Vaccinia virus (VACV), the etiological agent of an exanthematic disease, has been associated with several bovine outbreaks in Brazil since the end of the global vaccination campaign against smallpox. It was previously believed that the vaccine virus used for the WHO global campaign had adapted to an unknown wild reservoir and was sporadically re-emerging in outbreaks in cattle and milkers. At present, it is known that Brazilian VACV is phylogenetically different from the vaccinia virus vaccinal strain, but its origin remains unknown. This study assessed the seroprevalence of orthopoxviruses in domestic and wild animals and farmers from 47 farms in three cities in the southwest region of the state of São Paulo with or without official reports of outbreaks in cattle or humans. Our data indicate a low seroprevalence of antibodies in wild animals and raise interesting questions about the real potential of wild rodents and marsupials as VACV reservoirs, suggesting other routes through which VACV can be spread.

A new outbreak of Vaccinia virus was observed in Southwest region of São Paulo State, Brazil. The disease was observed in small dairy farms with manual milking. Lesions in cattle were observed on teats and udder (Figure 1) characterized by vesicules and ulcers. Milkers presented lesions on hands and one child, 11 years of age, presented lesions in mouth and nose. Age of the milkers ranged from 22 to 63 years of age. Three of them were previously vaccinated against smallpox. Lymphadenopathy, headache, and fever were reported, varying in severity and persisting for ∼2-5 days. Lesions in the ocular region were observed in one milker, 22 years of age, correlated with several lesions on the hands ( Figure 2

Foram examinados 176 eqüídeos (15 muares e 161 eqüinos) do município de Monte Negro, Rondônia, Amazônia Ocidental Brasileira, frente a agentes virais e bacterianos. A amostra correspondeu ao total de eqüídeos no município, considerando um nível de confiança de 99%, prevalência esperada de 50% e erro padrão de 10%. As infecções virais foram investigadas pelas provas de Imunodifusão em gel de Agar (Anemia Infecciosa Eqüina - AIE), Inibição da hemaglutinação (Influenza eqüina tipos 1 e 2 IE-1 e 2) e Soroneutralização em cultura celular (Arterite Viral Eqüina - AVE, Herpesvírus Eqüino tipo 1 HVE1, Estomatite Vesicular - EV e Encefalomielite Eqüina do Leste - EEE, do Oeste - WEE e Venezuela - VEE). Para o diagnóstico da leptospirose, foi utilizada a prova de Soroaglutinação Microscópica (SAM); para o diagnóstico da brucelose, o teste do Antígeno Acidificado Tamponado (AAT) foi utilizado como teste de triagem e as provas de Soroaglutinação Lenta em Tubos (SLT) e 2-mercaptoetanol como testes diagnósticos. Foram constatados 9,6% dos eqüídeos reativos para AIE, 22,7% para HVE1, 19,9% para IE-1, 42,0% para IE-2, 21,0% para EEE, 11,3% para VEE, 3,4% para Brucella spp. e 91,4% para Leptospira spp. Os sorovares de leptospira mais freqüentes foram Bratislava (10,5%), Icterohaemorrhagiae (8,7%) e Autumnalis (8,7%) nos eqüinos e Patoc (26,6%) nos muares. Não foram encontrados animais com anticorpos contra AVE, EV e WEE.

Neospora caninum, a cause of abortion and stillbirth in cattle, was studied by histology, immunohistochemistry, and nested-PCR, using primers from the Nc5 region of the genomic DNA (PCR PLUS) and primers from the ITS1 region of the ribosomal DNA (PCR JB). A total of 105 fetal samples sent to the Centro de Pesquisa e Desenvolvimento de Sanidade Animal do Instituto Biológico from January 2006 to May 2008 were examined for evidence of N. caninum. Histological examination revealed 71.4% with non-suppurative inflammation in the heart, lung, liver, kidney, placenta, and brain. Immunohistochemistry detected infections in 8.6% of the samples, mainly in the brain, placenta, and heart. Nested-PCR JB revealed 6.7% with infections, while nested-PCR PLUS returned 20.9% positive results, mainly in brain and placenta, and in the pooled liver and heart. Kappa statistics demonstrated little agreement among the three techniques. The three methods are complementary, since they have distinct diagnostic characteristics and were combined to give a positivity rate of 24.8%.

Infecção por Toxoplasma gondii foi associada com perdas reprodutivas em um rebanho caprino no Rio Grande do Sul, Brasil. Lesões macroscópicas foram observadas em dois de seis produtos caprinos enviados para diagnóstico e incluíram linfonodos mesentéricos pálidos e aumentados e pulmões com consistência firme e áreas claras intercaladas com vermelhas. Lesões histológicas, especialmente caracterizadas por infiltrados linfoplasmocitários no cérebro e pulmões, foram observadas em todos os fetos. Nefrite intersticial linfoplasmocitária, linfadenite necrosante e hepatite periportal linfoplasmocitária também foram observadas. Enquanto tanto o exame bacteriológico quanto o teste de imunofluorescência direta para Leptospira sp. foram negativos em todos os casos, a PCR e a imunoistoquímica resultaram positivamente para T. gondii em quase todas as amostras testadas. Anticorpos anti-T.gondii, em titulações de 1:512 a 1:2048, foram detectados nas amostras de soro sangüíneo das cabras que pariram natimortos (3), abortaram (1) ou cujos neonatos morreram (2). Este trabalho descreve os achados clínicos, patológicos, sorológicos, imunoistoquímicos e de PCR observados em um rebanho caprino infectado por T. gondii.

Neosporosis is a parasitic disease that occurs in sheep and is associated with reproductive failure. The aim of this study was to verify seroprevalence of Neospora caninum antibodies in 12 sheep flocks in Uberlândia county, Minas Gerais, Brazil, using Indirect Immunofluorescence Assay (IFA). A total of 334 sheep blood samples were analyzed and an epidemiologic questionnaire was applied for each farm in order to correlate with risk factors of neosporosis: gender, age, breed, abortion problems, contact with cattle and dogs, and presence of wild canids. The prevalence of Neospora caninum antibodies in sheep was 8.1%, with titers ranging from 50 to 3200 and 83.3% of farms detected at least one ovine reactive. There was statistically significant difference in flocks with abortion problems associated with the presence of Neospora caninum antibodies (p < 0.05).

Oocyst stage of Toxoplasma gondii is characterized by a durable wall that confers a strong protection to this protozoan parasite in face of harsh environmental conditions. Thus, it is considered the key for transmission of T. gondii. Analysis of oocyst wall composition is mandatory therefore; the aim of this study was to identify novel T. gondii oocyst wall proteins and test their use in detection of these oocysts in environmental samples. Five candidates of novel T. gondii oocyst wall proteins (TgOWPs) were identified and named TgOWP8 through TgOWP12. Recombinant protein of TgOWP8 was expressed in E. coli using glutathione S-transferase as fusion protein. Polyclonal antibody was produced and validated by indirect immunofluorescence antibody assay (IFA). For detection by IFA, we used different methods for fixation and permeabilization of oocysts to improve the antigen-antibody detection. Specificity to wall of T. gondii oocyst was confirmed and revealed absence of cross reactivity with bradyzoite cyst wall and tachyzoites. Although some TgOWPs were identified previously, our study represents a continuation of molecular investigations of oocyst wall proteins as an essential structure for the longevity and infectivity of this stage and also provided new trial to improve T. gondii oocysts detection.
