Tick-borne rickettsiosis is an important emerging disease in Panama; to date, there have been 12 confirmed cases, including eight fatalities. To evaluate the distribution of rickettsiae in Panamanian ticks, we collected questing and on-host ticks in urban and rural towns in elevations varying between 0 and 2300 m. A total of 63 sites (13 urban and 50 rural towns) were used to develop models of spatial distributions. We found the following tick species: Rhipicephalus sanguineus s.l. (present in 54 of 63 towns and cities), Amblyomma mixtum (45/63), Dermacentor nitens (40/63), A. ovale (37/63), Rhipicephalus microplus (33/63), A. oblongoguttatum (33/63), Ixodes affinis (3/63), and Ixodes boliviensis (2/63). Rhipicephalus sanguineus s.l. was present in urban and rural towns, and other species were present only in rural towns. DNA was extracted from 408 R. sanguineus s.l., 387 A. mixtum, 103 A. ovale, and 11 A. oblongoguttatum and later tested for rickettsiae genes using PCR. Rickettsia DNA was detected in ticks from 21 of 63 localities. Rickettsia rickettsii was detected in five A. mixtum (1.29%), and Candidatus "Rickettsia amblyommii" was found in 138 A. mixtum (35%), 14 R. sanguineus (3.4%), and one A. ovale (0.9%). These results suggest that much of rural Panama is suitable for the expansion of tick populations and could favor the appearance of new tick-borne rickettsiosis outbreaks.
Introducción. Aunque el perro se considera el principal reservorio doméstico de Leishmania infantum en varios países de Latinoamérica, se desconocen las tasas de infección por parásitos del género Leishmania en perros de algunos focos de leishmaniasis visceral del norte de Colombia. Objetivo. Establecer las tasas de infección por Leishmania spp. en poblaciones caninas del departamento de Sucre en el norte de Colombia. Materiales y métodos. Se estudiaron perros de zonas rurales de los municipios de Sincelejo, Sampués y Ovejas. Los parásitos del género Leishmania se detectaron mediante la amplificación de un segmento de ADN de la subunidad ribosómica pequeña (rSSU). Se analizó la relación entre variables como sexo, grupos etarios y presencia de signos clínicos, respecto a la infección por Leishmania. Resultados. Se amplificó un fragmento de 561 pb de la rSSU de Leishmania spp. en 34,9 % (29/83), 35,7 % (15/42) y 11,1 % (1/9) de los perros analizados de Sincelejo, Sampués y Ovejas, respectivamente. La prevalencia total de infección por Leishmania spp. fue de 33,6 %. En dos perros de Sincelejo, dos de Sampués y uno de Ovejas, se obtuvo una banda de 650 pb, característica de parásitos del género Trypanosoma. El 64,2 % de los perros que mostraron al menos un signo clínico indicativo de leishmaniasis canina, fue positivo por PCR. Se halló una relación estadísticamente significativa entre los perros mayores de 33 meses de edad y el resultado positivo para Leishmania spp. (p=0,043). No se encontró relación entre el sexo del perro y el resultado positivo para Leishmania spp. (p=0,85). Conclusión. Un tercio de la población canina analizada se encontró infectada con parásitos del género Leishmania, lo cual podría sustentar su posible papel como fuente de infección para el insecto vector.
Abstract. Chikungunya fever, an acute and often chronic arthralgic disease caused by the mosquito-borne alphavirus, chikungunya virus (CHIKV), spread into the Americas in late 2013. Since then it has caused epidemics in nearly all New World countries, the second largest being Colombia with over 450,000 suspected cases beginning in September, 2014, and focused in Bolivar Department in the north. We examined 32 human sera from suspected cases, including diverse age groups and both genders, and sequenced the CHIKV envelope glycoprotein genes, known determinants of vector host range. As expected for Asian lineage CHIKV strains, these isolates lacked known Aedes albopictus-adaptive mutations. All the Colombian strains were closely related to those from the Virgin Islands, Saint Lucia, Mexico, Puerto Rico, and Brazil, consistent with a single, point-source introduction from the southeast Asia/Pacific region. Two substitutions in the E2 and E1 envelope glycoprotein genes were found in the Colombian strains, especially E1-K211E involving a residue shown previously to affect epistatically the penetrance of the E1-A226V A. albopictus-adaptive substitution. We also identified two amino acid substitutions unique to all American CHIKV sequences: E2-V368A and 6K-L20M. Only one codon, 6K-47, had a high nonsynonymous substitution rate suggesting positive selection.
Rocky Mountain spotted fever (RMSF) has been reported in Colombia since 1937. Most recent outbreaks were in 2006-2008, followed by the absence of recognized cases. This report describes new clinical cases of RMSF and epidemiologic investigations. Medical records were reviewed, and fieldwork, serological and molecular diagnostic testing, and bacterial isolation were performed. Fever, hypotension, abdominal discomfort, and rash accompanied by thrombocytopenia and leukopenia were the most characteristic manifestations. Two convalescent sera from the index case and sera from two dogs revealed high anti-spotted fever group Rickettsia antibody titers. Rickettsia rickettsii was isolated from case 4. Cases 2 and 3 were identified by epidemiological connection with the index case. Thus, a new cluster of cases of RMSF was identified in Antioquia, Colombia, with the occurrence of fatal cases, which indicates the active circulation of the bacteria and a potential risk for the population.

El gen mitocondrial de la subunidad 16S del ARN ribosómico (16S) es uno de los más usados, mientras que el gen mitocondrial de la citocromo oxidasa 1 (COX1) se ha empleado recientemente y se propone como un marcador genético alternativo frente al 16S. Objetivo. Evaluar la utilidad de los genes 16S y COX1 en los estudios genéticos de las garrapatas mediante el análisis de secuencias en tres especies de la región Caribe de Colombia.Resultados. El análisis de secuencias mostró que los dos genes permitieron identificar las tres especies con mucha confiabilidad y con niveles de divergencia genética interespecífica relativamente similares (19 a 22 %), aunque solo el gen COX1 permitió detectar la variabilidad genética intraespecífica (hasta de ~0,8 %). El análisis de saturación de sustituciones indicó que el gen 16S no se saturó con transiciones, mientras que el COX1 mostró saturación a partir de distancias de ~17 %. Conclusión. Los resultados indicaron que el gen 16S parece tener mejores características para los análisis filogenéticos interespecíficos dada su alta divergencia genética y baja saturación de transiciones, mientras que el gen COX1 parece ser más útil para estudios de variabilidad genética intraespecífica. Sin embargo, dado que el estudio se hizo a escala local, se requieren más investigaciones en diferentes escalas biogeográficas para establecer su utilidad en circunstancias más amplias y complejas. Introduction: In recent decades the analysis of mitochondrial genes has been used for population and phylogenetic studies of ticks allowing many advances in their systematics. Mitochondrial ribosomal 16S (16S) subunit is one of the most frequently used among those genes available for tick analysis, whereas cytochrome oxidase gene 1 (COX1) has recently been used and proposed as an alternative to the traditional 16S gene marker. Objective: To evaluate the usefulness of 16S and COX1 in genetic studies of ticks by analyzing sequences of three species commonly found in the Caribbean region of Colombia. Results: The analysis of both genes sequences allowed us to identify the three species with high levels of confidence and interspecific genetic divergence (19-22%), although only COX1 allowed us to detect intraspecific genetic variability (up to ~0.8%).
