts at different concentrations compared to that of a blank (0.5% PDA), using the amended-medium procedure [ 69 ]. The culture medium contained 2.4% PDB and 1.5% bacteriological agar in 100 mL of distilled water.…”

“…A microscale amended medium protocol was performed following the previously reported procedure. 44 Briefly, 12-well plates (autoclavable glass), semisolid medium (1.2 g of potato-dextrose broth, and 0.5 g of agar per 100 mL of distilled water) were used. The final volume (for treatments and controls) was 200 μL per well (1 cm diameter).…”
